Abstract

Using a removable denture can caused chronic inflammation commonly triggered from oral microorganism, specifically Candida albicans. The prevalence of denture stomatitis on removable denture user has been reported to be 15-70%. Yakult® probiotic drink contains Lactobacillus casei Shirota that is known for its role in interfering the binding of pathogen microorganisms. The purpose of this study is to determined whether Yakult® probiotic drink can decrease the number of Candida albicans on heat-cured acrylic resin plate. This study used pre-post test control group design method with n=25, consisting of 5 groups. Acrylic plates are contaminated with Candida albicans and initial counting was conducted. Then the plates were immersed in Yakult® probiotic drinks on 15, 30, 60 minutes treatment groups and on Fittydent® and aquadest control groups. Afterwards the plantation and final counting were done. The result of the study is normally distributed and homogenous, then it was analyzed using Paired T-Test (p<0,05) with mean P1 12,20±4,38, P215,60±5,72 and P315,80±3,76 CFU/ml. LSD test shown significant differencesbetween positive controls with all treatments and not significant differences between treatments. This study shows that probiotic Lactobacillus can inhibit the growth of Candida albicans. It can be concluded that Yakult® probiotic drink can decrease the number of Candida albicans colonies on heat-cured acrylic resin plates immersion, with the most effective time being 60 minutes.

Abstract

Introduction: Denture stomatitis is a chronic inflammation caused by wearing dentures, mainly caused by Candida albicans. Heat-polymerized acrylic resins are often used as denture bases, but lack the ability to absorb liquids due to their porosity and surface roughness. The basis of the latest dentures is thermoplastic nylon because it is more aesthetically, hypoallergenic and more flexible. Soursop leaf extract contains alkaloids, tannins, flavonoids, and saponins as antifungal. The purpose of this study was to examinate whether there were differences in the number of Candida albicans colonies on heat-polymerized acrylic resin plates and thermoplastic nylon in soursop leaf extract immersion. Materials and Methods: The research design was an experimental pre-post test with control group design, n = 50 plates were divided into 2 groups, heat-polymerized acrylic resin (n=25) and thermoplastic nylon (n=25), divided into 5 groups, 3 treatment groups using 10%,15%,25% soursop leaf extract, Fittident® and aquadest in the control groups. Samples were contaminated with Candida albicans suspension and incubated, counted before immersion for 8 hours, then put in Saboroud’s bath and counted again. Results and Discussions: Wilcoxon test analysis showed significant differences in all groups, except the aquadest group. The Mann-Whitney test showed significant differences between the treatment groups, except between the 25% extract and the Fittident® control group. Also, showed a significant difference in the 15% concentration group between heat-polymerized acrylic resin and thermoplastic nylon. Conclusion: There was a difference in the decrease in the number of Candida albicans colonies on heat-polymerized acrylic resin plates and thermoplastic nylon in soursop leaf extract at a concentration of 15%, while at concentrations of 10% and 25% there was no difference.

Abstract

Thermoplastic nylon is a denture base that is in high demand by dentists because it has several advantages over acrylic resin materials. Streptococcus mutans is one of the bacteria that can cause the formation of dental plaque and halitosis. Beetroot (Be-ta vulgaris L.) is a natural ingredient as an anti-inflammatory that contains phenol compounds, flavonoids, saponins, tannins, and betalains. Through this study, the effectiveness of beetroot extract on thermoplastic nylon denture users against the growth of S.mutans was determined. The experimental laboratory study used Fittydent® as positive control and sterile distilled water as negative control. Based on one-way Anova test, there was no significant difference between 10% and 40% beetroot extract (p>0.05). Immersion of thermoplastic nylon plates in 10% and 40% beetroot extract had an effect on the growth of S.mutans bacteria. This is due to the nature of thermoplastic nylon which easily absorbs water and phenol compounds contained in beet-root extract by diffusion. It is concluded that beetroot extract has the effectiveness in inhibiting the growth of S.mutans on ther-moplastic nylon plates.
