Abstract

Antibiotics have been widely used to treat tuberculosis (TB), yet the emergence of drug-resistant strains remains a serious global concern, highlighting the need for new molecular biomarkers to assess treatment success more accurately. Mycobacterium tuberculosis adheres to host tissues through the fbpA gene, which plays a vital role in bacterial virulence and persistence. The novelty of this study lies in exploring the fbpA gene as a potential biomarker for evaluating TB treatment response, differing from previous research that mainly targets drug-resistance genes such as rpoB, katG, or inhA. This study offers a new perspective by focusing on a virulence-related gene that may reflect bacterial survival and therapy outcomes, particularly in Indonesian clinical samples where such data remain scarce. The study aimed to assess the potential of the fbpA gene as a treatment biomarker by detecting a 900 bp fragment using molecular techniques. Ten sputum samples were collected from the Tuberculosis Unit of HUM-RC, Hasanuddin University Hospital, Makassar, and underwent sputum decontamination, Ziehl-Neelsen staining, acid-fast bacilli examination, DNA isolation, PCR amplification, and gel electrophoresis. The results showed the presence of DNA bands in all samples, with variations in thickness and intensity corresponding to DNA concentration. These findings suggest that the fbpA gene could serve as a rapid and reliable molecular biomarker for monitoring bacterial load and evaluating treatment efficacy, supporting the development of more precise diagnostic tools for multidrug-resistant TB management.

Abstract

AbstractBackground: Colorectal cancer originates from progressive genetic alterations in colorectal epithelial cells. While current therapies (surgery, radiotherapy, and chemotherapy) remain cornerstone treatments, chemotherapy often induces systemic toxicity, adverse effects, and acquired resistance. Sulfated polysaccharides (SPs) from the green alga Caulerpa racemosa demonstrate higher sulfate content than red algal derivatives, correlating with enhanced bioactivity. Despite their potential, SPs from green algae remain understudied compared to brown and red algal counterparts. This study evaluated the anticancer potential of C. racemosa SPs against colorectal cancer through viability inhibition and apoptosis induction.Materials and Methods: SPs were extracted via Microwave-Assisted Extraction (MAE) and characterized using iodine testing and FTIR spectroscopy. Cytotoxicity was evaluated in WiDr colorectal cancer cells using MTT assay after 24-hour exposure. Apoptotic mechanisms were investigated through in silico molecular docking targeting Caspase-3 activation.Results: SPs were confirmed by a blue color change and FTIR absorption at 1232 cm-1. At 100 µg/mL, low toxicity was observed based on abundant formazan crystals. Concentrations of 200–400 µg/mL showed predominant viable cells, whereas 500 µg/mL caused significant growth inhibition and cell death. In silico analysis demonstrated that SPs may induce apoptosis by Caspase-3 activation.Conclusion: SPs of C. racemosa inhibit colorectal cancer cell viability at a concentration of 500 µg/mL and may induce apoptosis via Caspase-3 activation.Keywords: apoptosis, Caulerpa racemosa, colorectal cancer, sulfated polysaccharides

Abstract

Edukasi tentang kesehatan gigi dan mulut masih perlu dilakukan terutama berkaitan dengan konsumsi makanan sehat yang dapat menjaga kesehatan gigi dan mulut terutama untuk balita . Penggunaan  probiotik dan prebiotik pada makanan dapat  menjaga kesehatan terutama untuk kesehatan gigi dan mulut. Solusi yang akan dilakukan dalam kegiatan pengabdian pada masyaakat ini adalah  sosialisasi tentang bahan pangan fermentasi yang mengandung probiotik dan prebiotik yang dapat digunakan untuk kesehatan tubuh , mengolah probiotik dan prebiotik menjadi produk fermentasi yang bahan bakunya dengan mudah diperoleh dan dapat dibuat dengan mudah oleh semua kalangan sehingga bisa dikonsumsi sebagai bahan pangan yang membantu meningkatan kesehatan gigi dan mulut serta upaya pencegahan penyakit pada gigi dan mulut. Metode yang dilakukan secara bertahap yang dimulai dengan edukasi tentang pentingnya perawatan gigi dan mulut untuk menambah pengetahuan masyarakat tentang perawatan kesehatan gigi dan mulut. Tahap selanjutnya dilakukan penyuluhan tentang probiotik dan prebiotik, demonstrasi pembuatan  pangan fermentasi. Hasil kegiatan menunjukkan masyarakat memahami perawatan gigi dengan mulut dengan sangat baik, tetapi untuk probiotik dan prebiotik umumnya para kader posyandu yang mengikuti kegiatan pengabdian ini belum memahami dengan baik manfaat probiotik dan prebiotik serta mengenali bahan pangan fermentasi.Selama kegiatan pengabdian para kader posyandu sangat antusias dalam mengikuti selutuh kegiatan pengabdian .
