Abstract

The use of paracetamol in excessive doses can increase the hepatotoxicity effect, which causes acute hepatocellular injury. Dewa leaves (Gynura pseudochina (L.) DC) contain flavonoids, saponin, and tannin. This study aimed to determine the effect of ethanol extract from dewa leaves in reducing the impact of paracetamol induction on SGPT and SGOT levels and liver histology of white rats (Rattus norvegicus). The test was grouped into five treatment groups, namely: normal control; negative control with paracetamol induction 315mg/KgBW; Dewa leaf extract was administered with three variations in dosage, namely 3.15mg/KgBW, 3.50mg/KgBW and 3.85mg/KgBW and each treatment group consisted of 5 rats as replications. Dewa leaf extract was administered to the experimental animal, followed by the administration of paracetamol 8 hours after the extract was given orally; both treatments were given for 14 days. Measurement of SGPT and SGOT levels uses the photometric method. The hepatoprotective effect was further verified by histopathology of the liver. Liver histology was made using the paraffin method. SGPT levels, SGOT levels, and Hepatocyte Damage Scores were analyzed using Analysis of Variance (ANOVA); if the results are significant, continue with the Least Significance Difference (LSD) test at a significance level of 5%. The results of this study show that the administration of dewa leaf extract was not able to significantly reduce SGPT and SGOT levels in rats. The administration of dewa leaf extract at doses of 3.50mg/KgBW and 3.85mg/KgBW significantly prevented histological damage to rat livers.

Abstract

Praktikum menjadi salah satu sarana dalam peningkatan kompetensi IPA bagi siswa di sekolah. Implementasi dari praktikum dapat memberikan dampak positif dalam peningkatan kompetensi karena pembelajaran akan lebih berkesan dan mudah dipahami jika dilaksanakan dengan aksi yang dikerjakan langsung oleh siswa. Hal tersebut yang mendasari sekolah, baik di tingkat SMP maupun SMA, melakukan banyak praktikum untuk meningkatkan kompetensi IPA yang dimiliki muridnya. Berdasarkan dari hasil survei yang telah dilakukan, banyak implementasi praktikum yang selinier dengan pembelajaran IPA di jenjang SMP sehingga program kerja yang dilakukan berfokus pada kegiatan praktikum yang dapat menunjang peningkatan kompetensi siswa di SMP Djama’atul Ichwan, Surakarta. Program-program tersebut terdiri dari 5 program kerja yang satu di antaranya merupakan program kerja sama dengan dosen Grup Riset Biomaterial Hewan Prodi Biologi FMIPA UNS berupa pendampingan praktik mikroskopis. Kelima program kerja tersebut di antaranya, sosialisasi pengelolaan limbah organik melalui praktikum pembuatan eco-enzyme; praktikum pembuatan insektarium dalam upaya meningkatkan pembelajaran klasifikasi dan sistematika hewan; praktikum pengamatan morfologi dan anatomi hewan uji laboratorium; praktikum pembuatan preparat sederhana sebagai objek pengamatan dengan mikroskop; serta kegiatan mini project yang menekankan skill kerja sama dan kepemimpinan antarsiswa dengan mengimplementasikan pembelajaran IPA yang telah didapat.

Abstract

Komposisi media tanam yang tepat akan menentukan pertumbuhan bibit tanaman sehingga perlu dilakukan percoban yang dapat memberikan informasi efektifitas pengaruh media tanam terhadap pertumbuhan bibit tanaman tersebut. Tujuan penelitian adalah untuk mengetahui pengaruh media tanam topsoil, serabut sawit dan kombinasinya dalam menunjang pertumbuhan tunas stek sungkai (Peronema canescens JACK). Stek dari batang ortotrop (tunas vertical). Stek tidak terlalu tua dan muda. Parameter yang diukur adalah persentase tumbuh dan pertumbuhan panjang tunas. Rancangan percobaan menggunakan pola rancangan acak lengkap (RAL) dengan 3 perlakuan media tanam yaitu topsoil (P0), media tanam serabut sawit (P1) dan kombinasi topsoil 50% dan serabut sawit 50% (P2) dengan 15 kali ulangan. Hasil penelitian: (1) Persentase tumbuh tunas stek sungkai (P. canescens JACK) media tanam P0= 66.67%, P1=73.33% dan P2=86.67%. (2) Rata-rata pertumbuhan panjang Tunas. P2=13.66 cm, P1=7.75 cm dan P0=4.93 cm. Kesimpulan: Media tanam topsoil, serabut sawit dan kombinasi topsoil 50% dengan serabut sawit 50% menunjukan pengaruh yang berbeda terhadap pertumbuhan tunas stek sungkai (P. canescens Jack). Media kombinasi topsoil 50% dengan serabut sawit 50% adalah media terbaik untuk pertumbuhan tunas stek sungkai (P. canescens Jack).

