Abstract

The use of coconut water, green bean sprouts and onions is chosen as natural growth regulator because it is easily obtained at a fairly affordable price. The aim of the study was to determine the effect of various natural growth regulator different concentrations to the growth of cempedak (Artocarpus champeden Lour.) seedlings.The research was conducted from May to July 2018 in the garden of the Tissue Culture Laboratory Faculty of Agriculture, Mulawarman University. This study used a factorial Completely Randomized Design with five replications. The first factor was the types of growth regulator, namely: coconut water, green bean sprout extract, and onion extract. The second factor was the concentration of the natural growth regulator substances,consisted of five treatment levels, namely: 0, 25, 50, 75 and 100 mL L-1. The parameters observed were plant height, number of leaves, stem diameter, root length, fresh weight (leaves, stems and roots), dry weight (leaves, stems and roots) and leaf area. Data were analyzed by analysis of variance with F-test and continued with the Least Significant Difference (LSD) test with a level of 5%. The results showed that there was no interaction between the type and concentration level of the natural growth regulator to the growth of cempedak seedlings.The growth regulator concentrationalso had no significant effect to all growth parameters observed. However, of the type of natural growth regulators showed a significant in the fresh root weight,stem dry weight, and plant height at harvest time.

Abstract

Sugarcane orchid (Grammatophyllum speciosum Blume) are known as a type of giant orchid, adult orchids can weigh 1 ton, panicle can grow up to 2.5 – 3 meters with a diameter of about 1.5-2 cm. Each panicle has dozens, even reaching one hundred flower buds, each with a diameter of about one 10 cm. Sugarcane orchids are a type of plant that is protected because its existence is increasingly scarce so that it needs in vitro propagation techniques to support its survival. To improve the growth of Grammatophyllum speciosum Blume in vitro shoot tip was growing on MS medium with varying concentration of kinetin (0, 1, 2, 3, and 4 mg L-1) with 5 replications. Shoot tip from in vitro culture around 1 cm lengt was used as explants. Observations carried out for 8 weeks after inoculation.The results showed that shoot explants tip  growth well in MS basic medium. The addition of kinetin to the medium, increases growth rate of culture. Increasing the concentration of kinetin tends to increase the growth. The best concentration for growth induction on plant height, stem diameter,  leaves length and  length of roots was 2 mg.L-1 kinetin, On the other hand 3 mg.L-1 kinetin gave best response on number of leaves and 4  mg.L-1 kinetin was the best concentration on leaves width and number of roots.

Abstract

Eucalypt is known as the fastest growing tree species in the world. Some advantages such as fast growing, hight yielding, short rotation, less desease, lots of purposes and good economic benefit have made Eucalypt as the most important forest tree product for industrial raw material in Indonesia. However, those advantages of Eucalypt including fast growing, high yielding, short rotation and less desease can only be achieved through selecting a good clone. Cutting propagation is considered as one of the main methode to produce good seedling. Sprouts of two clones of Eucalyptus pellita F. Muell (E. 13 and E. 77) were collected from mini hedge. Total of 1080 of each clone was planted in greenhouse. The result showned that Clone E. 77 was better than E. 13 in term of growth of rooting, the increment of height, stem diameter and number of leaves at 60 and 75 days after planting

Abstract

The objectives of the research were: (1) to determine growth and yield response of two tomato varieties upon the application of POC super ACI, and (2) to obtain suitable concentrations of POC super ACI for tomato crop.This research was polybag trials using completely randomized design (CRD) with factorial analysis and 5 replications.  The first factor was the variety, consists of two levels: Ranti and Ratna.  The second factor was the concentration of liquid organic fertilizer, consists of 4 levels, namely 0.00; 0.75; 1.50; and 2.25 ml/l.  Data were analyzed using analysis of variance and if there was a significant difference, then the test used Least Significant Difference (LSD) at 5%.The results revealed that: (1) Treatment of varieties affected significantly on the plant height increment at age of 15, 30, 45, and 60 days after planting and harvesting; days to flowering; number of fruit per crop and fruit fresh weight. Ratna variety produce fruit weight of 239.63 g/polybag while Ranti was 79.08 g/polybag;  (2) The concentration of POC Super ACI affected significanlty on the plant height increment at age of 15, 30, 45, and 60 days after planting and harvesting; number of fruit per crop and fruit fresh weight.  The best fertilizer concentration was 1.50 ml/l water that can produce fruit fresh weight of 194.68 g; and (3) The interaction between varieties and fertilizer concentrations did not affect significantly on all variables observed.

Abstract

Eucalyptus pellita F Muell is a fast-growing tree that has great benefits. The uses of wood are  the raw material  for pulp and paper industry, building construction, and also for charcoal. Propagation can be done by using seeds, cuttings and tissue culture.  Propagation from seed has a weakness for very large offspring genotype variation while the vegetative propagation will produce offspring same with the parent. The Advantages of propagation by tissue culture is able to  produce seedling in large quantities in a short time.The research objective was to determine the growth of Eucalyptus pellita F Muell  seedling in the field  that propagated using three propagation  methods, tissue culture,  cutting,  and seed.The study was conducted in PT Surya Hutani Jaya, Sebulu, Kutai, East Kalimantan.  Seedlings from tissue culture was planted on 13.7 ha,  seedling from cutting was planted on 14.3 ha, and  seedlings from the seeds was planted  on 14.4 ha.  Each seedling was  planted with spacing  3 m x 2.5 m.  All plants had similar treatment in fertilizing, weeding,  pest and disease control. Samples are taken one per cent of the total population of the plant.The results showed that the  plants  from tissue culture seedlings showed the best results on plant height, diameter and volume.

