Abstract

Cyclooxygenase is an enzyme that plays a role in the formation of prostaglandins, which can cause inflammation and pain when overexpressed. This study aims to determine the interaction between COX-2 macromolecules (receptors) with their ligands, namely indomethacin and diclofenac in silico using the Molecular Docking method. The COX-2 receptor was downloaded in the form of a 3D structure from the RCSB GDP with code 5F19. Diclofenac Ligand and Indomethacin were downloaded in the form of a 3D structure from the RCSB GDP with 4ZBQ code and 4IK7 code. The results showed that the interaction between COX-2 and indomethacin was the interaction of hydrogen, which linked indomethacin with amino acid Leu531 and steric interactions between indomethacin and amino acids Trp387, Tyr385, Tyr355, Leu352, and Val523. The interaction of [COX-2 – Indomethacin] produces a value of ∆G of -103.136 kcal/mol, and the value of RMSD is 1.244 Å. Whereas, the interaction that occurs in COX-2 with diclofenac is the steric interaction that happens between diclofenac with amino acids Leu390, Trp387, Gln203, His388, His207, and Thr206. The interaction parameter between [COX-2-Diclofenac] obtained ∆G value of -7.843 kcal/mol and an RMSD value of 2.07851 Å.

Abstract

Xilanase merupakan enzim ekstraseluler hasil isolasi dari jamur Trichoderma viride yang mampu menghidrolisis xilan menjadi xilosa. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan Zn2+ terhadap aktivitas xilanase. Pada penelitian ini digunakan serbuk kulit pisang sebagai induser untuk produksi enzim. Aktivitas xilanase ditentukan berdasarkan banyaknya gula pereduksi yang dihasilkan (xilosa) oleh 1 mL enzim per menit. Pengaruh Zn2+ terhadap aktivitas xilanase dilakukan dengan beberapa konsentrasi, yaitu 25, 30, 35, 40, 45 dan 50 mM. Analisa data menggunakan analisis ragam Pola Rancangan Acak Lengkap (RAL). Hasil penelitian ini menunjukkan bahwa aktivitas ekstrak kasar xilanase dengan penambahan ion Zn2+ 40 mM diperoleh sebesar 1,821 µgmL-1menit-1 dan Zn2+ berpengaruh sangat nyata terhadap aktivitas xilanase (P<0,01). Penambahan Zn2+ berfungsi sebagai aktivator untuk meningkatkan aktivitas xilanase. Nilai konstanta kinetika tanpa penambahan ion Zn2+ diperoleh Vm sebesar 5,32 µgmL-1menit-1 dan KM sebesar 8,05 %, sedangkan pada penambahan Zn2+ diperoleh Vm sebesar 6,80 µgmL-1menit-1 dan KM sebesar 2,87 %. Abstract

Sabun pembersih wajah umumnya mempunyai kemampuan daya bersih yang baik, khususnya pada kulit yang berjerawat. Salah satu bahan alam yang membantu pembersihan tersebut ialah daun jambu biji . Ekstrak daun jambu biji memiliki kandungan beberapa metabolit sekunder, seperti golongan tanin, triterpenoid, glikosida dan flavanoid, yang memiliki aktivitas antijerawat. Selain itu, pemilihan bentuk sediaan juga mempengaruhi daya bersih suatu sabun pembersih. Oleh karena itu, pada penelitian ini telah dilakukan pembuatan emulgel sabun pembersih yang berasal dari daun jambu biji serta dilakukan evaluasi fisik sesuai spesifikasi dan persyaratan yang ditentukan. Hasil penelitian menunjukkan emulgel sabun pembersih dengan kadar ekstrak 9,779% memenuhi spesifikasi evaluasi organoleptis, pH, viskositas, daya sebar, daya bersih, dan tinggi busa. Namun, uji homogenitas masih belum memenuhi persyaratan sehingga diperlukan optimasi terkait pemilihan teknik pembuatan.

Abstract

Usually, papaya leaf can be used as malaria disease treatment, prevent acne, appetite enhancer, milk supplement even making a toothache. In this research, papaya leaf is made as snack chips. The ingredients of papaya leaf’s chips are papaya leaf’s, rice flour, tapioca flour, red chili pepper, salt, eggs, guava leaf’s, clay, coriander, candlenut, flavorings, cooking oil, garlic, and water. The papaya chips also treat by organoleptic tests that consist of 5 criteria includes color, smell, texture, appearance, a taste of spices and moisture content test. The process of making papaya leaf’s chips include wash the papaya leaf, cutting, boiling used clay and guava then add salt to remove the bitter taste. The next step is making flour dough, frying, and slicing. The results of the organoleptic test 90% mean that consumers are very like papaya chips and the results of moisture content test are 4.564%.