Abstract

Kandungan hara mineral pada media tumbuh dapat mempengaruhi pertumbuhan tanaman. Pemupukan merupakan cara untuk menambah kandungan hara mineral pada media tumbuh. Penggunaan Pupuk kimia dapat merusak tanah dan lingkungan. Penggunaan kompos dan teh kompos merupakan usaha alternatif penambahan hara mineral tanah yang ramah lingkungan. Penelitian ini bertujuan untuk mengetahui pengaruh pupuk organik kompos dan teh kompos dengan konsentrasi (dosis) dan interval waktu pemupukan terhadap pertumbuhan tinggi dan diameter tanaman cabai rawit. Penelitian ini dilakukan dengan menggunakan Rancangan Acak Lengkap dengan pola Faktorial dengan 3 faktor yaitu faktor A yang terdiri dari 2 taraf (Kompos dan Teh Kompos), faktor B yang terdiri dari 3 taraf (0 gram, 150 gram dan 300 gram) dan faktor C yang terdiri dari 3 taraf (1 minggu, 2 minggu dan 3 minggu). Hasil penelitian ini adalah perlakuan jenis pupuk organik berpengaruh sangat nyata dan interaksi perlakuan jenis pupuk organik dengan interval waktu berpengaruh nyata terhadap rata-rata laju pertumbuhan tinggi dan diameter Penggunaan pupuk teh kompos ternyata menunjukkan hasil rata-rata laju pertumbuhan tinggi dan diameter tanaman cabe rawit yang lebih tinggi dari pada pupuk kompos. Interval waktu pemberian pupuk teh kompos yang berdampak baik terhadap rata-rata laju pertumbuhan tinggi tanaman cabe rawit sebaiknya 2-3 minggu sekali, sedangkan interval waktu pemberian pupuk kompos yang berdampak baik terhadap rata-rata laju pertumbuhan tinggi tanaman cabe rawit sebaiknya 1 minggu sekali.

Abstract

The use of the main growth medium in the form of wood sawdust in mushroom domestication is still higher than that of other organic materials, even though its availability is limited, so efforts need to be made to find a growth medium composition that minimizes the percentage of sawdust use. The purpose of this study was to determine the comparison of treatment 1 (80% teak sawdust, 10% bran, 5% tapioca flour, and 5% agricultural lime) with treatment 2 (70% teak sawdust, 15% bran, 10% tapioca flour, and 5% agricultural lime) on the percentage of growth and mycelium growth of white oyster mushrooms.  Composting of growing media was carried out for 5 days, then baglog making was carried out, after which the baglog was sterilized for 4 hours. After sterilization, the baglog was cooled and inoculated. After inoculation, the baglog is incubated to grow mycelium. Then, after 15 days, data collection and calculation were carried out. The parameters measured were the percentage of growth and mycelium growth of white oyster mushrooms in baglogs. Results showed that the percentage of growth and mycelial growth of white oyster mushrooms in treatment 1 is better than in treatment 2. Reducing the percentage of the main growing medium of teak sawdust and adding the percentage of additional growing media of bran and tapioca flour has a negative impact on the number of baglogs that grow mycelium (percentage of growth) and the growth of white oyster mushroom mycelium. This means that reducing the main media of teak sawdust and adding additional media is not the right growing media composition for domestication of white oyster mushrooms, so it cannot be used as an alternative growing media composition to reduce the use of wood sawdust.

Abstract

Kaliandra (Calliandra calothyrsus Meisn) is a plant capable of producing large amounts of biomass, potentially as a renewable energy source that supports the reduction of dependence on fossil fuels. The transition from fossil fuels to renewable energy is an urgent and long-term solution, as the use of fossil fuels as an energy source contributes to global warming and climate change.This study aims to evaluate the effect of compost fertilizer dosage and compost tea on the growth of height and diameter of kaliandra seedlings. Using a one-factor complete randomized design (CRD) method with treatment of compost fertilizer doses K0 (0 grams), K1 (60 grams), K2 (75 grams) and K3 (90 grams) and compost tea doses TK1 (0 ml), TK1 (50 ml), TK2 (75 ml) and TK3 (100 ml). Research results: 1. The treatment of compost doses and compost tea showed significant differences in the height growth of calliandra seedlings. 2. Compost fertilizer with a dose of 90 grams (K3) showed a better height growth rate than compost tea fertilizer with a dose of 100 ml (TK3), 75 ml (TK2), and 50 ml (TK1). 3. Compost tea fertilizer with doses of 50 ml (TK1) and 75 ml (TK2) showed a better height growth rate than compost fertilizer with doses of 60 grams (K1) and 75 grams (K2). 4. The compost fertilizer dose treatment showed a significant difference in the growth rate of Calandra seedling diameter, while the compost tea dose treatment did not show a significant difference. 5. Compost fertilizer with doses of 75 grams (K2) and 90 grams (K3) showed better diameter growth than compost tea fertilizer with doses of 75 ml (TK2) and 100 ml (TK3). 6.  The optimal dosage recommendation in this study is 90 grams of compost fertilizer and 75 ml of compost tea.