Abstract

Kalimantan whose existence is increasingly extinct so it needs to be preserved by in vitro propagation. This study aims to determine the effect of a combination of the growth regulators, Benzyl Amino Purin (BAP), Kinetin, Naphthalene Acetic Acid (NAA) and banana extract on the growth of sugar cane orchids in vitro. The basic media used were Half Murashige and Skoog (1/2 MS), on the base media were added 30 g.L-1 sugar, 8 g.L-1 seaweed commercial powder,  plus BAP 3 ppm, Kinetin 3 ppm, NAA 0.5 ppm, and ambon banana extract 100 g.L-1, the acidity (pH) of the media is adjusted to around 5.8. The research design used was a Completely Randomized Design (CRD), each treatment was repeated six times. Data were analyzed using variance analysis and continued with DMRT (Duncan Multiple Range Test) tests with a level of 5%.            The results showed that the half media Murashige and Skoog was good for the propagation of sugarcane orchids.  The addition of BAP growth regulators, Kinetin, NAA, and banana extracts showed a good response to growth culture. The addition of BAP 3 ppm + NAA 0.5 ppm showed the best results for the height increased, leaf number increased, leaf length increased, leaf width increased, and the number of shots increased. The addition of Ambon banana extract to media containing growth regulators tended to decrease the growth response, seen in the parameters of plant height increased, leaf number increased, leaf length increased, leaf width increased, the number of shoots increased, the number of roots increased and in root length increased.

Abstract

Eucalyptus pellita  is fast growing species and have many advantages such as: pulp and paper industry raw material, building construction, charcoal, and medicine.  The research was carried out to study plant morphogenesis from shoot explant of E. pellita F. Muell by in vitro technique. The experiment was conducted at Tissue Culture Laboratory, Agriculture Faculty of Mulawarman University from March until June, 2016.Single factor experiment was arranged in Completely Randomized Design with 10 replications.  The factor was BAP concentrations (B) consisted of 1.00 mg BAP L-1 (b1); 2.00 mg BAP L-1 (b2); 3.00 mg BAP L-1 (b3); and 4,00 mg BAP L-1 (b4). The explants was innoculated on MS medium supplemented with BAP according to the treatments. Data analyzed by analysis of variance (anova) and continued by Least Significant Difference  (LSD) test  with level 5%. Results of the experiment showed that the BAP concentrations can promote shoot formation of E. pellita.  Statistical analysis indicated that the  effect of BAP concentrations significantly difference on number of shoots at 8 weeks after innoculation (WAI), shoot length, and number of leaves at  4; 6; and 8 WAI, however it’s no significantly difference on shoot formation time and number of shoots at 4 and 6 WAI.  The highest number of shoots,  shoot length, and number of leaves achieved by 4,00 mg BAP L-1.

Abstract

Limbah ternak kelinci memberikan manfaat yang besar untuk budidaya Kedelai. Penelitian ini bertujuan :  (1) mengetahui interaksi antara pemberian pupuk organik dari limbah ternak kelinci padat dan cair terhadap pertumbuhan dan hasil tanaman kedelai, (2) mengetahui dosis pupuk organik padat kelinci yang terbaik terhadap pertumbuhan dan hasil tanaman kedelai, dan (3) mengetahui konsentrasi pupuk organik cair urin kelinci yang terbaik terhadap pertumbuhan dan hasil tanaman kedelai.Metode penelitian menggunakan rancangan percobaan faktorial 4x4 yang disusun pada Rancangan Acak Lengkap (RAL) dengan ulangan sebanyak empat kali. Faktor pertama adalah pupuk organik padat (P) dengan dosis : 0 Mg.ha-1 (p0), 5 Mg.ha-1 (p1). 15 Mg.ha-1 (p2) dan 25 Mg.ha-1 (p3). Faktor kedua adalah konsentrasi pupuk organik cair urin kelinci (U) dengan konsentrasi 0 mL.L-1air (u0), 200 mL.L-1air (u1), 400 mL.L-1air (u2), dan 600 mL.L-1air (u3). Data dianalisa dengan uji F dan untuk membandingkan antara rata-rata dua perlakuan dilanjutkan dengan Uji Beda Nyata Terkecil (BNT) dengan taraf 5 %.Penelitian ini menunjukkan bahwa interaksi terbaik dari masing-masing perlakuan terdapat pada dosis pupuk organik padat kelinci pada dosis 15 Mg.ha-1 dengan pupuk organik cair pada konsentrasi 600 mL.L-1air (p2u3) dan berpengaruh nyata terhadap variabel tinggi tanaman umur 42 HST, dan jumlah cabang per tanaman. Konsentrasi 600 mL.L-1air (u3) pupuk organik cair urin kelinci memberikan pengaruh terbaik terhadap pertumbuhan dan hasil tanaman kedelai serta berpengaruh nyata pada variabel tinggi tanaman umur 14, 28, dan 42 HST, waktu berbunga, berat kering polong isi, berat kering biji, dan berat 100 butir biji.
