Abstract

The escalating demand of shallot has forced the increase of domestic production, including through extension of harvest area by utilization of arable land on peatland. This study was aimed to analyze genetic profile of shallot adapted in peatland areas using SSR markers. Twenty-one shallot genotypes were tested in the field and eighteen primers dispersed throughout the genome was applied to analyze genetic diversity of the peatland-adapted shallots. Phenotypic evaluation revealed that shallot yield potential ranged from 6.66 to 14.21 t/ha. Of these, seven shallot clones (11 NA, 1111 TA, 12 NA, 12 NC, 20 NA and 22 N) had good yield potential and comparable with those of five released shallot varieties. Nine out of 12 shallot clones were moderately resistant to Alternaria porii. Clustering analysis showed that shallot genotypes were clustered into two main groups, Clustered I and II which consisted of 13 and 8 genotypes, respectively. The closest genetic relatedness was observed between 8NC and 8NA (0.85), while the farthest ones was between 11NA and Kramat2 (0.51). This result implies that cross combination between 11NA and Kramat2 is valuable and suitable for breeding programs aimed at improving shallot potential yield in the future.

Abstract

One of the internal factors that play an important role in seed quality is the level of the phytohormone auxin. Efforts to increase auxin in seeds include developing new varieties with increased indigenous auxin, such as those found in mutant tomatoes from Micro-Tom, namely iaa9-3 and iaa9-5. This research was conducted to determine the germination response of iaa9-3 and iaa9-5 mutant tomato seeds at different levels of the fruit maturation stage. The research was carried out at the Seed Technology Laboratory, Faculty of Agriculture, Padjadjaran University, and the Seed Testing Laboratory of the Center for Standard Testing of Vegetable Plant Instruments. The response design used was a completely randomized factorial design consisting of two factors and repeated three times. The first factor was the iaa9-3, iaa9-5, and Wild-Type Micro-Tom (WT-MT) mutants as controls. The second factor is the fruit maturation stage, which consists of Breaker, Pink, and Red. The research results showed that there was an interaction between genotype and fruit maturation stage on the parameters of germination, growth simultaneity, and hypocotyl length.  The harvest stage for tomatoes to produce normal strong seed germination was the pink stage in all tomato genotypes tested. The best germination rate was shown by WTMT seeds at the pink stage, genotype iaa9-3 at the red stage, and genotype iaa9-5 at the red or pink stage. The effect of fruit maturity stage on the synchronization of sprout growth was relatively not significantly different in mutant tomatoes, but it had an effect in WTMT tomatoes, namely the best pink stage.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh jenis kemasan terhadap mutu fisiologis  benih bawang daun varietas Fragrant yang telah disimpan selama beberapa periode simpan. Penelitian dilaksanakan pada tahun 2011-2012 dan disusun dalam Rancangan Acak Lengkap pola faktorial yang terdiri atas dua faktor yaitu periode simpan (4 (empat), 8 (delapan) dan 12 (bulan) dan jenis kemasan (kantong alumunium foil kedap, kantong alumunium foil, kantong kertas dan plastik klip)  dengan empat kali ulangan. Parameter yang diamati adalah mutu fisiologis benih yang terdiri dari daya berkecambah benih, kecepatan berkecambah benih, panjang hipokotil, dan laju pertumbuhan kecambah. Hasil penelitian menunjukkan bahwa mutu fisiologis benih bawang daun dipengaruhi oleh periode simpan dan jenis kemasan. Pada suhu ruang, mutu fisiologis benih bawang daun menurun seiring bertambahnya masa simpan dengan menggunakan semua jenis kemasan. Waktu penyimpanan dan jenis kemasan meningkatkan daya berkecambah dan kecepatan laju perkecambahan bawang daun. Untuk mempertahankan daya berkecambah benih bawang merah selama 4 bulan dapat menggunakan seluruh kemasan, namun untuk penyimpanan lebih dari 4 bulan harus menggunakan kemasan alumunium foil. Kata kunci: benih bawang daun, mutu fisiologis, penyimpanan, periode simpan ABSTRACT The research aims was to study the effect of packaging on physiological seed quality of bunching onion (Fragrant variety) that have been stored for some period. The research was conducted from 2011 to 2012. A Factorial Completed Randomized Design with two factors was used i.e. , storage period (four (4), 8 (eight) and 12 months) and the type of packaging (bag-tight aluminum foil, aluminum foil bags, paper bags and plastic clips) with four replications. The observations were made on seed germination, speed of germination, hypocotyl length, and seedling growth rate. The results showed that the bunching onion seed physiological quality is affected by the store period and type of packaging. At room temperature, physiological quality of bunching onion seeds decreases with increasing storage period using all types of packaging.  Key words: bunching onion seed, fisiology quality, storage, storage periodÂ