Abstract

Sengon buto (Enterolobium cyclocarpum) is a long-lived tree species that grows rapidly, is adaptable to various soil types, and is drought-tolerant, making it suitable for land conservation and rehabilitation. This study aimed to determine the effect of seed soaking duration of E. cyclocarpum in shallot (Allium cepa L.) extract on germination percentage, germination rate, and germination power. The research employed a Completely Randomized Design (CRD) with one factor and three treatments (A, B, and C) with 30 replications. The results showed that: (1) there was no significant effect between treatments; (2) germination percentages were: A = 36.66%, B = 40%, C = 13.33%; (3) germination rates were: A = 4.91 (t), B = 5.92 (t), C = 7 (t); (4) germination power: A = 53.33%, B = 73.33%, C = 80%. These results indicate that soaking in A. cepa L. extract did not have a significant effect on the germination parameters of E. cyclocarpum seeds

Abstract

The increased activity of the hyaluronidase enzymes speeds up the degradation of hyaluronic acid in the skin, leading to reduced elasticity and the formation of fine wrinkles, eventually contributing to skin aging. Mackerel scad (Decapterus macarellus) is a promising candidate as a skin anti-aging substance due to its particular amino acid composition. Native collagen and collagen peptides from mackerel scad skin were extracted using pepsine soluble collagen and hydrolysis with collagenase II enzyme. The amino acid profile of collagen was determined using HPLC analysis. An anti-hyaluronidase activity test was done using the spectrophotometry assay to express the content of N-acetyl glucosaminoglycan, and IC50 was calculated. Results showed that mackerel scad collagen contains 17 amino acids, with the highest content of glutamic acid, 8.20%; aspartic acid, 6.70%; glycine, 5.37%; arginine, 4.24%; and proline, 3.84%. The collagen from the extraction results had relatively low anti-hyaluronidase activity (IC50 326.05 ± 6.77 ppm). However, when it was broken down into smaller collagen peptides, the anti-hyaluronidase activity increased to IC50 100.78 ± 0.17 ppm. This indicated that the hydrolysis of collagen into collagen peptides with a smaller molecular weight increased its capacity to inhibit hyaluronidase. These findings suggest that D. macarellus collagen peptides have the potential to inhibit skin aging by inhibiting hyaluronidase enzyme activity.

Abstract

ABSTRAK Jamur tiram (Pleurotus sp.) merupakan salah satu jenis jamur yang digemari masyarakat. Jamur tiram merupakan komoditi yang mempunyai prospek sangat baik untuk dikembangkan. Salah satu varietas jamur tiram yang layak untuk dibudidayakan adalah jamur tiram merah muda (Pleurotus flabellatus). Usaha budidaya jamur tiram (Pleurotus sp.) yang dilakukan oleh petani sering menemui kendala dalam hal penyediaan bibit . Tujuan penelitian adalah untuk mengetahui persentase tumbuh dan laju pertumbuhan miselium P. flabellatus yang ditumbuhkan pada media PDA dan media campuran jagung dan dedak. Prosedur biakan murni (FO): penyiapan media PDA dan media campuran jagung dan dedak, sterilisasi, inokulasi, inkubasi, pengumpulan dan perhitungan data. Hasil penelitian: (1) Persentase tumbuh miselium P. flabellatus dengan media PDA adalah 60% sedangkan pada media campuran jagung dan dedak sebesar 80%. (2) Laju pertumbuhan miselium P. flabellatus pada media campuran jagung dan dedak lebih tinggi dari media PDA. Kesimpulan: Dari data persentase tumbuh dan laju pertumbuhan miselium P. flabellatus pada media campuran jagung dan dedak diketahui miselium P. flabellatus berhasil tumbuh 80% dan menunjukan laju pertumbuhan yang lebih baik dari media PDA, hal ini mengindikasikan bahwa tahapan pembuatan bibit biakan murni (F0) dapat dilakukan langsung pada media biji-bijian (jagung dan dedak) tanpa melalui media PDA yang seringkali tingkat kegagalannya tinggi.
