Abstract

The rapid advancement of medical biotechnology plays a crucial role in healthcare practices, particularly in genetic engineering for sickle cell anemia. Currently, there is no definitive cure for sickle cell anemia, but in recent years, gene therapy has been extensively researched as an alternative solution. Gene therapy trials on sickle cell anemia patients have been conducted using CRISPR technology, and its effectiveness is still under investigation. The principle involves manipulating the abnormal hemoglobin gene in erythrocytes, which causes sickle cell disease, by extracting stem cells from the patient’s bone marrow and replacing the faulty gene with a normal one using CRISPR (Clustered Regulatory Interspaced Short Palindromic Repeats). The critical components required are Cas9, an endonuclease restriction enzyme, and gRNA, which guides Cas9 to the target gene for manipulation. Once the target gene is successfully cut by Cas9, DNA polymerase naturally initiates a gene repair mechanism through the HDR (Homologue-directed repair) pathway, a process of repairing damaged DNA sequences in CRISPR. Based on tests conducted on 44 patients, 28 patients showed promising results, with their bone marrow producing functional, normal erythrocytes.

Abstract

Hydroxyapatite is one of important material in orthopedic application because its features like human bone, and it has potential to replace metal and ceramic in implantation. Hydroxyapatite may get from waste which rich calcium like sea shell and snail shell. In this research, the characterization of raw materials of hydroxyapatite from freshwater snailshell (Sulcospira sp.) was investigated. The shell was milled with ball mill in 200 rpm during 15 minute with 4 steps. Between the milling was conducted heat treatment process at temperature 900°C for 4h. The shell powder was then sorted based on its roughness with siever machine. Powder characterization was done with Scanning Electron Microscope (SEM), Energy Dispersive X-ray (EDX) and X-ray Diffraction (XRD). Result of this study show that the minimum size of powder about 67 µm and percentage of calcium in the powder is increased until 55%.

Abstract

The new student registration process at Sanggar Kegiatan Belajar (SKB) Ujung Pandang was previously conducted manually, resulting in several issues such as data processing delays, file accumulation, and lack of transparency in selection. This study aims to develop a web-based registration information system integrated with the Simple Additive Weighting (SAW) algorithm to enhance the efficiency and objectivity of the selection process. The system was developed using the Waterfall model and implemented using PHP and MySQL-based web technology. The implementation results show that the system can automate registration and selection in real-time. The SAW algorithm effectively produces objective participant rankings based on criteria such as exam scores, age, and domicile. Evaluation indicates that the system improves selection speed, result accuracy, and facilitates data management for users. It can be concluded that this system provides significant benefits for both SKB administrators and applicants and is relevant in supporting the digital transformation of non-formal education.

Abstract

Berdiri semenjak 2018 ternyata BUMNag 12 Suku masih masuk dalam kategori rintisan, hal ini disebabkan oleh masih buruknya pengelolaan BUMNag oleh pengurus BUMNag periode 2018-2020. Selama peride tersebut sudah mengalami pergantian pengurus sebanyak empat kali yang mengakibatkan BUMNag tidak bisa menjalankan usahanya sesuai dengan target pemerintahan nagari. Yang menjadi permasalahan selanjutnya adalah selama periode tersebut BUMNag menjalankan usaha dengan tidak mempertimbangkan peluang dan potensi Nagari Salimpauang. Oleh karena itu, kegiatan pengabdian kepada masyarakat ini bertujuan untuk memberikan sumbangsih pemikiran dan ilmu terkait strategi pengembangan BUMNag. Kegiatan pengabdian dilaksanakan di Nagari Salimpauang Kabupaten Tanah Datar. Metode yang digunakan dalam kegiatan pengabdian ini adalah sosialisasi dalam bentuk penyampaian materi terkait tata kelola BUMNag, selain itu juga menggunakan metode Participatory Rural Appraisal (PRA) diskusi dan pendampingan dalam menggali apa yang menjadi potensi nagari hingga dapat dijadikan unit usaha BUMNag. Hasil yang ingin dicapai dari kegiatan ini adalah adanya peningkatan pengetahuan pengurus BUMNag dan aparatur nagari terkait dalam tata kelola BUMNag, adanya kemampuan pemetaan potensi nagari, dan peningkatan kinerja dan produktivitas BUMNag melalui unit usaha yang sesuai dengan potensi nagari.

