Abstract

The Tumor Protein P53 Inducible Nuclear Protein 1 (TP53INP1) gene is expected to be a crucial candidate for mineral content.  This research aimed to analyse the genotype polymorphism and investigate its association with the TP53INP1 gene and sheep mineral content. To analyse gene polymorphisms and conduct an association study, 30 rams of Javanese thin-tailed sheep were utilised. The NlaIII restriction enzyme was applied to investigate the genotype polymorphism of the TP53INP1 gene through Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP). Regarding the association with analysing TP53INP1, a General Linear Model (GLM) was used. The findings indicate that the TP53INP1 gene shows polymorphism, revealing three distinct genotypes identified by the results: AA, AG, and GG. The genotypes were detected using the Hardy-Weinberg Equilibrium (HWE) principle. According to the association analysis, a significant association (P < 0.05) was found between the TP53INP1 gene and mineral content, particularly iron (Fe). A higher mineral content was associated with the GG genotype, whereas a lower mineral content was related to the AA genotype. The TP53INP1 could be the candidate gene for sheep's mineral content.

Abstract

Pituitary Tanscription Factor 1 (POU1F1) has been identified as the pituitary spesific transcription factor that regulates the expression of the growth hormone (GH) and prolactin (PRL) in the anterior pituitary. The objective of this research is identification the polymorphism of POU1F1 gene of etawah grade goat in BPTU KDI-HPT Pelaihari South Kalimantan use Polymerase Chain Reaction- Restriction Fragment Length Polymorphism (PCR-RFLP). PCR products of POU1F1 digested with Hinf1 enzyme which restric G|AnTC.  Samples are used as the material of DNA from blood samples of 115 PE goats.  POU1F1 gene amplification was performed with annealing temperature of 62 oC for 20 seconds resulting in a fragment with the length of 601 bp which is located at intron 5 and exon 6. The result of PCR-RFLP analysis indicated that the POU1F1 gene was monomorphic.  B allele had 3 fragments at 275, 243, and 83 bp respectively.

Abstract

Etawah-grade goat is a graded up line between kacang and etawah goats, which well adapted to Indonesia’s humid tropical climate. The good reproductive trait of Etawah-grade does are inherited from kacang goat (prolific goat). This study used six heads of Etawah-grade does at 1–2 years old (I1 dental condition). IPB University. The body variables were withers height, hip height, body length, chest width, chest depth, thurl width, rump length, chest girth, and cannon circumference, whereas for head variables were acrocranion–prosthion, basion–prosthion, lower jaw length, head height, tuber facial left-right, nasion–rhinion, entorbitale left-right, euryon left-right, supraorbitale left-right. Body score and head score were obtained from Principal Component Analysis (PCA). PCR-RFLP technique was used to determine genotypes of fecundity genes BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1) and KISS1 (intron 1) with restriction enzymes AluI (AG|CT), SduI (GDGCH|C), BsrI (ACTGG|) and MwoI (GCCTAAG|TAGC), respectively. Progesterone leves were measured by EIA (enzyme immuno assay). The results showed that the association between crude fat consumption and progesterone profile was found in this study, but it was not related to twin births with certain genotypes of gene fecundity BMP15 (exon 1), BMPR1B (exon 1), KISS1 (exon 1) and KISS1 (intron 1). The progesterone profile during the pregnancy period did not correlate to their body size and head size.
