Abstract

Phalaenopsis amabilis (L.) Blume orchid is one of the most popular orchid species in the world. However, this ornamental plant is threatened in its natural habitat, the Meratus Mountains of South Kalimantan, Indonesia. This study determines and analyzes the genetic relationship among several morphological characteristics of P. amabilis from this region combined with molecular (RAPD) markers. A total of ten orchid samples comprised of nine moth orchids (P. amabilis) and one species (P. cornu-cervi) as an outgroup, as well as ten RAPD primers were used in this study. Based on the morphological markers, the moth orchids have a moderate level of genetic diversity, indicated by Shannon's index value of 0.5. In contrast to molecular markers, this germplasm shows high genetic polymorphism, shown by the polymorphism degree of 100% for all primers used. The cluster analysis shows that this germplasm can be divided into two clusters for morphological and five for molecular markers. Following these markers, the grouping of moth orchids was nearly corresponding to their origin. Thus, this information could be useful as a reference for orchid conservation and breeding programs in the future.

Abstract

Critical thinking is one of the fundamental competencies needed by humans in the 21st century to produce quality human resources which can be seen in the implementation of the Tri Dharma Higher Education activities, it is necessary to develop learning resources that can direct students to improve and bring up knowledge, attitudes and critical thinking skills. One of the learning resources that can train students' critical thinking skills is a learning resource in the form of popular scientific books based on local potential which is presented and described in simple sentences, looks attractive and easy to understand and in accordance with the characteristics of critical thinking skills to facilitate students in the learning process. This study aims to describe the practicality of the popular scientific book Cyperaceae in the Mangrove area of Sungai Bakau Village on students' critical thinking skills. Data on the practicality of BIP were obtained from individual tests to determine student readability using a readability questionnaire, small test and field test to determine the practicality of expectations and actual practicality using student response questionnaires and observations on the implementation of BIP use using an implementation questionnaire. Research respondents are students who have passed the Higher Plant Botany course with research design development through formative evaluation from Tessmer.Abstrak Berpikir kritis merupakan salah satu kompetensi fundamental yang diperlukan manusia di abad 21 untuk menghasilkan SDM yang berkualitas yang terlihat dalam pelaksanaan kegiatan Tri Dharma Perguruan Tinggi maka perlu adanya pengembangan sumber belajar yang dapat mengarahkan mahasiswa untuk meningkatkan dan memunculkan pengetahuan, sikap dan keterampilan berpikir tingkat kritis. Salah satu sumber belajar yang dapat melatihakn keterampilan berpikir kritis mahasiswa adalah sumber belajar berupa buku ilmiah populer berbasis potensi lokal yang disajikan dan dijabarkan dengan kalimat sederhana,tampilan menarik dan mudah dipahami serta sesuai dengan karakteristik keterampilan berpikir kritis untuk memudahkan mahasiswa dalam proses pembelajaran. Penelitian ini bertujuan untuk mendeskripsikan kepraktisan buku ilmihah populer Cyperaceae di daerah Mangrove Desa Sungai Bakau terhadap keterampilan berpikir kritis mahasiswa. Data kepraktisan BIP diperoleh dai uji perorangan untuk mengetahui keterbacaan mahasiswa menggunakan angket keterbacaan, uji kecil dan uji lapangan untuk mengethaui kepraktisan harapan dan kepraktisan aktual menggunakan angket respon mahasiswa dan observasi terhadap keterlaksanan penggunaan BIP menggunakan angket keterlaksanaan. Responden penelitian merupakan mahasiswa yang telah lulus mata kuliah Botani Tumbuhan Tinggi dengan desain penelitian pengembangan melalui evaluasi formatif dari Tessmer.

Abstract

Garcinia genus has a complicated taxonomy due to the high similarities in morphological characters of its members. The phylogenetic analysis on Garcinia species based on rRNA gene sequence intended to find out the evolutionary relationship amongst the species. This research employed 20 sequences of the rRNA gene of Garcinia species selected from the GenBank of the National Center for Biology Information (NCBI).  The sequences were aligned using ClustalW with the MEGA X application. A phylogenetic tree was constructed Maximum Likelihood method approximation and Kimura 2-parameter model. The results of the analysis showed that the cladogram had monophyletic parameter properties and classified into 3 clades. Clade I consisted of G. celebica, G. hombroniana, G. opaca, G. mangostana, G. malaccensis, G. penangiana, G. scortechinii, G. hanburyi, and G. urophylla. Clade II included G. atroviridis, G. bancana, G. forbesii, G. griffithii, G. cowa, G. nigrolineata, G. globulosa, and G. parvifolia. Clade III composed of G. rostrata, G. nervosa, and G. praininiana.  The species of Garcinia considered the most primitive and the closet to their ancestor is G. nervosa.

Abstract

Critical attitude is born from critical thinking skills in the neighborhood (surrounding environment) and the daily life of the individual. Critical thinking skills can be trained in the learning process by using appropriate learning strategies and resources. One of the learning resources that can be used to practice critical thinking skills is a contextual electronic handout (e-handout). This study aims to describe the practicality of the e-handout “Surrounding Wetlands Environment” to practice critical thinking skills. Assessment of the practicality of the e-handout through one-on-one evaluation and small group evaluation. The research subjects were 5 students of Biology Education at Universitas Lambung Mangkurat (one-on-one evaluation) and 5 other students (small group evaluation). The data collection instruments used were readability test questionnaires, implementation test questionnaires, student response questionnaires, and critical thinking skills measurement activity sheets. The results showed that the e-handout "Surrounding Wetlands Environment" is very practical to practice critical thinking skills.
