Abstract

Forensic is a multi-discipline science that is used to obtain evidence of various criminal cases, such as rape. DNA analysis on sperm specimen is needed to identify the rapist. However, the success of this analysis depends on the DNA isolation method used. Several methods of DNA isolation from human sperm have been developed, but no method has been proven effective for the forensic analysis need. This study aimed to determine the effective sperm DNA isolation method for forensic analysis. In this study, the DNA of sperm specimens was isolated using three methods: Boiling Water, modified TRIzol, and Chelex-100. The DNA isolation result was visualized using agarose gel electrophoresis method. The concentration and purity of isolated DNA were measured using a Nanodrop by comparing the absorbance of DNA at λ 260 nm and protein at λ 280 nm. The effectiveness of the sperm DNA isolation method was determined based on the concentration and purity of DNA, the specimen volume, the implementation time, and the costs involved. The result showed that the successful methods for isolating sperm DNA were TRIzol and Chelex-100. The quantity of DNA isolated using the modified TRIzol method was 1,5 times higher than Chelex-100 but equired 120 times more specimen volume than Chelex-100. From 25 µl sperm specimens, the concentration of DNA isolated using the Chelex-100 method was 612.6 ng/µl with a purity of about 1.7. Therefore, it can be concluded that the Chelex-100 is the most effective method for isolating sperm DNA for forensic analysis.

Abstract

Peningkatan angka kejadian Covid-19 terjadi disebabkan akibat beban populasi yang cukup tinggi di wilayah Jawa Barat. Cluster-cluster baru terdiri dari kasus di pemukiman penduduk, di mana rata-rata saat dilakukan penelusuran bisa menginfeksi anggota keluarga lainnya. Peningkatan cluster di pemukiman penduduk cukup dikhawatirkan karena beberapa penduduk sudah kurang menerapkan protokol kesehatan untuk Covid-19. Penduduk di daerah lingkungan Stikes Jenderal Achmad Yani pun termasuk kurang menerapkan protokol kesehatan, seperti sering terlihat kerumuman warga dan lalai menggunakan masker, sehingga dikhawatirkan menjadi pembawa penyakit untuk keluarga dan menimbulkan cluster baru. Lingkungan Stikes Jenderal Achmad Yani dekat dengan Rumah Sakit Pusat Rujukan Covid-19 di Jawa Barat. Program Studi Teknologi Laboratorium Medis (TLM) D-4 sebagai salah satu program studi yang terlibat dalam diagnosis penyakit, berinisiatif untuk melakukan pemeriksaan skrining Covid-19. Kegiatan ini dilakukan dengan memeriksa antibodi SARS-COV-2 dengan metode imunokromatografi terhadap 50 warga di sekitar kampus. Semua kegiatan pemeriksaan dilakukan sesuai dengan protokol yang diberlakukan oleh Kementrian Kesehatan Republik Indonesia dan World Health Organization. Hasil pemeriksaan menunjukkan 98% sampel non reaktif dan 2% reaktif. Sampel dengan hasil reaktif ditindaklanjuti dengan pemeriksaan lanjut menggunakan teknik Polymerase Chain Reaction. Selain pemeriksaan, sampel diberikan edukasi untuk tetap mengikuti protokol kesehatan dan menjaga lingkungan dari penularan Covid-19.

Abstract

The microalgae Chaetoceros calcitrans has potential as a natural antibacterial but is rarely applied to pathogens that cause skin infections such as Staphylococcus aureus and S. epidermidis. The purpose of this study was to determine the optimum concentration of C. calcitrans extract to inhibit the growth of S. aureus and S. epidermidis. The antibacterial activity of C. calcitrans was tested by the Kirby Bauer diffusion method. The results showed that C. calcitrans extract dissolved in DMSO with the concentrations of 5, 10, 15, and 25 mg mL-1 could produce inhibition zones on S. aureus and S. epidermidis. The average diameter of the largest inhibition zone resulted in the concentration of 25 mg mL-1, namely 10.1 ± 0.5 mm in S. aureus and 9.3 ± 0.5 mm in S. epidermidis. It can be concluded that the extract of C. calcitrans has antibacterial activity against bacteria that cause skin infections S. aureus and S. epidermidis with the optimum concentration of 25 mg mL-1.
