Abstract

The aim of this research is to isolate, characterize, and identify bacteria on infected African catfish in Banyumas Regency. The researcher used survey method and purposive random sampling technique. Samples were infected African catfish. They were taken from three areas that had potential cultivation for African catfish in Banyumas, namely Rempoah, Kebanggan, and Singosari. In each area, the researcher chose two African catfish cultivating ponds randomly and each pond was taken randomly 3 fishes. Symptoms of the disease in African catfish sample were carefully observed, both externally and internally with surgically. External observation covered wounds and bleeding in some parts of the body, sores or ulcers on the body, protruding eyes, while the internal observation included kidney. Isolation of bacteria carried from the skin (scales), body sores, fin, and kidney. Characterization was done by observing the structure of macroscopic, microscopic, and biochemical tests. The results indicated that 130 isolates of bacteria, and identified to be genus Alcaligenes, Bacillus, Arachnia, Acidomonas, Aeromonas, Amphibacillus, Salmonella, Neisseria, Pseudomonas, Micrococcus, and Clostridium. Abstract

Mucus is one of a non-specific defense mechanism, since this is the first element of aquatic organisms, which contact physically, chemically, or biologically with the environment.  The mucus self defense mechanism investigation was carried out on fresh  water fish tilapia (Oreochromis mosambicus).  Eight (8) types of lectine were used to examine residual carbohydrate-based protein from mucous component based on histological and histochemical observation method.  The review was directed as basic information for detail review about physiology adaptation aspects. The results showed that mucous in goblet cells from palatal, gills primary lamella, ecophagus and skin reacted with WGA (Wheat Germ Aglutinin) lectine.  In another part, mucous from the goblet cells in palatal and esophagus cells reacted with PNA (Peanut Aglutinin).  Based on these results, therefore, it can be concluded that mucous from goblet cells in esophagus contains residual of N-asetil glucosamine and/or similar acid β-galactose and α-N-acetyl galactomine.  Mucous from goblet cell in palatal contains residual of X-acetyl glucosamine and/or sialat acid and galactose.  While mucous in the gills lamella contains carbohydrate residual, namely N-acetyl glucosamine and/or sialat acid

Abstract

Masalah utama dalam budidaya lele dumbo adalah penyakit, terutama yang disebabkan oleh bakteri Aeromonas hydrophila. Penyakit tersebut dikenal dengan penyakit MAS (Motile Aeromonas Septicemia). Salah satu penanggulangan penyakit MAS adalah dengan vaksinasi. Masalah lain yang sering muncul adalah tingginya biaya pakan sedangkan harga jual ikan cenderung stabil, sehingga pembudidaya ikan cenderung merugi. Perlu strategi untuk membuat pakan sendiri dari bahan-bahan yang masih memiliki kualitas gizi yang baik tetapi harganya murah bahkan memanfaatkan limbah, bahan tersebut mudah diperoleh dan tersedia setiap saat. Penelitian ini bertujuan untuk menguji secara lapang penggunaan pakan bervaksin pada lele dumbo di daerah Cilacap. Uji lapang dilakukan di Desa Purwodadi, Kecamatan Nusawungu, Kabupaten Cilacap. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 3 perlakuan dan 5 kali ulangan. Perlakuan terdiri dari P1 : pemberian pakan bervaksin selama 10 hari; P2 : pemberian pakan bervaksin selama 15 hari; dan P3 : kontrol (non vaksin). Pakan diberikan sebanyak 5% /bb/ekor/hari. Penelitian menggunakan kolam terpal plastik dengan ukuran panjang x lebar x tinggi : 60 x 60 x 80 cm. Lele dumbo yang digunakan berumur 2 bulan, berukuran panjang 12-15 cm dengan berat 16-25 g. Ikan dipelihara selama 8 minggu. Parameter yang diamati adalah respons imun berupa titer antibodi, pertambahan berat dan panjang, serta sintasan ikan. Parameter pendukung yang diamati adalah parameter kualitas air meliputi suhu air, pH, dan oksigen terlarut. Data dianalisis menggunakan Analysis of Variance (Anova) dan Duncan Multiple Range Test (DMRT) dengan taraf uji 5%. Abstract

Agricultural tools are important for agrarian communities. However, for some people in Banyumas Regency, Central Java, very few people have businesses in the agricultural tools sector. In fact, this can be a promising business opportunity. Knowledge about making blacksmith agricultural tools is generally obtained from generation to generation. Some blacksmiths in Banyumas generally only produce agricultural tools such as kudi and work machetes, because the need for these tools never stops, is always needed by farmers or workers, and the equipment produced does not require high design, even tends to be monotonous because the main thing is its function as work equipment. That is why Science and Technology for the Community (IbM) was implemented at the Pandai Besi Kirso production house on November 6, 2025, as a method of increasing production. IbM was continued with training in techniques for selecting raw materials, making potions that can improve the quality of iron, and making a variety of new products with high quality both in terms of strength and aesthetics. It was identified that agricultural tool craftsmen generally (1) Have limitations in equipment, (2) Lack expertise in metal blending, (3) Lack of innovative product variations, not only quality but also unique ethnic ones and (4) Low management quality. The implementation of IbM is expected to be able to minimize the problems of agricultural tool craftsmen's businesses, so that the blacksmith craftsmen's community businesses can develop and have a positive impact on the existence of MSMEs in Banyumas Regency.
