Abstract

The high population of waterfowl subsequently with the high case fatality of poultry and people in West Java regency caused by HPAI H5N1 can raise possibility that waterfowl was a natural reservoir. This research aimed to prove that waterfowl in West Java served as reservoir of AI virus (primarily H5N1) and also identify the virus pathotype based on cleavage site of amino acid sequence. Cloacal swab sample was obtained from healthy and unvaccinated waterfowl from Sukabumi and Bogor Regency. Cloacal swab was propagated in 9 days old embryonic chicken eggs. Allantoic fluid was harvested at the 4th day of incubation and then tested for hemagglutination, and positive isolate continued with virus sub-typing using PCR method. H5 gene from H5N1 isolate then sequenced using dideoxy termination method. Multiple alignment of nucleotide sequences were analysed using MEGA-3.1 program. Sub-typing using PCR method indicated the existence of 25 strain H5N1, 16 strain HxN1, 4 strain H5Nx and 9 virus ND. Characterization of cleavage site amino acid sequence indicated that all H5N1 sample were pathogenic with sequence QRERRRKKR (23 sample) dan QRESRRKKR (2 sample). Waterfowl was HPAI H5N1 virus reservoir. Asymptomatic infection in waterfowl, but the virus shedding gradually occurred and therefore it became potential source of H5N1 virus infection. Our findings suggest that immediate action is needed to prevent the transmission of highly pathogenic avian influenza viruses from the apparently healthy waterfowl into terrestrial poultry or human. Abstract

Avian influenza viruses (AIV) subtype H5N1 isolated from waterfowls in West Java pose the known characteristic of highly pathogenic strains, with polybasic amino acid sequence of cleavage site QRERRRKKR and QRESRRKKR. This research aimed to analyze the important domains of hemagglutinin (HA) gene of those isolates. Fragment of HA gene was amplified using RT-PCR method with specifically-designed primer pairs and sequenced using dideoxy termination method with ABI automatic sequencer (Applied Biosystems). Multiple alignment of nucleotide and deduced amino acid sequences were analyzed using ClustalW of MEGA-3.1 program. Some of biological domains of HA, i.e. antigenic sites, receptor binding pocket, and glycosylation sites of the isolates were polymorphic. The viruses also pose conserved glutamine (Q) and glisin (G) residues at the known receptor binding site, at the position 222 and 224 respectively. This findings clearly show that all AIV subtype H5N1 isolaed from waterfowl preservesthe α-2,3NeuAcGal avian receptor specificity. Abstract

A study was carried (1) to analyze the phylogenetic relationship of fragment hemaglutinin (HA) geneof avian influenza viruses (AIV) subtype H5N1 isolated from apparently healthy backyard waterfowls inWest Java with representative of animal and human isolates from Indonesia and some countries in Asia;(2) to find out cross-reactivity of those viruses with a standard Indonesian strain. Nucleotide sequences ofHA gene of AIV H5N1 from backyard waterfowls along with other H5N1 isolates of Indonesian and Asianorigin were aligned using with ClustalW of MEGA 3.1 program. Estimation of genetic distance and theconstruction phylogenetic tree were conducted by Neighbor Joining method and calculation of distancematrix using Kimura 2-parameter. Antigenic analysis was conducted using hemagglutination inhibition(HI) test. Result of phylogenetic analysis indicated that all viruses from backyard waterfowls form threedistinct sublineages. One lineage was located in Indonesia cluster and two lineages in Asia cluster. In thephylogenetic analysis, it was concluded that multiple introductions of AIV H5N1 to Indonesia have occurred.Six AI H5N1 viruses from backyard waterfowls (IPB1-RS to IPB6-RS) appeared to be different ancestorsthose isolated previously in Indonesia. Cross-antigenic analysis showed that nine viruses isolates used inthis study were antigenically different to Legok 2003 chicken strain of AIV H5N1. The HI titer of anti-Legok 2003 antibody with all newly isolated viruses is up to 6 log lower then the HI titer using homologstrain.

