Abstract

The cytotoxic activity experiment of n-hexane extract of kersen (Muntingia calabura Linn.) leaves has been carried out using the Brine Shrimp Lethality Test (BSLT) method. The purpose of this research is to identify the secondary metabolites, extract characterize, and determine the LC50 value of the extract against larvae shrimp Artemia salina Leach. Screening result showed that n-hexane extract of kersen leaves contain triterpenoid. Extract characterization showed the results water content of 2.59±0.18 %w/w, water soluble extractive content of 1.9%±0.19 %w/w, the ethanol soluble extractive content of 17.41±1,87 %w/w and total ash value 0.25% w/w. Extract showed LC50 value is 278,72 ppm were calculated by probit analysis. The LC50 value indicated that the n-hexane extract of kersen leaves potentially has cytotoxic activity.

Abstract

The characterization of simplisia, phytochemical screening, extraction, isolation and identification of chemical constituens  from thallus Turbinaria decurrens Bory have been carried out. The examination of simplisia characteristics gave the water soluble extract the value of 10.59%, ethanol soluble extract valued at 0.93%, total ash valued at 15.64%, the acid insoluble ash value 0.79% and the water content valued at 8.66%. The result of phytochemical screening showed that there was triterpens/steroids present. The extraction process was carried out by percolation and then was separated by liquid vacum column chromatography. Then by preparative thin layer chromatography isolate B1 was obtained. The analysis of isolate B1 by infra red spectrophotometry showed hydroxyl, aliphatic C-H bond, C=O bond, double bond of C=C, C=O bond, C=H bond of CH3 and CH2, were present. Ultra violet spectrophotometry exhibited a maximum absorption at 242 nm and mass spectrometric fragmentation pattern exhibited that the molecular weight of isolate B1 was 394, it was suspected ergosta -4,7,22 –trien -3 one.

Abstract

Abstract. The cytotoxic activity experiment of ethylacetatefraction of kersen (Muntingia calabura) leaves has been carried out using the Brine Shrimp Lethality Test (BSLT) method withf ive concentration variables; they are 10 ppm, 100 ppm, 200 ppm, 500 ppm, and 1000 ppm. The research aims to determine the secondary metabolite; the extract characterization and determine cytotoxic activity of the ethyl acetate fraction of kersen (Muntingia calabura L.) leaves. Extraction was done with maceration method using ethyl acetate solvent to the kersen dried leaves dregs of n-hexane extract and the value of LC50 was determine by using probit analysis. The results of phytochemical screening showed that ethyl acetate fraction of kersen (Muntingia calabura) leaves contained three secondary metabolites, namely tannin, saponin and flavonoid with extract characterization results are the water content of 3.85%3 ± 0.35%, the water soluble extractive of 17.65% ± 0.27%, ethanol soluble extractive of 62.24% ± 0.07%, the total ash value of 0.89%± 0.005%, and acid insoluble ash value of 0.69%± 0.004%. The LC50 value of ethyl acetate fraction of kersen (Muntingia calabura) 84.92 ppm showed that the extract has  cytotoxic activity as medium toxic.
