Abstract

Population growth increase, without support from good economic conditions, requirer population control. One of the program to control population growth is family planning program (KB). The participation of men in family planning programs is very important. Male contraception can utilize natural materials derived from plants. One of the herb is guava leaves (Psidium guajava L.) which contains many active substances that can inhibit the process of spermatogenesis. The purpose of this study was to determine the effect of ethanol extract guava leaves to the quality (concentration, viability, morphology, and motility) of spermatozoa and the expression of Cyclooxygenase-2 (COX-2) in the testes of mice (Mus musculus L.). The research used experimental design with completely randomized sampling method. Four treatment groups, namely P0 using aquadest as a control 0,5 mL, P1 using 15 mg/25g body weight extract orally, P2 using 20 mg/25g body weight extract orally, and P3 using 25 mg/25g body weight extract orally. Data were analyzed using One Way ANOVA followed by Bonferroni Post Hoc test 5%. The results showed that ethanol extract guava leaves with a dose of 15mg/25g body weight had significantly decreased sperm quality and increased expression of COX-2 in the testes of mice. This research indicates that guava leaves have a potential to be used as a herbal contraceptive.

Abstract

ABSTRAKKaryawan Mebel di Pusat Rehabilitasi Kusta Gema Kasih Galang banyak yang tidak menggunakan alat pelindung diri seperti masker, kacamata kerja, sepatu kerja dan sarung tangan saat bekerja. Hanya mengandalkan baju dan celana dari rumah saja padahal lingkungan kerja banyak debu dan serpihan potongan kayu. Mereka merasakan aman dan nyaman karena belum pernah mengalami kecelakaan kerja. Tujuan dari program penyuluhan kesehatan ini adalah untuk mengetahui tingkat pengetahuan karyawan mebel tentang pentingnya keselamatan dan kesehatan saat bekerja dan pentingnya penggunaan alat pelindung diri. Metode yang dilakukan berupa penyuluhan kesehatan kepada 15 orang karyawan Mebel di Pusat Rehabilitasi Kusta Gema Kasih Galang. Evaluasi telah dilakukan dengan memberikan pre test dan post test. Hasil evaluasi sebelum dilakukan penyuluhan tentang keselamatan dan kesehatan kerja (K3) adalah mayoritas tingkat pengetahuan peserta kurang yaitu sebanyak 10 orang (66,6%) belum mengetahui konsep dari Keselamatan dan Kesehatan Kerja (K3) dan belum mengetahui tentang jenis-jenis Alat Pelindung Diri (APD). Setelah dilakukan penyuluhan tentang keselamatan dan kesehatan kerja (K3) menunjukkan adanya peningkatan pengetahuan karyawan sebanyak 13 orang (86,6%) yang memiliki pengetahuan baik dan menggunakan alat pelindung diri saat bekerja. Perbandingan pengetahuan peserta sebelum dan sesudah dilakukan penyuluhan tentang keselamatan dan kesehatan kerja (K3) adalah terjadinya peningkatan pengetahuan peserta sebesar 73,3% untuk kategori baik.             Kata kunci: keselamatan dan kesehatan kerja; pusat rehabilitasi kusta gema kasih; alat pelindung diri ABSTRACTMany Furniture employee in the Gema Kasih Galang Leprosy Rehabilitation Center Furniture don’t use personal protective equipment such as masks, work glasses, work shoes and gloves at work.  Only rely on clothes and pants from home even though the work environment is full of dust and pieces of food.They feel safe and comfortable because they have never had an accident at work. The purpose of this health education program is to find out the level of knowledge of furniture employees about the importance of safety and health at work and the tmportance of using personal protective equipment. The method used is in the form of health education to 15 furniture employees at the Gema Kasih Galang Leprosy Rehabilitation Center. Evaluation has been done by giving pre test and post test. The results of the evaluation before counseling on occupational safety and health (K3) was that the majority of the participants’ knowledge level was lacking as many as10 people (66,6%) did not know the concept of occupational safety and health (K3) and did not know about the types of personal protective equipment (PPE). After counseling on occupational safety and health (K3) showed an increase in employee knowledge as many as 13 people (86.6%) who had good knowledge and used personal protective equipment while working. Comparison of knowledge of participants before and after counseling on occupational safety and health (K3) is an increase in participants’ knowledge of 73.3% for the good category.    Keywords: occupational safety and health; kasih galang leprosy rehabilitation center; personal protective equipment