Abstract

Penelitian bertujuan untuk mendeteksi adanya bakteri Escherichia coli O157 pada air minum kemasan, air minum isi ulang, dan air sumur di Kelurahan Sekaran Gunungpati Semarang. Sampel yang diambil sebanyak 20 sampel yang terdiri atas 4 merk air minum kemasan, 8 sampel air minum isi ulang, dan 8 sampel air sumur. Penelitian diawali dengan tahap isolasi E.coli pada medium Eosin Methylen Blue Agar (EMBA), yang dilanjutkan ke medium Sorbitol MacConkey Agar (SMAC) untuk identifikasi E.coli O157 dilanjutkan uji lateks aglutinasi (OXOID) dan diakhiri dengan uji konfirmasi gen rfbE menggunakan teknik Polymerase Chain Reaction (PCR). Hasil penelitian menunjukkan 8 sampel yang positif E.coli pada medium SMAC menunjukkan positif E.coli O157 (colorless). Uji lateks aglutinasi juga menunjukkan 8 sampel positif E.coli O157 seperti kontrol ATCC 43894. E.coli ATCC 43894 dan 8 sampel E.coli dari berbagai air minum di Kelurahan Sekaran Gunungpati Semarang menunjukkan positif E.coli O157.

Abstract

TK Wiji is a nursery school which was built at Jl. Untung Suropati no. 10  kelurahan Tegalrejo kota Pekalongan based on NPSN 69879295 of 2005 by Yayasan 99. TK Wiji is growing fast due to its education system which is in line to the 2013 national curriculum with the vision of toodler characteristic development. The most fundamental of character education is environment education. Considering to this situation, we have found that the most common litter of the nursery is disposable diaper. This kind of diaper is hardly degraded into its monomer. Colon’s research in 2013 has found a strategy to use this diaper as compost. Based on the problem of disposable diaper wastes in the nursery and also the result of the background research about exploiting the waste as compost, then the environment education for toodler of TK Wiji by using diaper waste as a material for hidroponic cultivation was done. In the beginning, guidances were given to the teacher in 2 phases which are making diaper waste composite into organic fertilizer and making hidrogel for interior plant cultivation by using the gel of the waste. In the next step, the teachers were asked to make improvisation regarding toodler environment education by using the skills. In spite of this, there was still an argument about the time management in the learning process by using diaper waste as the material. Thus an implementation of the skill for toodler extracuricular was stated.

Abstract

Problem Based Learning dengan Mind Mapping merupakan alternatif pembelajaran biologi yang menarik, membantu siswa berpikir kritis dan mampu memecahkan masalah, meningkatkan kreatifitas dan aktivitas siswa. Penelitian ini bertujuan untuk menganalisis pengaruh model Problem Based Learning dengan Mind Mapping terhadap hasil belajar siswa pada materi sistem gerak manusia kelas VIIIdi SMP Negeri 01 Ungaran. Jenis Penelitian ini eksperimen kuasi dengan rancangan penelitian Post-test only design. Sampel penelitian ditentukan secara Purposive Sampling, yaitu kelas VIII A sebagai kelas eksperimen dan kelas VIII B sebagai kelas kontrol. Data utama penelitian adalah hasil belajar kognitif melalui post-test dan lembar diskusi siswa dianalisis menggunakan t-test. Data pendukung berupa data hasil belajar keterlaksanaan pembelajaran, tanggapan siswa dan tanggapan guru. Hasil penelitian menunjukkan bahwa pembelajaran dengan Problem Based Learning dengan Mind Mapping pada kelas eksperimen berbeda terhadap kelas kontrol dengan analisis hasil posttest, t-test menunjukkan thitung 3,474> ttabel 2,042. Rata-rata nilai hasil belajar afektif siswa kelas eksperimen 86 sedangkan pada kelas kontrol 81,43. Berdasarkan hasil penelitian dapat disimpulkan bahwa penerapan model Problem Based Learning dengan Mind Mappingberpengaruh terhadap hasil belajar siswa pada materi sistem gerak pada manusia kelas VIII. Kata kunci:, mind mapping, Problem Based Learning, hasil belajar

