Abstract

The aim of this study was to identify the genetic polymorphisms of calpastatin (CAST), calpain 3 (CAPN3) and myostatin (MSTN) on local sheep at Jonggol Animal Science Teaching and Research Unit (JASTRU). A total number of 294 blood samples were collected from JASTRU. The identification of polymorhism in CAST and CAPN3 genes performed by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) while MSTN gene by using PCR-SSCP methods. The results showed that CAST|MspI, CAST|NcoI and CAPN3|MaeII loci were polymorphic, whereas The MSTN locus was monomorphic for G (1.0). The frequency of allele M (0.87) on the locus (CAST|MspI) higher than the N allele (0.13). At locus CAST|NcoI, the frequency of allele M (0.96) higher than the N allele (0.04). At the CAPN3|MaeII, allele G (0.85) and allele T (0.15). Locus CAST|NcoI has higher observed heterozygosity (Ho = 0.92) compared to CAPN3|MaeII and CAST|MspI (Ho = 0.74-0.77), however has lower compared to CAPN3|MaeII and CAST|MspI in expected of heterozygosity (He = 0.08 vs 0.23-0.26) and in index fixation (Fis = -0.04 vs 0.03-0.12).

Abstract

ABSTRAKKinesin Family 12 (KIF12) merupakan gen yang terlibat dalam mediasi kaskade antioksidan dalam sel beta sebagai sebuah target intraselular dari kelebihan asupan lemak atau lipotoksit. Penelitian ini bertujuan untuk mengidentifikasi karakterisasai gen KIF12 melalui identifikasi keragaman dan asosiasi gen KIF12 terhadap komposisi asam lemak pada domba. Sampel domba yang digunakan sebanyak 35 ekor yang terdiri dari Domba ekor Gemuk (DEG) dan Domba Ekor Tipis (DET) masing-masing 20 dan 15 ekor. Titik mutasi gen KIF12 berada pada posisi g.9617965 C>T.  Identifikasi keragaman gen KIF12 dianalisis menggunakan Polymerase Chain Reaction - Restriction Fragment Length Polymorphism (PCR-RFLP) dengan menggunakan enzim BfaI. Hasil keragaman gen KIF12 bersifat polimorfik dengan ditemukan tiga genotipe yaitu CC, CT, dan TT. Hasil Uji chi square menunjukkan bahwa lokus KIF12 berada pada keseimbangan Hardy-Weinberg (HWE) pada pada DET, hal berbeda ditunjukakan pada DEG. Titik mutasi gen KIF12 berasosiasi secara signifikan  (P <0.05) terhadap asam lemak jenuh (SFA)  asam laurik (C12:0), asam miristik (C14: 0) and asam heptadenoik (C17: 0) juga asam lemak jenuh tunggal (MUFA), asam miristoleinik (C14: 1) and asam oleik (C18: 1n9c). Gen KIF12 memiliki potensi untuk digunakan sebagai penanda seleksi terhadap komposisi asam lemak.Kata kunci:  domba, asam lemak, KIF12, PCR RFLPABSTRACTKinesin Family 12 (KIF12) is one of gene which involved in mediates an antioxidant cascade in beta cells as an intracellular target of excess fat intake or lipotoxicitys. This study was aimed to identify the polymorphism and association of KIF12 gene as candidate gene for fatty acid composition in Indonesian sheep including fat-tailed sheep and thin-tailed sheep. The sample of sheeps used 35 heads consist fat-tailed sheep and thin-tailed sheep as many as each 20 and 15 samples. Identification of polymorphism of KIF12|BfaI gene were analyzed using Polymerase Chain Reaction - Restriction Fragment Length Polymorphism (PCR-RFLP). The results of polymorphism of KIF12 gene were polymorphic for three kind of genotypes of CC, CT, and TT. The chi square revealed that the locus of KIF12 was deviated in Hardy-Weinberg equilibrium (HWE) in fat-tailed sheep, but was in HWE in thin-tailed sheep. A SNP of KIF12 was associated (P <0.05) with saturated fatty acids (SFA) including lauric acid (C12:0), myristic acid (C14: 0) and heptadecanoic acid (C17: 0) as well as monounsaturated fatty acids (MUFA), namely miristoleinic acid (C14: 1) and oleic acid (C18: 1n9c).The KIF12 gene was the potential to be used as a marker of selection for fatty acid compositions.   Keywords : sheep, fatty acids, KIF12, PCR-RFLP

