Effect of 3-Day Storage and Temperature on ADP-induced Platelet Aggregation Puspita, Nanda Ayu; Suryawati, Suryawati

Abstract

Objective: To observe platelet functions during the 3 days of platelet storage and to evaluate the effect of storage temperature towards platelet aggregation response. Methods: Two conditions of platelet storage were used in this study: at room temperature and in a cold condition at 4 °C to observe the temperature effect on platelet aggregation. The aggregation test was performed using a 96-wells plate platelet aggregation method. Results: At room temperature on day 1, the response of platelet aggregation reduced to 34.9±10.6%, which was less than half of the aggregation of fresh platelet (100%). The aggregation on day 2 reduced modestly (26.8±4.2%), in comparison with that seen on the first day. On day 3, platelet aggregation deteriorated significantly (6±1.2%), which was comparable to the control group without the presence of platelet agonist (6.1±1.4%). When platelets are stored at 4 °C, the aggregation response to ADP stimulation was slightly different to that shown by platelets stored at room temperature (22 °C). At the 4 °C, the aggregation response was 68.11±34.2%, 57.1±21.4%, and 5.9±2.6% at day 1, day 2, and day 3, respectively. The result showed that storing the blood at 4 ˚C or room temperature was not able to preserve platelet function, as the deterioration of platelet aggregation response over time was still taking place. Although was not statistically significant, our result indicated that cold storage reduced platelet responsiveness toward ADP activation. Conclusions: This current study provides evidence of the deterioration of platelet function during first 3 days of platelet storage. Moreover, we found that storing platelets in 4 °C showed no significant benefit in preserving the ADP-induced platelet aggregation capacity compared to that stored in 22 °C. Abstract

Medicinal plants have been used worldwide as supportive therapy in reducing symptoms of various diseases, including COVID-19 symptoms. The objective of this study was to investigate the use of medicinal plants and herbal products in respondents who experienced symptoms of COVID-19 in Aceh, Indonesia. A cross-sectional study was conducted, involving 384 respondents from the age group over 18 years old residing in Aceh Province, Indonesia. Data was collected via an online questionnaire consisting of 4 structured questions to assess the types and names of medicinal plants, the time of use, and the purpose of using medicinal plants. The results showed that nearly half of the study population (181 respondents) consumed medicinal plants and commercial products, mainly for the reason to enhance their immunity (49.7%), followed by to overcome the COVID-19 symptoms (26.7%). Amongst all medicinal remedies mentioned in this study, Ginger (Zingiber officinale) and turmeric (Curcuma longa) are the most widely used, which were consumed by 65.5% of medicinal plant and herbal products users. The other medicinal plant chosen by the study population were lemon (Citrus lemon), Java Ginger/temulawak (Curcuma zanthorrhiza), and Habbatus sauda (Nigella sativa). Although the majority of the respondents had used the herbal remedies prior contracting COVID-19 symptoms (45.5%), a third of the respondents consumed traditional medicine during the symptomatic period only (less than two weeks). This study has demonstrated the characteristics of the consumption of medicinal plants and their commercial products in Aceh, during the occurrence of COVID-19 symptoms in the study population.

Abstract

Diarrhea is an atypical state of the digestive system characterized by a soft or watery texture in bowel movements. Antibiotic-related diarrhea is common in the elderly due to extensive antibiotic use. Probiotics are vital microorganisms that support the intestinal flora and reduce bacterial colonization in the intestinal wall. This study aimed to assess the effectiveness, type, and dose of probiotics for diarrhea in the elderly. A systematic review with meta-analysis was conducted using PubMed, ScienceDirect, and Google Scholar. Seven records with a total of 2,087 participants were included. A quantitative analysis was carried out using Review Manager version 5 software. A meta-analysis was conducted to assess the frequency of diarrhea. The results showed that using probiotics significantly reduced the risk of antibiotic-related diarrhea 0.53 times compared to the placebo administration (OR 0.53; CI 95% 0.29 to 0.98; I2 = 70%). The type of probiotics frequently given was the genera of Lactobacillus, Bifidobacterium, and Streptococcus, with consumption durations varying from 3 days to a maximum of 4 weeks. The dose of probiotics ranged from a minimum of 1.0 × 10⁶ CFU to a maximum dose of 2 × 10¹⁰ CFU. To conclude, probiotic administration is more effective than placebo in reducing the risk of antibiotic-related diarrhea in the elderly.

Abstract

Perkembangan teknologi memberikan dampak yang positif pada berbagai bidang, termasuk bidang pendidikan. Internet merupakan bagian dari teknologi informasi yang dapat menyediakan berbagai informasi tentang lembaga pendidikan, baik formal maupun nonformal, sesuai dengan kebutuhan masyarakat. Di Indonesia terdapat lembaga pendidikan nonformal, salah satunya Alam Privat Indonesia. Ditengah perkembangannya saat ini, sangat penting dalam meningkatkan user experience pada website agar dapat meningkatkan pengalaman pengguna dan menciptakan branding atau identitas perusahaan yang kuat. Evaluasi dilakukan dalam beberapa tahap menggunakan User Experience Questionnaire (UEQ) untuk mengidentifikasi area pengalaman pengguna (user experience) yang memerlukan perbaikan. Selain itu, implementasinya dilakukan dengan pendekatan metodologi Human Centered Design (HCD) guna meningkatkan nilai usability yang meliputi efisiensi, kepuasan pengguna, dan efektivitas. Skor rata-rata UEQ dari hasil penelitian ini adalah 1,94 (Excellent) untuk aspek attractiveness, 1,67 (Above Average) untuk aspek perspicuity, 1,81 (Good) untuk aspek efficiency, 1,61 (Good) untuk aspek dependability, dan 1,83 (Excellent) untuk aspek stimulation, serta 1,07 (Above Average) untuk aspek novelty. Hasil ini menunjukkan bahwa prototype yang dihasilkan mengalami peningkatan dalam semua aspek UEQ dan prototype dapat dikatakan berhasil.

Abstract

Objective: To evaluate morphological changes of breast cancer cell line MCF-7 following the development of acquired resistance towards cisplatin. Resistance towards anticancer agents still plays an important role in the breast cancer chemotherapy failure.Methods: The cisplatin resistant subline MCF-7/CisR was developed in-vitro by cultivating the parental cell line cisplatin-sensitive MCF-7 with raising concentration of cisplatin (from 0.01 to 2.3 µM) for four months. The cell morphology was observed by giemsa staining. Results: The resistance was shown by the increase of IC50 by 9 folds on MCF-7/CisR compared to the initial IC50 value of MCF-7. The change in cisplatin cytotoxic potency on the resistant cell line was accompanied by the morphological modification, including the enlargement of cell size, the increase of nucleus and cytoplasm ratio, and the increase of the number of microvesicular and cytoplasmic granules.Conclusions: This result has supported the underlying mechanism of cisplatin resistance, including the ability of the cells to decrease intracellular cisplatin concentration and repair DNA damage effects.