Abstract

Kemajuan teknologi sekuensing menyebabkan peningkatan ketersediaan sekuen genom organisme. Ribuan strain dan isolat dari berbagai populasi organisme telah diisolasi serta diketahui sekuen genomnya. Informasi genetik populasi organisme dapat dimanfaatkan sebagai marka molekuler. Marka Single Nucleotide Polymorphism (SNP) terdapat dalam jumlah banyak namun tidak seluruhnya informatif. Metode seleksi yang telah ada belum efektif menyeleksi SNP informatif, oleh karena itu perlu dilakukan pengembangan metode seleksi SNP yang efektif. Metode seleksi SNP dikembangkan menggunakan metode statistik dengan FST sebagai filter (penyaring) utamanya dan digabungkan dengan Linkage Disequilibrium (LD). Struktur populasi dari SNP diketahui menggunakan Principal Component Analysis (PCA), Principal Coordinate Analysis (PCoA), pairwise FST, dan neighbor-joining population tree. Kriteria SNP informatif diketahui dengan menghitung FST dan Minor Allele Frequency (MAF). Metode statistik diuji efektivitasnya dalam menyeleksi SNP informatif menggunakan simulasi data genetik populasi. Hasil penelitian menunjukkan pengembangan metode statistik dengan menggunakan FST sebagai penyaring utama efektif dalam menyeleksi SNP informatif. Kriteria SNP informatif adalah SNP dengan MAF 0,2-0,4 serta FST 0,1-0,4 dan 0,8-1,0.

Abstract

The Covid-19 pandemic that has occurred since the end of 2019 until now, requires people to reduce activities outside the home, and increase activities at home. This makes people's productivity levels decrease. One of the efforts to reduce the activity of residents gathering and doing activities outside the home is to provide alternative activities at home. This community service activity is intended to provide education and skills for hydroponic strawberry cultivation as an alternative activity at home that is healthy for humans and the environment around the residents of Larangan Kulon Village, Mojotengah District, Wonosobo Regency, Central Java. To achieve this goal, several method steps were used, namely observation, lectures/counseling,  demonstrations, and direct practice of the application of hydroponic strawberry plants. The service activity succeeded in transferring the knowledge and skills of hydroponic strawberry cultivation to the participants. Participants' understanding of concepts, tools and materials, cultivation and care methods and types of hydroponics is very good, as evidenced by 78.13% of participants getting a score of ≥80. Hydroponic cultivation of strawberry plants can be practiced by participants and can be used as an alternative to productive activities at home

Abstract

Kampung jamu di kelurahan Wonolopo kecamatan Mijen Semarang adalah sentra produsen jamu Gendong. Wilayah pemasarannya masih di Semarang karena produk tidak awet, sehingga berpotensi menurunkan daya saing dengan produk pabrikan. Tujuan kegiatan pengabdian kepada masyarakat (PPM) ini adalah melatih keterampilan mitra membuat jamu serbuk yang berkualitas, mengemasnya dan dapat memasarkan produknya secara online sehingga wilayah pemasarannya lebih luas Metode kegiatan PPM adalah pelatihan membuat jamu serbuk dan pengemasannya serta pelatihan pembuatan branding dan berjualan online. Pelatihan dilaksanakan pada 15 Juli dan 3 September 2022, kepada 15 orang anggota Paguyuban produsen jamu gendong “Sumber Husodo”. Hasil pelatihan adalah peserta dapat mempraktekkan membuat jamu serbuk dan mengemasnya. Peserta pelatihan juga dapat membuat branding produk jamunya. Peserta juga dapat mempraktekkan penggunaan aplikasi berjualan online, menggunakan facebook, instagram dan tiktok. Simpulan dari kegiatan PPM ini adalah mitra memiliki bekal untuk melakukan diversifikasi produk dan memperluas wilayah pemasarannya, sehingga diharapkan dapat bertahan dan bersaing dengan produk jamu pabrikan.
