Abstract

The extraction of kratom (M. speciosa) leaf powder was optimized with preliminary extraction to be further optimized with the Box-Behnken experimental design. The individual and interactive effects of process variables (sample-to-solvent ratio, extraction time, solvent concentration) were assessed. The preliminary extraction results showed that ultrasound-assisted extraction (UAE) and methanol were chosen for further optimization. The experimental data were analyzed by Pareto analysis of variance (ANOVA) and second-order polynomial models were developed using multiple regression analysis. The model developed showed a good fit with the experimental data with a high coefficient of correlation (R2) and predictive ability (predicted R2). An optimization study was performed and the optimal extraction conditions were sample-to-solvent ratio value 1.5:10; extraction time of 10 minutes, and methanol concentration of 100%.

Abstract

Kratom (Mitragyna speciosa (Korth.) Havil.) is a plant that originated from the rainforest in Southeast Asia, mainly grows in Thailand, Malaysia, and Indonesia. Kratom has been used traditionally as an herbal remedy for the treatment of various illnesses. Kratom gained notoriety due to its potential as an analgesic, opiate withdrawal treatment, anxiolytic, antidepressant, and antidiabetic with an unclear risk of addiction and toxicity fueled by a false sense of security due to its identity as a member of the coffee family. This article is a narrative review on kratom to highlight its pharmacological and toxicological properties, and the analytical method of Kratom, especially its potential as an opioid withdrawal therapy and its risk of abuse.

Abstract

Peptida bioaktif merupakan fragmen protein spesifik yang tidak hanya memiliki fungsi sebagai nutrisi, namun juga memiliki efek positif pada fungsi, kondisi dan kesehatan tubuh. Peptida bioaktif dapat terbentuk selama proses proteolisis baik melalui hidrolisis enzimatik in vitro, fermentasi, maupun pemecahan selama pencernaan makanan di saluran cerna. Peptida bioaktif memiliki berbagai manfaat pada kesehatan manusia dengan mempengaruhi sistem pencernaan, endokrin, kardiovaskular, syaraf dan imunitas. Beberapa jenis peptida bioaktif juga telah diteliti dan diketahui memiliki aktivitas antihipertensi, antioksidan, imunomodulator, antimikroba, antihiperlipidemia, sitomodulator dan antigenotoksik. Peptida bioaktif juga merupakan komponen yang fleksibel dalam penggunaannya dikarenakan variasi rantai samping yang dapat berinteraksi dan bekerja pada berbagai jenis lingkungan. Beberapa tantangan dalam penggunaan bioaktif peptida adalah lingkungan asam pada lambung serta efek dari enzim protease pada lambung dan usus yang tentunya akan berdampak terhadap ketersediaan hayatinya. Selain itu, meskipun peptida dapat mencapai sirkulasi sistemik, peptida dapat mengalami inaktivasi yang menyebabkan peptida kehilangan aktivitasnya dan hanya berperan sebagai nutrisi. Peptida dengan berat molekul besar juga dapat mengalami kesulitan dalam proses penyerapannya di usus, menyebabkan hilangnya peptida yang bermanfaat dari sistem pencernaan. Meskipun begitu, peptida bioaktif memiliki potensi besar yang dapat dikembangkan. Review ini berfokus pada potensi peptida bioaktif sebagai pangan fungsional dan nutrasetikal.

Abstract

Hypertension is a chronic disease and a major risk factor for cardiovascular disease, often referred to as a "silent killer" due to its asymptomatic nature. Its prevalence in Indonesia is increasing, reaching 34.1% among individuals aged 18 years and over in 2018. Traditional herbal medicine can treat degenerative diseases, but its use is still limited due to inadequate public knowledge. This community service program aims to provide education about hypertension and the use of traditional herbal medicine in Dusun Beji, Bantul, Yogyakarta. The program includes health education, blood pressure checks, and promotion of herbal medicine for hypertension management. A total of 34 participants (85% attendance) attended the session, with 26 completing pretest and posttest questionnaires to evaluate increased knowledge. The majority of participants were elderly (?61 years, 50%) and female (69.2%), with 69.2% showing abnormal blood pressure levels. Educational materials, including presentations and brochures, were provided along with live demonstrations of herbal preparations. Post-test results showed a slight increase in mean score from 7.73 to 7.88, with increased homogeneity of data and an increase in median from 7 to 8. Although paired t-test showed no statistically significant difference (P=0.885), descriptive analysis showed an increase in understanding and awareness. Active participation during the discussion, especially regarding herbal preparations, reflected the community's interest in adopting alternative approaches to hypertension management. This initiative has succeeded in increasing awareness of hypertension and the benefits of herbal medicines to effectively manage the chronic disease and its complications.