Abstract

Instant drinks, which are often consumed by the public, are mostly sweetened drinks that contain high calories and artificialsweeteners and can cause various diseases, such as obesity, cancer, diabetes and others. This can be controlled and prevented by consuminglow-calorie sweeteners. Stevia is a plant that has a sweetness of 300 times that of sucrose, so it can be used as a low-calorie sweetener, suchas in the functional drink Pletok beer. This study aims to identify glycosides and to evaluate the physical evaluation of the low-caloriesweetener formula for Stevia (Stevia rebaudiana B.) in the functional beverage of Bir Pletok. Physical evaluation and hedonism test werecarried out on 20 panelists and 1 expert panelist, then analyzed descriptively. In this study, several tests of instant preparations were carriedout, namely organoleptic test, pH test, specific gravity test, movable volume test and hedonism test on 20 panelists and 1 expert panelist. Theresults of the research that have been done show that the leaves of Stevia (Stevia rebaudiana B.) contain diterpenoid glycosides and based onall test preparations the best formula of the three formulas is Formula II. Abstract

One of the indicators used in assessing the success of drug use is prescription. Fluoroquinolone antibiotiks are one of the most frequently prescribed drugs. if the use is too high it will cause antibiotik resistance. The purpose of this study was to determine the overview of fluoroquinolone antibiotik prescription in outpatients at Rumah Sakit Ibnu Sina Gresik with a descriptive observational study design that carried out retrospective secondary data collection. This study used a population of all outpatient prescriptions who received fluoroquinolone class antibiotik therapy on February 1, 2020 - February 29, 2020 at the Outpatient Pharmacy Installation of Rumah Sakit Ibnu Sina Gresik with a sample size of 311 sheets of prescriptions taken by total sampling technique. It can produce the highest prescription profile from each of the analysis variables, namely 1 antibiotik item of 88.75%, lung clinic 21.87%, capsule preparation 58.80%, over 55 years of age 47.91%, generik fluoroquinolones 81.67% BPJS patients were 67.52% and fluoroquinolone second generation 69.77%. These results can be seen from the overview of outpatient prescriptions at Rumah Sakit Ibnu Sina Gresik on February 1 - February 29, 2020. Keywords: Prescribing, Antibiotik, Fluoroquinolone

Abstract

Coronavirus Disease 2019 (Covid-19) is a new type of disease that has never been identified before in humans. The virus that causes Covid-19 is called Sars-CoV-2. This research was conducted to educate the public about the use of ginger and lemongrass functional drinks to improve immunity. The method of implementing Community Service is carried out interactively with a video conversion application ZOOM on 25 youth cadets in the village of Mojogede, Balongpanggang Gresik on June 14, 2020. Before starting the event the participants were given a face shield that must be used during the event as an effort to implement health protocols. Furthermore, pre-test is given first, then material exposure is carried out and continued filling in post-test with the same problem. Based on the results of the pre-test and post-test obtained an increase in public knowledge which was 79.09% to 89.23%.

Abstract

At this time, community does not understand classes of drugs that can be purchase or stored at home. This can have two impacts, positive and negative. The positive impact is community become more responsive to health, while the negative impact is increasing use of drugs which raises problems related to how to get, use, store and dispose of drugs properly or commonly known as Dagusibu. This community service activity aims to educating housewives as "pharmacists" in the household regarding how to get, use, store, and dispose of drugs properly in Kedanyang Village RT/RW 003/003, Kebomas, Gresik. Method that we use in this community service is counseling and training using property and followed with monitoring. We use questionnaire for monitoring a month after counseling and training. Our method to counseling and training involve several variable, including Question of 5”O”, Get, Use, Save and Discard (Dagusibu). The result shows that this activity is effective with the percentage of correct answer for each variable are above 75%, including question about 5”O" as much as 97%, get 84.38%, use 87.14%, save 76%, and waste 93%. This proved that the method chosen in this community service is appropriate, where the community still remember and understand.

Abstract

Xylanase is a hydrolase enzyme that can hydrolyze hemicellulose into xilo-oligosaccharide and xylose. This research is aimed to investigate the in vitro and in silico xylanase (isolated from the fungus Trichoderma viride) immobilized on the zeolite matrix activated with HCl 0.4 M solution. The study was performed using in silico docking molecular methods to investigate the interaction between the xylanase primarily to its ligand. Xylanase activity was determined by reducing sugar produced (xylose) by 1 mL of enzyme per minute. The optimum conditions of immobilized xylanase were measured according to the time agitation and concentration of xylanase. The time variation agitation used were 1, 2, 3, 4, and 5 hours, while variations in the concentrations of a xylanase used were 11.500, 15.653, 20.444, 25.875, and 31.944 mg/mL. The optimum conditions of immobilized xylanase was obtained in the shaking time for 3 hours at a concentration of xylanase 15.653 mg/mL and immobilized xylanase activity generated at 46.755 μg.g-1.min-1. Immobilized xylanase activity was greater than the purified xylanase (15.976 μg.mL-1.min-1). These results were due to the cofactors Al (AlO4) and Si (SiO4) of zeolite was able to increase the kinetic energy caused the reaction rate between xylanase with the larger substrates. Cofactor also increased the kinetic energy and can enhance the rate of reaction between a xylanase with its substrate, in order to give greater activity. Immobilized xylanase was stable and its reusability as much as 6 times which afforded the activity 21.331 μg.g-1.min-1 and the efficiency of 56.77%.
