Abstract

The extraction method plays a crucial role in obtaining high-quality DNA samples, which is indispensable for various molecular biology techniques and analyses, enabling a deeper comprehension of genetic information and biological processes. The objectives of the study were: a) to optimize the chloroplast DNA extraction protocol by comparing modified CTAB methods and GeneAid for both leaf and wood samples of Shorea leprosula, a major commercial timber species, and b) to identify a suitable cpDNA region that exhibits variability and universality across taxa. Total DNA was analyzed by gel electrophoresis followed by Sanger sequencing to determine the amplification success. The results revealed that trnL intron, trnL-trnF, and trnG yielded readable sequences of the expected length (maximum 586 bp, 480 bp, and 908 bp, respectively), while the rps 16 intron failed to assemble a contig. The petL-psbE region provided long readability for reverse sequences (769 bp) but not for the forward sequence (195 bp). Higher successful DNA extraction was achieved from the leaves compared to the woods. The lower sequencing quality may be attributed to suboptimal primer design, the structural features of the regions resulting from extensive repetitive sequences, and the suboptimal condition of the extraction method in eliminating wood chemical compounds.

Abstract

Rosewood (Dalbergia latifolia) is a hardwood plant, resistant to termite attack, its wood has beautiful patterned fibers and textures, so it is widely used for furniture in luxury categories with high economic value. According to CITES (Convention on International Trade in Endangered Species of Wild Fauna and Flora), rosewood trade status is included in appendix II. In the trade of rosewood in both domestic and international markets, there are relatively many obstacles and difficulties in identifying the origin of rosewood. It is very important to establish a reference on the identity of wood based on the location of origin as an effort to prevent illegal trade, namely by using a molecular-based identification approach (DNA fingerprinting). In the isolation of rosewood DNA, special techniques are required due to its hard wood characteristics. This study aims to compare DNA isolation techniques from wood and leaves of rosewood using the CTAB (Cetyl Trimethyl Ammonium Bromide) protocol and the commercial kit protocol. The research used molecular biology-based identification methods. PCR results showed that the DNA isolation protocol using the commercial kit outperformed the CTAB protocol for both leaf and wood samples. DNA isolation from wood samples was more difficult than leaf samples. Overall, the best protocol for DNA isolation from rosewood leaves and wood was to use a commercial kit.

Abstract

The research method used is a qualitative method with observation and interview techniques with selected informants with purposive sample. The results of the study show that bathing in the Balimau Kasai procession is a tradition that is still highly maintained by the local community to this day as a form of gratitude to Allah SWT and a sense of joy because the holy month of Ramadhan has come and as a symbol of self-purification both physically and mentally ahead of the fasting month of Ramadhan. The traditional procedure for bathing in the Balimau Kasai procession includes parades, distributing cakes and water, bathing in the Balimau Kasai parade, and eating together. Bathing is carried out by several community leaders, religious leaders, and government agencies using water that has been mixed with lime. Bathing in Balimau Kasai hamlet of Simpang Baru Teluk Latak village has social, religious and cultural values for the local community

Abstract

Islam views money as a medium of exchange, not as a commodity. Widespread awareness of the role of money aims to eliminate injustice and injustice in the exchange economy. they are classified as fadl riba. Exchange is an unrealistic method of exchange and proves the unimportance of the overall market mechanism. Literature study is data deepening based on theoretical studies and scientific work references. descriptive qualitative research. Data collection techniques in literature study, namely the collection of literature related to research, then analysis and conclusions about public goods, namely the general public can use it without disturbing the use by other people or certain parties. The money saved is not used for the manufacturing sector (idle assets). This amount is reduced because of the obligation of zakat on Muslims. So the money must be rotated (the concept of cash flow). Should not be productive and used as a transaction tool. Like the spirit of Fastabigul Hoyrot, Islam encourages competition in the economic field. alms, waqf, and other activities. The money must also be used for investments and contracts such as musyarokah, mudharabah, wadiah, qardh, and others, in the real sector, as exemplified by the Prophet Muhammad.

Abstract

This classroom action research (CAR) aims to improve children's creativity through sand painting activities. The background of this study is the low level of creativity observed among children in group B3 at Az-Zahrah Islamic Kindergarten, Palembang. Many children still face difficulties in developing ideas, imagination, and creativity. They tend to imitate given examples, showing limited flexibility, originality, and innovation, and often appear hesitant to try new approaches. Furthermore, some children lack interest in coloring activities. Based on pre-cycle observations, out of 15 children, 13 (87%) were categorized as “Starting to Develop” (MB), while only 2 children (13%) reached the “Developing as Expected” (BSH) category. The research was conducted over two cycles, with each cycle consisting of six meetings. The participants were selected using purposive sampling, focusing on group B3 students. Data were collected through observations, checklists, and field notes. The results of the study show that in Cycle I, 11 children (73%) achieved the BSH category. In Cycle II, the number increased to 12 children (80%) in the BSH category, and 4 children (20%) reached the “Very Well Developed” (BSB) category. These findings indicate that sand painting activities are effective in enhancing children's creativity. Therefore, it is recommended that teachers incorporate sand painting into their learning activities to foster creativity among young learners.