Abstract

The aim of this study was to evaluate the effect of diet containing mung bean sprout waste (MBSW) and feeding times on the hematological profiles and metabolite status of garut lambs. Experimental design used in this study was a factorial randomized block design with two factors of diet and two feeding times. Two factors of diets were 60% concentrate 1+40% natural grass and 60% concentrate 2+40% MBSW. Feeding times were in the morning (6:00-7:00 am) or the evening (5:00-6:00 pm). The animals were reared in individual cages and fed with 5% DM/kg body weight/day. The hematological profiles of the goat were examined and analyzed with Anova. The results showed the treatment of rations significant effect on blood glucose, whereas feeding time very significant to glucose, erythrocytes and hemoglobin, and the interaction of both treatments significantly affect hemoglobin, hematocrit, and glucose level. Meanwhile,leukocytes, neutrophils, lymphocytes, neutrophils/lymphocyte ratio (N/L), urea-N, and blood cholesterol were not affected by the treatments and their interactions. But overall the number of erythrocytes 8.72-12.78x106/mm3, hemoglobin 7.6-10.02 g/dL, leukocytes 8.94-12.27x103/mm3, neutrophils 33.00-52.20%, lymphocytes 38.80-52.40%, N/L 0.78-1.66, glucose 41.94-54.24 mg/dL, urea-N 29.91-35.87 mg/dL, and blood cholesterol 24.57-30.28 mg/dL. These results suggest that diets containing 40% MBSW together with a morning or an evening feeding time did not cause disturbances in haematological profile and blood metabolite status of garut lamb. ABSTRAK Penelitian ini bertujuan untuk mengevaluasi efek ransum mengandung hijauan limbah tauge dan waktu pemberian yang berbeda terhadap profil hematologi dan status metabolit darah domba garut. Materi penelitian berupa 20 ekor domba garut berumur 6-7 bulan (bobot badan 15,42 ± 2,42 kg) dipelihara selama tiga bulan dan dikandangkan secara individu serta diberi pakan sebanyak 5% bahan kering/kg berat badan/hari. Penelitian ini menggunakan rancangan acak kelompok pola faktorial dengan dua perlakuan yaitu jenis ransum dan waktu pemberian pakan. Dua jenis ransum yang diformulasikan dengan basis bahan kering (BK) adalah Ransum 1 (60% konsentrat 1 + 40% rumput lapang) dan Ransum 2 (60% konsentrat 2 + 40% limbah tauge). Waktu pemberian pakan terdiri dari pemberian pakan pada pagi hari (06.00-07.00) dan sore hari (17.00-18.00). Pengambilan sampel darah dilakukan pada akhir bulan kedua setelah masa pemeliharaan. Data yang diperoleh dianalisis dengan sidik ragam. Perlakuan jenis ransum berpengaruh nyata terhadap kadar glukosa darah. Waktu pemberian pakan berpengaruh nyata dan sangat nyata terhadap kadar glukosa, eritrosit dan hemoglobin. Interaksi kedua perlakuan tersebut berpengaruh nyata terhadap hemoglobin, hematokrit, dan glukosa darah. Leukosit, neutrofil, limfosit, rasio neutrofil/limfosit (N/L), urea-N, dan kolesterol darah tidak dipengaruhi oleh kedua perlakuan dan interaksinya. Secara keseluruhan jumlah eritrosit 8,72-12,78 x 106/mL, hemoglobin 7,6-10,02 g/dL, leukosit 8,94-12,27 x 103/mL, neutrofil 33,00-52,20%, limfosit 38,80-52,40%, N/L 0,78-1,66, glukosa 41,94-54,24 mg/dL, urea-N 29,91-35,87 mg/dL, dan kolesterol darah 24,57-30,28 mg/dL. Simpulan penelitian adalah ransum yang mengandung 40% limbah tauge dengan waktu pemberian pakan pagi atau sore hari tidak menyebabkan gangguan pada profil hematologis dan status metabolit darah domba garut.

Abstract

The availability of superior seeds garut sheep relatively few and the unavailability of seed supply ofsuperior male garut sheep that is continuously high productive is also a problem. . The success ofcryopreservation is influenced by the diluent being used. This study aimed was to compare the Tris EggYolk containing omega-3 (TEYO) and Tris used commercial egg yolk plus omega-3 fish oil salmon (TEYOS)to the success of cryopreservation of garut sheep semen. Five garut ram aged 1,5 – 2,0 years were used inthis study. Semen was collected using an artificial vagina, then it was evaluated and divided into twotubes, each of them was diluted with TEYO and TEYOS (50x106 sel/straw), before they then packed intostraws, equilibrated (5°C), and frozen with liquid nitrogen vapour (-130oC) for10 minutes and storedthem in the container (-196oC) for further evaluation. The results showed that post thawing values of thesperm motility, viability and intact plasma membrane were 53,75±2,46; 60,75±2,17and 72,58±2,12%respectively, diluted with TEYOS were higher (P<0,05) than those diluted with TEYOS with the spermmotility, viability and intact plasma membrane only at 41,50±1,25; 50,50±1,04 and 57,58±1,03%respectively. This result demonstrated that the recovery rate of spermatozoa in TEYOS diluent washigher (P<0,05) than TEYO. Its concluded that fish salmon oil omega-3 supplemented in TEYOS better incryopreservation of garut ram semen compared to the TEYO diluent.

