Abstract

The Self-Nanoemulsifying Drug Delivery System (SNEDDS) is a drug delivery system developed to enhance the solubility and bioavailability of lipophilic compounds. Optimization of SNEDDS formulation is an essential step to obtain a stable and efficient system, one of which can be achieved using the Simplex Lattice Design (SLD) method that mathematically determines the optimal composition of oil, surfactant, and co-surfactant. This study employed a narrative review approach by collecting literature from Google Scholar, ScienceDirect, and NCBI databases using the keywords “SNEDDS AND simplex lattice design AND optimization AND physicochemical” within the publication range of 2016-2025. Based on the literature screening, five relevan articles met the inclusion and exclusion criteria. The review results showed that the application of the SLD method consistently produced formulations with droplet sizes of 16-200 nm, PDI <0.3, zeta potential values ranging from -31 to -61 mV, emulsification time between 8-47 seconds, and %transmittance of 95-98%, indicating a stable and homogeneous system. SLD has proven effective in optimizing SNEDDS formulations as it can reduce research time and cost while improving efficiency and system stability. Therefore, this method has great potential to be applied in the development of lipid-based drug delivery systems to enhance the physicochemical characteristics and bioavailability of liphophilic drugs.
