Abstract

Latar belakang: Pencemaran logam berat meningkat seiring dengan meningkatnya perkembangan industri. Paparan tembaga dalam jumlah besar dapat menyebabkan kerusakan ginjal dan hati. Berdasarkan penelitian terdahulu rimpang kunyit putih (Curcuma zedoaria) memiliki manfaat diantaranya sebagai antioksidan, antiinflamasi, antiproliferasi, antikanker, dan antimikroba. Kurkumin yang terkandung dalam rimpang kunyit putih pada penelitian sebelumnya digunakan sebagai senyawa nefroprotektif.Tujuan: Penelitian ini bertujuan apakah rimpang kunyit putih (Curcuma zedoaria) memiliki efek nefroprotektif dengan paparan CuSO4 pentahidrat. Metode: Penelitian ini adalah penelitian eksperimental dengan rancangan the post test only control group design. Pengambilan sampel pada penelitian ini menggunakan teknik simple random sampling. Tikus putih jantan galur wistar dibagi menjadi enam kelompok; kontrol normal, kontrol kunyit putih (45 mg/ 200 g), kontrol CuSO4 pentahidrat, dan kelompok co-treatment (curcumin selama 14 hari diikuti CuSO4 selama 5 hari). Hasil: Data dianalisis menggunakan uji One Way Anova diikuti uji Post-Hoc LSD. Untuk Kelompok 4 (kelompok preventif) kadar ureum kreatinin serum tidak ada perbedaan yang bermakna dengan Kelompok 1 dan 2 (p > 0,05) dengan kerusakan struktur histologis yang ringan. Namun, didapatkan perbedaan yang bermakna (p < 0,05) pada Kelompok 3, 5, dan 6. Kesimpulan: Dosis ekstrak kunyit putih 45 mg/ 200 g menunjukkan adanya penurunan kadar ureum kreatinin serum dan kerusakan histologi jaringan ginjal yang ringan.

Abstract

Photoaging is caused by exposure to sunlight and ultraviolet radiation (UV-B) through the reactive oxygen species (ROS) molecules formation, resulting in decreased levels of collagen. It is characterized by wrinkles, sagging and rough surfaces. Black cumin has been used in medicine due to its high antioxidant potential. The aim of this study is to identify the effect of black cumin cream on collagen levels and hydration of the skin of Rattus norvegicus exposed to ultraviolet-B rays. This is experimental study in which 30 male Wistar strain rats were classified into 6 sub-groups, namely the KN group (without treatment), KP (getting Nivea cream moisturizer), P1 (basic cream ), P2 (black cumin seed extract cream 0.25%), P3 (cream 0.5%) and P4 (cream 0.75%). P4 rat collagen levels increased significantly before and after the intervention (24 ± 2.23 vs 42.6 ± 14.6, p <0.05).Cream with lower concentrations did not provide a significant increase (p> 0.05). P4 rats also showed increased skin hydration (38.6 ± 19.3 to 63.0 ± 4.47 (p <0.05)), and this was not found in other groups. The administration of topical black cumin seed extract 0.75% can inhibit decreased levels of collagen and increase skin tissue hydration.

Abstract

Antioxidants are chemical compounds that can donate one or more electrons to free radicals, so that these free radicals can be quenched. There are two sources of antioxidants, namely natural and synthetic antioxidants. Andaliman (Zanthoxylum acanthopodium DC.) is one of the many spice plants found in Toba Samosir and North Tapanuli Regencies, North Sumatra. Andaliman plants grow at an altitude of 1,500 m above sea level. Apart from North Sumatra. This research using method experimental research when collecting and processing andaliman fruit, making simplicia, making ethanol extracts and examining antioxidant activity tests using the ABTS free radical scavenging method (2,2'-azino-bis(3-ethylbenz-thiazoline-6-sulfonic acid) and antibacterial activity. using the minimum inhibitory concentration method. Testing the antioxidant activity of the ethanol extract of andaliman fruit with the ABTS method using a UV-VIS spectrophotometer with a wavelength of 734 nm. the regression equation (y = 0.4619 + 20.226) obtained an IC50 value of 64.4598 mg/mL which can be classified as a strong antioxidant activity. The results of the antibacterial activity test of the ethanol extract of the andaliman fruit showed that the extract had antibacterial activity in the moderate category at a concentration of 300 mg/ml with an inhibition zone of 10.
