Abstract

Latar Belakang: Periodontitis merupakan penyakit inflamasi pada jaringan pendukung gigi yang disebabkan oleh kelompok bakteri yang mengakibatkan kerusakan progresif ligamen periodontal dan tulang alveolar sehingga terjadi resesi gingiva dan pembentukan poket periodontal. Epitel gingiva merupakan lapisan terluar dari gingiva yang memiliki peran dalam melindungi jaringan dibawahnya dari trauma mekanis, kimiawi dan termal serta invasi mikroba. Kerusakan tulang alveolar ditandai dengan aktivitas sel osteoklas, yaitu sel yang meresorpsi tulang alveolar. Daun Kari (Murraya koenigii) mengandung senyawa alkaloid, saponin, flavonoid, dan tanin. Senyawa flavonoid dapat menurunkan jumlah sel osteoklas sehingga dapat mengurangi resorpsi tulang. Tujuan: penelitian ini bertujuan untuk mengetahui pengaruh gel ekstrak daun Kari (Murraya koenigii) terhadap ketebalan epitel gingiva dan jumlah sel osteoklas pada tikus Wistar jantan yang diinduksi periodontitis. Metode: penelitian ini bersifat eksperimental laboratoris dengan menginduksi periodontitis pada tikus Wistar jantan selama 21 hari dan dibagi menjadi tiga kelompok yaitu kelompok perlakuan (gel ekstrak daun Kari), kontrol positif (metronidazole) dan kontrol negatif (akuades). Penghitungan ketebalan epitel gingiva dan jumlah sel osteoklas dilakukan dengan menggunakan mikroskop cahaya. Hasil: Analisis data menggunakan one-way Anova memperlihatkan terdapat perbedaan yang signifikan terhadap peningkatan ketebalan gingiva pada semua kelompok dan menggunakan uji Kruskal-Wallis terlihat adanya perbedaan yang signifikan terhadap penurunan jumlah sel osteoklas tikus Wistar jantan yang diinduksi periodontitis pada semua kelompok. Kesimpulan: gel ekstrak daun Kari 10% (Murraya koenigii) memiliki pengaruh terhadap peningkatan ketebalan epitel gingiva dan penurunan jumlah sel osteoklas pada tikus Wistar jantan yang diinduksi periodontitis, tetapi terdapat perbedaan yang signifikan terhadap kontrol positif.

Abstract

In 2018, the percentage of Indonesians who have gingivitis was 96.58%. Accumulation of plaque and Porphyromonas gingivalis bacteria cause gingivitis, an inflammation of the gingival tissue. Cananga (Cananga odorata), according to research by Anggraini (2021), has an antibacterial activity against Porphyromonas gingivalis. Gum chewing can boost saliva production and reduce bacterial-produced acid. This study aimed to provide an alternative for gingivitis prevention. In general, the three steps of this research are in vitro test, preclinical test, and clinical test. Using the well diffusion method, the antibacterial potential of the cananga flower extract at concentrations of 2.5%, 5%, 10%, 20%, and 40% was examined. Tetracycline 30 μg was the control (+), while ethanol 96% was the control (-). In preclinical test, Lethal Concentration 50% (LC50) was established using the Brine Shrimp Lethality Test (BSLT) method. The clinical test was done using a pretest and posttest control group design method for a week. According to the study's findings, the average inhibition zones were 9.06 mm, 10.53 mm, 12.2 mm, 13.43 mm, and 14.8 mm at doses of 2.5%, 5%, 10%, 20%, and 40%, respectively. Based on the preclinical test, the product material's LC50 is 850.191 ppm. The clinical test showed a decrease in the plaque index of the treatment group by 0.344375. This study concluded that cananga flower extract have antibacterial activity against Porphyromonas gingivalis and cananga flower extract chewing gum has the potential to be effective in preventing and lowering the plaque index that causes gingivitis.

Abstract

Introduction: High nicotine levels from smoking can affect salivary composition and function, which in turn can contribute to oral health problems such as dental caries and periodontal disease. Objective: The purpose of this study was to investigate the relationship between smoking habits and nicotine levels with salivary flow rate and pH in students of the Faculty of Engineering, Universitas Syiah Kuala. Methods: This study was a cross-sectional research design and examined 100 students of the Faculty of Engineering, Syiah Kuala University. Saliva collected by the spitting method. Results: The results of research on students of the Faculty of Engineering, Universitas Syiah Kuala, show that smoking habits and nicotine levels have a significant on the flow rate and pH of saliva in student smokers. Conclusion: There is a significant relationship between smoking habits and nicotine levels in cigarettes, with flow rate and salivary pH in smoking students. The habit of smoking affects the decrease in salivary flow rate and changes in salivary pH, with a moderate to strong strength of relationship.