Abstract

Smoking behavior is still entrenched in Indonesian cause a variety of diseases and even death. Cigarettes have become necessities of life that can not be abandoned in everyday life. The smoking problem in essence has become a national problem, even international level. But most connoisseurs of cigarettes assume that smoking is a personal responsibility for the consequences caused. Scientific researchers revealed that smoking is the leading cause of premature death. Statistical data shows that 90% of deaths due to respiratory problems, 25% of deaths caused by coronary heart disease that is triggered by smoking. In cigarette smoke contains 4000 chemicals, one of which is carbonmonoksida (CO), which can disturb binding of oxygen with hemoglobin (Hb), which transformed into bonds HbCO. This research used a correlational analysis with a quantitative approach and cross-sectional study design. Samples used as many 100 respondents used purposive sampling technique. Samples fill out the questionnaire and measured HbCO levels in the blood HbCO qualitatively. Data were analyzed with univariate, bivariate with Product Moment correlation techniques, multivariate correlation analysis. The results showed that there is a correlation of smoking behavior (number of cigarettes smoked by the respondent, the respondent smoking duration and type of cigarettes consumed by the respondents) against HbCO levels. Smoking behavior (knowledge, attitudes, and actions on how cigarette smoking cigarettes made by respondents) have no correlation with HbCO levels.

Abstract

The family planning program needs to be carried out as an effort to reduce the rate of population growth. In this case the participation of men is very important and need research about natural ingredients that can be used as antifertility drugs. Aloe vera leaf (Aloe vera L.) is one of the plants that have an antifertility effect so that it can be used as a contraceptive for men. The purpose of this study was to determine the effect of ethanol extract  aloe vera leaves of the quantity of spermatozoa and the expression of Cyclooxygenase-2 (COX-2) in the testes of mice (Mus musculus L.). The research used four treatment groups, namely P0 as a control was given aquadest 0,5 mL, P1 was given extract 15 mg/25g body weight by oral, P2 was given extract 20 mg/25g body weight by oral, and P3 was given extract 25 mg/25g body weight by oral given for 4 weeks. The research used completely randomized sampling method and data were analyzed using One Way ANOVA followed by Bonferroni Post Hoc test 5%. Administration of ethanol extract aloe vera leaves resulted in a decreased sperm quantity and increase expression of COX-2 in the testes of mice so that aloe vera leaves had the opportunity as a herbal contraceptive for men.

Abstract

Chronic renal failure (CKD) is a progressive decline in kidney function so that it cannot maintain hemostasis in the body, in order to maintain hemostasis in the body, hemodialysis is carried out in patients with kidney failure, to replace kidney function. Examination of albumin is serum or plasma taken from venous blood. The purpose of this study was to analyze albumin levels in patients with chronic renal failure pre and post hemodialysis at Santa Elisabeth Hospital Medan. This research method is descriptive. The population in this study were 178 patients with a total sample of 64 respondents. The method used to examine albumin levels is Bromocresol Green (BCG). The results showed that pre-hemodialysis albumin levels in patients with chronic renal failure had an average of 3.080. g/dL. Post-hemodialysis albumin levels have an average of 3.683 g/dL. Based on the examination of albumin levels in patients with chronic renal failure pre and post hemodialysis, it was found that albumin levels had increased within normal limits. Thus, patients with chronic kidney failure undergoing hemodialysis check their blood albumin levels to avoid hypoalbumin and routinely consult nutrition.

