Abstract

This study aims to determine the effect of adding fructose to egg yolk diluent on the quality of Landrace pig spermatozoa. This research used a completely randomized design (CRD) method with 6 treatments and 5 replications. The following are the various treatments, namely: T0=NaCl fis 2.4 mL + TY 0.6 grams + 0 grams of fructose, T1= NaCl fis 2.4 mL + TY 0.6 grams + 0.015 grams of fructose, T2= NaCl fis 2 .4 mL + TY 0.6 gram + 0.030 gram fructose, T3= NaCl fis 2.4 mL + TY 0.6 gram + 0.045 gram fructose, T4= NaCl fis 2.4 mL + TY 0.6 gram + 0.060 gram fructose, and T5 = NaCl fis 2.4 mL + TY 0.6 grams + 0.075 grams of fructose. Good quality cement is diluted using treatment diluent, evaluated post-dilution, packaged in ependop then stored in a cool box with a temperature of 18-20°C. Evaluation of spermatozoa motility, viability, abnormalities and survival is carried out every 8 hours. The research results obtained showed that the 24th hour of storage with treatment T0 (47.00 ± 10.95), T1 (51.00 ± 7.42), T2 (55.00 ± 6.12), T3 (54.00 ±4.18), T4 (50.00±3.54), and T5 (44.00±4.18) were able to stay above 40% which is feasible for IB. Statistical tests showed that the treatments did not significantly differ on the quality of landrace pig spermatozoa. It was concluded that the semen quality of landrace pigs could be optimally improved by using a fructose level in the diluent of 0.30 grams.

Abstract

Tujuan penelitian ini yaitu untuk menguji efek penambahan level kuning telur puyuh (KTP) dalam pengencer Tris-modifikasi (TM) terhadap kualitas semen cair babi duroc. Penelitian ini menggunakan metode eksperimental dengan rancangan acak lengkap (RAL) terdiri dari 5 perlakuan dan 5 ulangan sehingga terdapat 25 unit percobaan. Perlakuan yang dimaksud adalah P1 =90% T-M + KTP 10%; P2=87,5% T-M + KTP 12,5%;P3 : 85% T-M + KTP 15%; P4: 82,5% T-M + KTP 17,5%; P5 : 80% T-M + KTP 20%. Semen yang berkualitas diencerkan dengan pengencer uji, kemudian dipreservasi pada suhu 18-20°C hingga motilitasnya 40%. Variabel yang diteliti meliputi motilitas, viabilitas, abnormalitas, dan daya tahan hidup spermatozoa yang di evaluasi setiap 12 jam. Data yang diperoleh kemudian dianalisis mengunakan analys of variance (ANOVA) dilanjutkan dengan uji duncan. Hasil pengamatan menunjukkan pada jam ke 60 preservasi perlakuan P3 menghasilkan motilitas spermatozoa tertinggi dan yang berbeda secara nyata (P<0,05) dengan semua perlakuan dengan nilai motilitas spermatozoa sebesar 43,40%, viabilitas spermatozoa 52,62%, abnormalitas spermatozoa 5,15%, dan daya tahan hidup spermatozoa 68,50 jam. Kesimpulannya penambahan 15% kuning telur puyuh menghasilkan kualitas semen cair yang optimal selama penyimpanan pada suhu 18-20°C.

Abstract

Artificial insemination (AI) is an appropriate technology that can be used to increase the productivity of cattle. The application of AI technology can use frozen semen, fresh semen or liquid semen. Semen quality as an important factor in the success of AI is influenced by the cement processing process starting from the collection, dilution, and preservation of cement. This study was conducted to determine the effectiveness of young coconut water-egg yolk diluent added with astaxanthin in maintaining motility, viability, abnormalities and survival of Angus cattle spermatozoa. Semen was collected from an angus bull about three years old who had previously been trained to collect semen with an artificial vagina. Semen of good quality is diluted with coconut water-egg yolk (CW-EY) with the addition of astaxanthin (As) at doses: 0 (T0), 0.25g (T1), 0.50g (T2), and 0.75g (T3) ). Semen was stored at 3-5ºC after being diluted, and evaluation of the quality of spermatozoa was carried out every 24 hours. Each treatment was repeated five times to form 20 experimental units. The results showed that spermatozoa in CW-EY diluent with the addition of As 0.25g (T1) compared to the other three treatments had higher quality (P<0.05), namely with motility reaching (35.50 ± 2.12% ), viability (47.41 ± 3.26%), abnormality (6.16 ± 1.51%) and survival (5.00 ± 0.00 days). The conclusion in this study was that the addition of 0.25g of astaxanthin in CW-EY diluent was effective in maintaining the quality of liquid semen of Angus cattle.