Abstract

The research objectives were to evaluate and compare of production performances and blood metabolicprofiles of garut and jonggol rams fed with mung bean sprout waste and omega-3. This research used 24rams aged of 1-2 years, consisted of 12 garut rams weighed of 36,43±1,45 kg (CV=13,87%) and 12 jonggolrams weighed of 23,09±0,57kg (CV=8,64%). Research used Completely Randomized factorial Design(CRD 2x3) with 2 main treatments x 3 factorials and four replications. The first factors as the maintreatments were different breeds of garut and jonggol. The second factors as the factorial treatments werethree different feed percentages which consisted of P0 (40% grass+60% concentrate I), P1 (40% mung beansprouts waste+60% concentrate II), and P2 (40% mung bean sprouts waste+60% concentrate II+omega-3).Data were analyzed by analysis of variance (ANOVA) and proceeded by Duncan Range Test. The resultsshowed that P1 had the highest performance on feed efficiency and average daily gain (ADG) parameters.Blood metabolic test showed that blood total protein, cholesterol, and glucose of all rams were normal. Inconclusion feeding rams with mung bean sprout waste as grass substitution have a positive effect on theperformance of the rams production performance.

Abstract

Proses pembelajaran biologi dikondisikan agar lebih menyenangkan melalui penggunaan model pembelajaran yang tepat dan dapat membangkitkan keterampilan berkomunikasi siswa, karena pembelajaran biologi di SMPN 2 Batukliang, umumnya guru lebih mendominasikan proses belajar mengajar dengan satu atau dua metode. Penelitian ini bertujuan untuk mengetahui bagaimana pembelajaran biologi dengan menggunakan keterampilan berkomunikasi dan hasil belajar IPA siswa kelas VIII SMPN 2 Batukliang  Tahun Pelajaran 2017/2018. Jenis penelitian yang digunakan adalah Penelitian Tindakan Kelas (PTK). Pendekatan penelitian dengan menggunakan pendekatan kualitatif  dan kuantitatif. Rancangan penelitian ini terdiri dari dua siklus. Subjek penelitian adalah  siswa kelas VIII SMPN 2 Batukliang Tahun Pelajaran 2017/2018. Hasil penelitian menunjukan bahwa data observasi keterlaksanaan pembelajaran pada siklus I sebesar 64,2% kategori baik dan pada siklus II sebesar 78,5% kategori baik. Sedangkan pada keterampilan berkomunikasi siswa hasil persentase pada siklus I sebesar 57,6% kategori cukup terampil dan pada siklus II meningkat menjadi 76,9% kategori terampil. Sedangkan untuk hasil belajar  siswa nilai rata-rata pada siklus I 62,30 dengan persentase 61,53%, setelah diadakan siklus II nilai rata-ratanya meningkat menjadi 70,19 dengan persentase 88,46%. Berdasarkan hasil penelitian, maka dapat disimpulkan bahwa dengan penggunaan strategi pembelajaran Active Knowledge Sharing dapat meningkatkan keterampilan berkomunikasi dan hasil belajar siswa pada mata pelajaran biologi kelas VIII SMPN 2 Batukliang Tahun Pelajaran 2017/2018

Abstract

Penelitian ini bertujuan untuk menganalisis peran analisis kebutuhan pasar terhadap inovasi dan daya saing kewirausahaan di Kota Tangerang. Pendekatan yang digunakan adalah kualitatif dengan metode deskriptif, di mana data diperoleh melalui wawancara mendalam, observasi, dan dokumentasi terhadap pelaku usaha mikro, kecil, dan menengah (UMKM) serta wirausahawan di Kota Tangerang. Hasil penelitian menunjukkan bahwa analisis kebutuhan pasar memiliki peran penting dalam mendorong kreativitas dan inovasi, baik dalam pengembangan produk, layanan, maupun strategi pemasaran. Faktor pendukung kreativitas wirausaha meliputi akses informasi digital, dukungan komunitas bisnis, serta program pelatihan dari pemerintah daerah, sedangkan faktor penghambat utamanya adalah keterbatasan modal, rendahnya literasi digital, dan keterampilan manajerial yang masih terbatas. Strategi yang terbukti efektif dalam menciptakan ide kreatif adalah melakukan analisis kebutuhan pasar secara rutin, berkolaborasi dengan sesama wirausaha, serta melakukan eksperimen produk skala kecil sebelum dipasarkan lebih luas. Temuan penelitian ini menegaskan bahwa analisis kebutuhan pasar memberikan dampak signifikan terhadap peningkatan inovasi, diferensiasi produk, kepuasan konsumen, serta daya saing wirausaha di Kota Tangerang. Oleh karena itu, penerapan analisis kebutuhan pasar secara berkelanjutan menjadi kunci bagi wirausahawan untuk menjaga eksistensi dan keberlanjutan usahanya di tengah persaingan bisnis yang semakin ketat. .
