Abstract

The fibroin extraction process was done base on temperature and time parameters without knowing exactly the rendement been maximum or not. Optimum rendement data are critical to the business feasibility for scaling up to the industrial level. The objective of this research is to obtain the optimum extraction process (degumming) to get the highest fibroin rendement. The method in this research was response surface method (RSM) where NaOH concentration, process temperature and time consumption were chosen to be independent variables. These variables were formulated in the central composite design (CCD) research method at RSM Program with the center point is NaOH 0.1 N, 105 °C for 30 minutes. The weight of fibroin yield (%) being use as a response condition (dependent variable) to get the optimum result of fibroin rendement. The optimum degumming process condition of Bombyx mori are achieved where the NaOH 0.018 N, 110.53 °C temperature degree for 55.51 minutes for a batch time process with the rendement recorded was 71.11 ± 0.98 %. As for “Attacus atlas” the optimum rendement was 83.06 ± 1.50 % achieved at 0.12 N NaOH concentration, process temperature was 79 oC and 42.65 minutes for twice degumming. The best selected fribroin degumming method will lead to the optimum yield achievement. ABSTRAKEkstraksi fibroin dilakukan dengan perlakuan suhu dan waktu tanpa diketahui maksimal atau tidaknya capaian rendemen yang dihasilkan. Padahal data tersebut sangat penting untuk estimasi profitabilitas pada usaha hilir ulat sutera. Penelitian ini bertujuan untuk mendapatkan formulasi ekstraksi (degumming) yang menghasilkan rendemen fibroin optimum. Metode yang digunakan adalah optimasi ekstraksi fibroin menggunakan Program Response Surface Methodology (RSM) dengan tiga variabel bebas, konsentrasi NaOH, suhu dan waktu. Variabel tersebut diformulasikan dalam rancangan percobaan Central Composite Design (CCD) di Program RSM menggunakan titik pusat NaOH 0,1 N, suhu 105 °C selama 30 menit. Pada rancangan percobaan tersebut, rendemen bobot fibroin (%) digunakan sebagai respon (variabel terikat) untuk menghasilkan kondisi optimum. Kondisi tersebut berupa formulasi variabel bebas yang mengoptimumkan rendemen fibroin. Kondisi optimum Bombyx mori dicapai pada konsentrasi NaOH 0,018 N, suhu 110,53 °C dan waktu 55,51 menit pada satu kali degumming dengan perolehan rendemen fibroin sebesar 71,11 ± 0,98 %. Rendemen fibroin optimum sebesar 83,06 ± 1,50 % pada Attacus atlas dihasilkan dari kondisi optimum NaOH 0,12 N, suhu 79 °C selama 42,65 menit pada dua kali degumming. Metode ekstraksi fibroin yang tepat akan menghasilkan rendemen fibroin yang optimum.

Abstract

Kefir is a highly acidic dairy product with various health benefits. The addition of forest honey plays a role in improving the quality and taste of kefir. This study aimed to evaluate the effect of honey addition and storage time on the physicochemical and organoleptic properties of cow's milk kefir during refrigerator storage. The study was conducted with a 3´3 factorial randomized block design with three replications. The first factor was the added honey (0%, 5%, 10%, and 15%), and the second was the storage time (0, 7, and 14 days). Analysis of variance and Duncan's test were used to analyze the data. The results showed that the added honey significantly affected water activity (P <0.05). In addition, the high concentration of added honey increased the panelists’ preference for kefir acceptance. This study confirmed that the addition of honey and storage time significantly affected the characteristics of the cow's milk kefir. The best treatment was a 15% addition of honey with a storage time of 0 day. Keywords: honey, kefir, milk, physicochemical, storage time

Abstract

Silk fiber is an organic waste that can pollute the environment due to its solubility in processing wastewater. The extraction from wastewater was carried out to reduce environmental pollution and produce natural bioactive. Therefore, this research aims to produce an extraction method that maximizes the protein yield of Attacus atlas (A. atlas) and Samia cynthia ricini (S. ricini) sericin and analyze the characteristics. The method consists of two stages, the optimization of sericin protein extraction with Response Surface Methodology (RSM) and its characterization. The optimization resulted in the optimum extraction of A. atlas and S. ricini sericin at a concentration of 0.08 and 0.03 N NaOH, temperature of 130.52 °C and 113.20 °C, time of 71.71 and 33.78 minutes with a yield of 17.39±1.24% and 20.24±2.30%. The sericin protein had a molecular weight of 8.99 and 7.08 kDa in A. atlas and S. ricini. The extraction produces glycine, alanine, and tyrosin for A. atlas and glutamic acid, glycine, and alanine for S. ricini. Therefore, the sericin extraction formulation did not change the secondary structure protein, as evidenced by the FTIR results.

Abstract

Manuka and forest honeys are types of honeys that are widely consumed because for their antioxidant properties. In this study, the properties of forets and manuka honey and the use of honey as a herbal drink with the addition of Qusthul hindi and turmeric were analyzed. The addition of Qusthul hindi (0,125 g and 0,375 g) and turmeric (0,125 g and 0,375 g) is given in two formulations. This study was conducted in two phase, namely analyzing the characteristics of manuka and forest honey and preparing herbal drinks to then analyzing their properties. The results showed that the properties of forest honey do not meet the standards for the variables of pH, HMF value, and diastase enzyme activity, while Manuka honey meets the standards only for variables of moisture, HMF value, and diastase enzyme activity. The addition of Qusthul hindi and turmeric to herbal drinks results in an increase in pH, DPPH inhibitoty activity, and antioxidant capacity, as well as a decrease of water content, but is not sensory evaluated because the addition of spices causes changes in taste and appearance.
