Abstract

Low quality of sperm after freezing and thawing process due to changes in extreme temperature and osmolarity. The sperm freezing capability and sperm membrane damage was to evaluate by measuring the levels of malondialdehyde (MDA) of  Simmental, Limousin and Friesian Holstein (FH) bull of a total of 10 bulls aged 4-8 years. Data were analyzed with a linear model (GLM) and Duncan s test. Results showed that breed influence sperm motility and MDA levels but not in the membrane integrity (MI) and viability. The FH bull had a low of recovery rate (RR) 57.53± 1.74% with high MDA level (0.81± 0.31 nmol/108 sperm level) and Limousine had the highest RR (59.70 ± 3.23% ) with the low MDA (0.52±0.25 nmol/108 sperm). Freezing decreased the sperm motility, viability and MI of all bulls. Sperm motility, viability and MI decreased by 28.32±1,45% and 29.73±1.54%, 21.58±4.09% and 22.55± 5.60% and 21.25±6.86% and 23.51±6.05 % respectively. Abstract

Present study aimed to determine the effectivenessof dimethylformamide (DMF)as cryoprotective agent (CPA) by determining its optimum concentration inTris Eggyolk (TEY) extender on the quality of post-thaw swamp buffalo semen. Only ejaculates having > 60% sperm motility were divided into eight s tubes. Four tubes were diluted in TEY with 4, 5, 6, 7% of glycerol and four other tubes were diluted in TEY with 4, 5, 6, 7% of DMF. Diluted semen then packed into ministraw (0.25 mL), equilibrated at 4°C for 4 hours and freezed at 8 cm above liquid nitrogen vapour for 10 minutes and then stored at liquid nitrogencontainer. After 24 hours of storage, straws were thawed at 37°C for 30 seconds. The result showed that sperm motility after equilibration tend to be the same at all four glycerol concentrations, whereas the decrease in motility was highest in extenders with a DMF concentration of 7% (P <0.05). Post-thaw sperm motility and recovery rate in TEY extender containing 5% DMF or 7% glycerol were highest (P<0.05) than other glycerol or DMF concentration. Sperm viability and membrane integrity did not differ among TEY containing 6% or 7% glycerol, both  were higher (P<0.05) than 4% or 5% glycerol. Viability and membrane integrity of sperm in 5% DMF was superior (P<0.05) to 4%, 6% or 7%. This research concluded that 5% DMF or 7% glycerol was considered to be the most suitable CPAs for swamp buffalo semen cryopreservation.

Abstract

The study was conducted to investigatethe effect of L-Carnitine supplementation in Tris-eggyolk (TEY) diluents to optimize the quality of Pasundan bull liquid semen. Semen samples were collected from three Pasundan bulls (6 – 7 years old)usinganartificial vagina. Semen samples were evaluated macroscopically and microscopically, semen having ≥70% progressive motility, ≥500×106ml-1 sperm concentration and ≤ 20% sperm abnormalities were divided into 5 equal part. Each part  wasdiluted with TEYand supplemented with0 mM(control), 1 mM, 2 mM, 3 mM and 4 mM L-Carnitine. Liquidsemen was storedat5°C.Sperm motility was evaluated every 12 h  objectively using Computer Asissted Sperm Analysis (CASA) until progressive motilityreached 40%. The best diluent was demonstratedby progressive motility (PM)by TEY supplemented with 1 mM L-Carnitine(43.08±0.49%) that remained up to 108 h compared tothecontrol group (43.63±0.70%) that remained up to 72 h (P<0.05). In conclusion, 1 mM L-Carnitine in TEY was the best concentration for preservation of Pasundan bull semen stored in the liquid form.

Abstract

The objectives of the present study were to compare and determine the best post-thawed characteristics of balinese bull sperm cryopreserved in three different extenders; animal based (Tris-clarified egg yolk (Tris-cEY)), and non-animal based extenders (Bioxcell® (lecithin based) and Optixcell® (liposome based)) in combination with three different equilibration times (30 minutes, 2 hours, 4hours). Thirty six ejaculates were collected from six Balinese bulls and frozen in three extenders (Tris-cEY, Bioxcell® and Optixcell®) after equilibration in three different times (30 minutes, 2hours and 4hours). Computer-assisted sperm analysis (CASA), hypo-osmotic swelling test (HOST) and eosin nigrosin staining were used in the post-thawed semen analysis. There was a significant interaction between equilibration time and extender type for sperm motility, viability and membrane integrity. Thirty minutes equilibration time had the lowest values (P<0.05) for all the evaluated parameters independent of extender type. Overall, semen extended in Tris-cEY, Bioxcell® and Optixcell® were similarly better when equilibrated at 4 hours (P>0.05). Moreover, post-thawed semen which were extended in Optixcell® for 2 hours equilibration showed a better motility compared with the other extenders (P<0.05). In conclusion, two hours equilibration of semen with Optixcell® is sufficient for semen freezing. Four hours equilibration has the best sperm survival, independent of the extender type.

