Abstract

Antioxidant is molecules that can inhibit the activity of free radical. White guava (Psidium guajava Linn) is one of many plants that have potential as antioxidant. This research was aimed to identify the flavonoid compounds and to determine IC50 values in the white guava leaf extract. Extraction was done by maceration method. The separation and purification was done by TLC and coloumn chromatography method. Flavonoid compound was identified using FTIR and UV-Vis spectrofotometry and antioxidant activity was tested with DPPH method. A 1000 grams of white guava leaf powder was extracted by n-hexane produced 103,40 grams of concentrated n-hexane extract and extraction procces by ethanol 70 % produced 128,49 grams of concetrated etanol extract. Eighty grams of ethanol extract partition produced 1.01 grams of n-hexane extract, 1.95 grams of chloroform extract, and 3.77 grams of n-butanol extract. The results of phytochemical test indicated that n-butanol extract positively contained flavonoids. The results of antioxidant activity towards DPPH from n-butanol extract showed IC50 of 37,14 ppm. The separation and purification n-butanol extract with n-hexane : ethyl acetate : n-butanol (8:2:1) as the eluent resulted 5 fractions (A, B, C, D, E), with C fraction positively contained flavonoids. FTIR analysis showed C isolates contained functional groups of CH aliphatic, CH aromatic, C = C aromatic, C = O, OH, and C - O. C fraction was analyzed with UV-Vis showing 2 peaks at ? 347.30 nm (band I) and ? 278.50 nm (band II) suggesting the presence of flavones group with possibility of OH groups attached to C-3, C-2’ atoms, ortho-dihydroxy groups attached to C-5', C- 6; C-4', C-5', and O-glycoside group at C-7 atom.

Abstract

The aim of this research was to find specific activity of bromelain enzyme extracted from crude extract of pineapple fruit (Queen variety) by salting out method using three different salts, which were (NH4)2SO4, NaCl, and Kusamba traditional salt. Crude extract of bromelain enzyme was obtained by precipitation method using salt with different levels of saturation. This study included determination of total protein concentration by using Biuret method and protease activity by using Anson method to get the specific activities of the crude extract. The results showed that the highest specific activities of bromelain enzymes precipitated with the three salts (NH4)2SO4, NaCl and Kusamba traditional salt with the ratio of 1: 0.94: 0.56 were 0.2887 U/mg (at 20% saturation) ; 0.2705 U/mg (at 40% saturation) and 0.1623 U/mg (at 80% saturation). Fractination of enzyme with the NaCl and Kusamba traditional salt gave smaller specific activity value than (NH4)2SO4. This results may caused of NaCl had smaller ionic strength, the presence of other ions in Kusamba traditional salt and the different solubility of bromelain enzyme in different salts. However, due to the ratio of NaCl approached (NH4)2SO4 and the availability of Kusamba traditional salt they could be used as alternative salts to replace (NH4)2SO4 in fractination bromelain enzyme from crude extract of pineapple fruit

Abstract

The research of purification process of crude glycerol using phosphoric acid and white cement as adsorbent has been carried out. The aim of this research was to study the effect of phosphoric acid concentration and weight of white cement used as adsorbent in the purification process to the purity of glycerol. The research was done by the addition phosphoric acid in crude glycerol followed by adsorption with white cement. Glycerol quality parameters were determined by assesment of glycerol density, viscosity, and by comparing its GC-MS spectra to GC-MS spectra of standard glycerol. The result showed that purification process of glycerol using 5 % phosphoric acid and 5 grams of white cement gave the highest level of glycerol which was 84.20%. The density of purified glycerol obtained in this research was 1.2595 ± 3.1622 x 10-5 g/mL, the viscosity was 1.4372 ± 3.8333 x 10-3 Pa.s, and the GC_MS spectra purified glycerol showed similar fragmentation pattern and retention time with the glycerol standard.

Abstract

DNA content in total honey microorganism can be used to study DNA fingerprint of honey. Metagenomic is a new approach to analyse complex genomes from unculturable total microorganism. The aim of this research was to get DNA metagenomic isolated directly from honey samples enriched with microorganisms growing media LB (Luria-Bertani) and without enrichment of microorganisms growing media LB in order to study DNA fingerprint of honey for the next research. The study was started with incubation of the LB enriched honey samples and honey without LB samples at variation time of 0, 24, 48, and 72 hours. Following the incubation process, the DNA metagenomic form both LB enriched and unenriched honey samples were isolated with direct cell lysis method. The DNA obtained was analyzed by spectrophotometer UV-Vis and agarose gel electrophoresis.  The result showed that DNA metagenomic can be isolated from LB enriched honey and without LB enriched honey samples. Analysis using the agarose gel electrophoresis indicated that the metagenomic DNA obtained from both honey samples was still contaminated with RNA. The purity ratio of metagenomic DNA from both honey samples analysed using UV-Vis Spectrophotometer at ? 260/280 nm was between 1.2 to 1.6. This ratio suggested that the metagenomic DNA isolated from both samples were contaminated with protein and fenol.

Abstract

Studies on synthesis of silica gel from rice hull and waste ofcombustion ash with prepitatian method have been. The studies was with the determination of the optimum temperature incineration of rice husk, the determination of the optimum concentration of NaOH in the manufacture of Sodium Silicate solution from rice hush and brick ash. Synthetic silica gel was obtained by boiling the ash with NaOH solution to create sodium silicate. This solution was neutralized with sulfuric acid  and silica gel was produced. After washing and drying the silica gel was analyzed by spectrophotometer IR and X-ray Diffractometer. The result showed that the optimum incineration temperature of rice husk was 700oC, the optimum concentration   of NaOH was 3M. Based on the characterization by IR, silica gel has been successfully created, marked by the absorption indicating and silianol of functional groups –OH at 3456,44-3487,30 cm-1and Si-O-Si or siloxane at 1103,28-1087,85 cm-1and X-RD analysis showed that silica gel formed was amorphous form.