Abstract

ABSTRACT Chronic Kidney Disease (CKD) is a progressive condition that leads to a decline in renal function and often results in anemia. Hemoglobin E (HbE) is a genetic variant that may exacerbate anemia in CKD patients, potentially complicating clinical outcomes. This study aimed to detect the presence of the HbE gene mutation in CKD patients using the electrophoresis method. This descriptive quantitative research was conducted on 25 CKD patients undergoing hemodialysis at Santa Elisabeth Hospital Medan in 2025. Total sampling was applied, and data collection included hemoglobin level measurement and DNA analysis through agarose gel electrophoresis. The results showed that all participants (100%) were diagnosed with anemia, with hemoglobin levels ranging from 7.3 g/dL to 11.4 g/dL. Electrophoresis revealed the presence of DNA bands in both sample and marker lanes; however, the bands appeared faint and lacked sufficient intensity, limiting clear interpretation. This condition leads to uncertainty in determining the presence or absence of the HbE gene mutation in patients with Chronic Kidney Disease. Based on the results, it is concluded that the percentage of the HbE gene in patients with Chronic Kidney Disease at Santa Elisabeth Hospital Medan cannot be determined Keywords: Hemoglobin, Hemoglobin E (HbE) Gene, Chronic Kidney Disease (CKD), Electrophoresis.  ABSTRAK Gagal Ginjal Kronik (GGK) merupakan kondisi progresif yang menyebabkan penurunan fungsi ginjal dan sering menimbulkan komplikasi anemia. Salah satu faktor genetik yang dapat memperparah anemia pada pasien GGK adalah mutasi gen Hemoglobin E (HbE). Penelitian ini bertujuan untuk mendeteksi keberadaan gen HbE pada pasien GGK dengan metode elektroforesis. Penelitian ini menggunakan desain deskriptif kuantitatif dengan teknik total sampling, melibatkan 25 pasien GGK yang menjalani hemodialisis di Rumah Sakit Santa Elisabeth Medan. Pengumpulan data dilakukan melalui pemeriksaan kadar hemoglobin dan analisis DNA menggunakan elektroforesis gel agarosa. Hasil penelitian menunjukkan seluruh responden (100%) mengalami anemia, dengan kadar hemoglobin berkisar antara 7,3 g/dL hingga 11,4 g/dL. Pemeriksaan elektroforesis menunjukkan adanya pita DNA pada sampel dan marker, namun tampak samar dan kurang intens. Hal ini menyebabkan ketidakpastian dalam menentukan ada atau tidaknya mutasi gen HbE pada pasien Gagal Ginjal Kronik. Dari hasil tersebut, disimpulkan bahwa presentase gen HbE pada pasien gagal Ginjal Kronik di Rumah Sakit Santa Elisabeth Medan tidak dapat ditentukan. Kata Kunci: Hemoglobin, Gen HbE, Gagal Ginjal Kronik, Elektroforesis

Abstract

ABSTRACT Thalassemia is a genetic disease characterized by abnormalities in hemoglobin production. One variant frequently found in Southeast Asia is Hemoglobin E (HbE). HbE screening is important for early detection and prevention of thalassemia. The purpose of this study was to determine the presence of HbE gene mutations in female students in the Medical Laboratory Technology Study Program at the Santa Elisabeth College of Health Sciences, Medan, using electrophoresis. This quantitative descriptive study employed a purposive sampling technique in 27 female students. Hemoglobin levels were measured using Hb strips, followed by DNA isolation, PCR amplification, and analysis using agarose gel electrophoresis. Seventy-four percent of respondents had Hb levels below normal. Electrophoresis results showed unclear DNA bands, making it impossible to confirm the presence of HbE gene mutations. This is suspected to be due to errors in reagent concentration and pipetting technique. The majority of female students had low Hb levels, but electrophoresis results could not confirm the presence of HbE gene mutations. Further research with optimal laboratory techniques and a larger sample size is needed.  Keywords: Thalassemia, Hemoglobin E, Electrophoresis, Genetic Screening  ABSTRAK Thalassemia merupakan penyakit genetik yang ditandai dengan kelainan produksi hemoglobin. Salah satu varian yang sering ditemukan di Asia Tenggara adalah Hemoglobin E (HbE). Skrining HbE penting dilakukan sebagai upaya deteksi dini dan pencegahan thalassemia. Tujuan mengetahui adanya mutasi gen HbE pada mahasiswi Program Studi Teknologi Laboratorium Medik STIKes Santa Elisabeth Medan menggunakan metode elektroforesis. Penelitian ini bersifat deskriptif kuantitatif dengan teknik purposive sampling pada 27 mahasiswi. Pemeriksaan kadar hemoglobin dilakukan menggunakan strip Hb, dilanjutkan isolasi DNA, amplifikasi dengan PCR, dan analisis menggunakan elektroforesis gel agarosa. Sebanyak 70,4% responden memiliki kadar Hb di bawah normal. Hasil elektroforesis menunjukkan pita DNA tidak tampak jelas sehingga tidak dapat dipastikan adanya mutasi gen HbE. Hal ini diduga akibat kesalahan konsentrasi reagen dan teknik pemipetan. Mayoritas mahasiswi memiliki kadar Hb rendah, namun hasil elektroforesis tidak dapat mengonfirmasi keberadaan mutasi gen HbE. Perlu dilakukan penelitian lebih lanjut dengan optimasi teknik laboratorium dan jumlah sampel lebih besar. Kata kunci: Thalassemia, Hemoglobin E, Elektroforesis, Skrining Genetik