Abstract

The objective of this research was to investigate the potential of snakehead albumin extract (channalbumin) for sorting X and Y sperm of Sumba Ongole (SO) and its characteristic. Semen was collected from three SO bulls using artificial vagina and the freeze dried channalbumin was extracted from snakehead fish. Channalbumin column was made with different concentration ratio of top and bottom fraction: 2%:4%; 3%:5%; 4%:6% respectively and BSA 5%:10% as control. Semen was put in top fraction then incubated for 30 min at room temperature then each fraction was centrifuged at 1800 rpm for 10 minutes. The pellet was evaluated for motility, abnormality, viability, membrane integrity and head sperm morphometric. The results showed that the channalbumin capable to maintain sperm motility in the top fraction better than the bottom fraction. Sperm viability and membrane integrity in control group were significantly higher (P<0.05) than all channalbumin treatment. BSA 5%:10% has highest proportion of X and Y sperm (69%:76.77%) compared with 2%:4% (42.33%:79.13%), 3%:5% (55.97%:75.73%) and 4%:6% of channalbumin (62.77%:68%). It’s concluded that channalbumin 4%: 6% was effective for separation of XY sperm with higher proportion.

Abstract

The objective of this study was to compare the quality of frozen semen from Toraya buffalo with different lecithins from different commercial diluents. Fresh semen from two 6-9 years old Toraya buffaloes were collected once a week in the morning through an artificial vagina. Fresh semen was examined macroscopically and microscopically. Semen with more than 70% sperm motility was divided into three tubes, each diluted with Andromed (soy lecithin), Steridyl (egg yolk lecithin), and Bovifree (synthetic lecithin) diluents at a concentration of 100 x106 mL-1 motile sperm. The diluted sperm were placed in 0.25 mL straws and allowed to equilibrate for 4 hours. The sperm was frozen above liquid nitrogen favour for 15 minutes and stored in liquid nitrogen containers for further evaluation. The quality of frozen semen was assessed 24 hours after freezing. The parameters tested were motility, viability, abnormalities, plasma membrane integrity, and sperm recovery rate (RR). The results showed no significant differences in the values of motility, viability, abnormalities, plasma membrane integrity, and sperm RR in the three diluents used. Sperm motility values ranged from 45.93% to 47.09% after freezing. Sperm viability ranged from 56.21% to 60.27%. The values of membrane integrity of the three diluents used ranged from 57.73% to 61.36%. The values of sperm abnormalities after freezing and thawing ranged from 2.74% to 3.18%. In conclusion, three commercial diluents containing animal, vegetable, and synthetic lecithin bases can be used as diluents for freezing Toraya buffalo semen with similar results.

Abstract

The quality of fresh and frozen semen as well as the potential to produce frozen semen, vary considerably between individual animals and cattle breeds. This study aimed to analyze the quality of fresh and frozen semen in Aceh cattle and calculate the potential production of frozen semen in Aceh bulls over 1 year. This study used primary data on the quality of fresh and frozen semen from five Aceh cattle and obtained secondary data from artificial insemination centers in Lembang and Singosari in 2022. Semen samples were collected weekly, and the quality of fresh and frozen semen samples were evaluated. The data were analyzed using a one-way analysis of variance at 95% significance level, followed by Tukey’s test. The results revealed differences in semen volume and pH. Based on microscopic characteristics, the semen samples from different bulls exhibited no discernible differences in mass movement, sperm motility, viability, morphology, and plasma membrane integrity, except for sperm concentration and acrosomal integrity. Bull 211605 exhibited the highest sperm concentration. Furthermore, the frozen semen samples from Aceh cattle showed no significant differences in viability, plasma membrane integrity, morphology, and acrosomal integrity. The total sperm motility of bull 211710 was higher than that of the other bulls. The highest progressive motility was observed in bulls 211710 and 211605, and the highest intact DNA was detected in bulls 211710 and 211605. The results of this study demonstrated that the quality of fresh and frozen semen in Aceh cattle is distinct, and the potential frozen semen production of Aceh cattle is estimated to range from 3,382 ± 1810 to 11,399 ± 2658 straws/year.

Abstract

This study was aimed to evaluate the effect of different IgY concentrations on the production performance and quality of semen sentul cocks with different total IgY in serum. This research used 20 sentul cocks with 4 month age consisted of 10 cocks with high IgY concentration and 10 cocks with low IgY concentration. Variabels observed included production performance (feed consumption, weight gain of feed conversion, final body weight, mortality)  and quality semen (macroscopic and microscopic). Study was designed using completely randomized design (RAL) with 2 treatments. All treatments were repeated 10 times. The results started that IgY concentrations treatment did not affect DMA production performance and quality of semen, but signicantly affecting (p<0.05) mass movement of spermatozoa.

Abstract

The aim ofthis study was to examine the preservability of local chicken semen in extender added with various level extra virgin olive oil (EVOO).Semen was collected from three roosters using massage method. Immediately after collection the semen evaluated macro and microscopically. Semen demonstrated   motility, individually divided into five tubes. Each of them diluted with ringer fructose that supplemented with 0%, 4%, 6%, 8% or 10% EVOO. Semen was stored in  refrigerator (5 oC) for sixty hours and evaluated every twelve hours for spermatozoa motility and viability. Results revealed that supplementing the diluent of roosters semen with EVOO and then storing it for different storage periods (, , ,  or  hours) resulted in significant (p .) improvement in spermatooa motility and viabilityin comparison with the control group.Moreover, used of 8% EVOO surpassed other treatments afterin vitro storage for up to 60 hoursand resulted fertilizing ability up to 73.81%.This research conclude that 8% EVOO can be used as antioxidant source for roosters semen during preservation.Keywords : extender, local chicken, extract virgin olive oil, sperma