Abstract

Nagari Salimpaung memiliki rencana untuk mengembangkan potensi wisatanya. Namun sayangnya, beberapa masalah membuat pengembangan pariwisata menjadi stagnan. Permasalahan yang mereka hadapi adalah 1) Belum adanya komitmen bersama antar stakeholders yang ada, 2) Belum adanya pemetaan potensi dan permasalahan dalam pengembangan pariwisata, dan 3) Belum optimalnya kelembagaan/aktor penggerak pengembangan pariwisata. Oleh karena itu, PKM ini bertujuan untuk membantu mitra dalam mengembangkan pariwisata dengan melibatkan pemangku kepentingan (elemen terkait) di nagari. Kegiatan yang telah dilaksanakan adalah: 1) Melakukan pretest untuk mengetahui tingkat pemahaman stakeholders dalam pengembangan wisata nagari, 2) Memberikan materi untuk memperkuat pemahaman stakeholders terhadap pengembangan desa wisata, dan 3) Menggali potensi wisata dengan menggali informasi dari pemangku kepentingan dengan menggunakan Participatory Rural Appraisal. Hasil kegiatan menunjukkan bahwa tingkat pengetahuan dan pemahaman pemangku kepentingan mengenai pengembangan wisata nagari masih rendah. Dengan demikian, para pemangku kepentingan harus meningkatkan pengetahuan dan pemahamannya, terutama dalam mencapai komitmen bersama untuk pengembangan pariwisata di Nagari Salimpaung.

Abstract

Program kegiatan Pengabdian Kemitraan Masyarakat (PKM) ini bertujuan untuk menyelesaikan permasalahan yang ada pada mitra. Mitra kami pada kegiatan PKM ini adalah Masyarakat (Ibu-ibu Kelompok Wanita Tani) di Nagari Salimpauang Kabupaten Tanah Datar, dengan permasalahan yang dihadapi yaitu masih rendahnya kesadaran dalam berwirausaha, serta kurangnaya pengetahuan dan pemahaman dalam merecanakan bisnis yang tepat. Sehingga dari permasalahan yang ada, solusi yang kami tawarkan adalah pemberian sosialisasi dan edukasi terkait perencanaan bisnis (business plan). Metode pendekatan yang akan dilakukan di dalam pelaksanaan pengabdian kemitraan masyarakat ini adalah ceramah, diskusi dan pelatihan penyusunan rencana bisnis sederhana. Berdasarkan hasil kegiatan yang telah dilaksanakan dapat disimpulkan bahwa Ibu-Ibu KWT di Nagari salimpauang mendapatkan pengetahuan terkait bisnis yang tepat untuk dikelola dengan memanfaat kekayaan sumber daya alam yang mereka punya.

Abstract

Cervical cancer occupies the third position in the cause of death from all cancers in Indonesia. PCR amplification examination can be used to detect HPV DNA Genotyping. Before carrying out PCR, DNA extraction is an essential step because it can determine the sensitivity of the examination. Sensitivity of HPV DNA detection Genotyping determined by concentration, purity and dot blot hybridization. The aim of this study was to compare the sensitivity of the selective membrane extraction method with heating lysis for the detection of HPV DNA Genotyping. In this research, a qualitative descriptive research method was used using a sample swab Cervical results previously known were six positive low risk, six positive high risk and six combinations of both at Laboratory X in Jakarta. The sampling technique used is disproportionate stratified random sampling. Work is done in duplicate. From the results of test analysis Chi-Square based on concentration, the value obtained is p=0.5 (>0.05), based on purity, the value obtained is p=0.5 (>0.05) and based on the results of HPV DNA hybridization Genotyping The p value obtained was 0.5 (>0.05), meaning there was no significant difference in sensitivity between the selective membrane extraction method and the heating lysis extraction method for the detection of HPV DNA. Genotyping. In this study both extraction methods can be amplified and used in the detection of HPV DNA Genotyping. Heating lysis extraction can be the main choice for use in HPV DNA detection Genotyping because it is more time and cost efficient.

Abstract

This study aims to determine (1) the effect of self-efficacy on students' interest in continuing their education to college (2) the effect of future orientation on students' interest in continuing their education to college (3) The effect of students' learning attention on their interest in continuing their education to higher education (4) The effect of teachers' attitudes on students' interest in continuing their education to higher education (5) The effect of the school environment on students' interest in continuing their education to higher education. The research was conducted in July 2025. This research is an associative study. The population in this study is students at SMK Negeri 1 Bonjol. The sample consists of 94 people using stratified random sampling. The data analysis techniques used are descriptive and inductive analysis, with the assistance of Eviews 8 and SPSS version 20.0. The results of this study indicate that: 1) self-efficacy has a significant effect on interest in continuing education to higher education, with a coefficient value of 0.396 and a t-value > t-table (4.705 > 1.66235). 2) Future orientation significantly influences students' interest in continuing their education to higher education, with a coefficient value of 0.221 and a calculated t-value > table t-value (2.194 > 1.66235). 3) Student learning attention significantly influences students' interest in continuing their education to higher education, with a coefficient value of 0.212 and a t-value > t-table (2.076 > 1.66235). 4) Teacher Attitude (X4) has a significant effect on students' interest in continuing their education to higher education, with a coefficient value of -0.324 and a calculated t-value > table t-value (-3.526 > 1.66235). 5) School environment significantly influences students' interest in continuing their education to higher education, as indicated by a coefficient value of 0.388 and a t-value > t-table (4.202 > 1.66235). 6) Self-efficacy, future orientation, student learning attention, teacher attitude, and school environment collectively significantly influence students' interest in continuing their education to higher education, as evidenced by the calculated F value of 16.322 > F table 2.36 and a significance level of 0.000 < 0.05.
