Abstract

ABSTRACTHigh Energy Diet to Develop Obese Animal Model in Cynomolgus Monkeys (Macacafascicularis). Obesity is a primary predisposition for diseases such as metabolic syndrome(insulin resistance, blood lipid abnormality, and hypertension), type 2 diabetes, andcardiovascular disease. The effort to overcome obesity is needed by understanding thedevelopment of obesity. Therefore, it is urgent to carry out preclinical trials by using an obeseanimal model however it is still limited. The objective of this research was to study the effect ofhigh energy diet with animal fat and soluble carbohydrate to produce obese cynomolgusmonkeys (Macaca fascicularis). Animals used in this study were 15 adult males dividedequally into three treatment groups and were given diets for 4 months. The 3 groups were 1)receiving diet A consists of beef tallow without egg yolk; 2) receiving diet B consists of beeftallow and egg yolk, (energy 4,207 cal/g, fat 19.62%, and starch 60.34% in both first two diet) 3)receiving diet C consists of monkey chow as control (energy 4,330 cal/g, fat 5.55%, and starch51.38%). Measurement were taken every four weeks for body weight, crown rump length, hipdiameter, abdominal skin thickness, body mass index (BMI), nutrient consumption anddigestibility. Results showed that animals received diet B had significant increase in bodyweight and BMI at week 4 and 8. Hip diameter and abdominal skin thickness were significantstarting at week 4 in animals receiving diet with egg yolk (p<0.05) compared to the other twogroups. Protein consumption and digestibility in group with diet A and B was lower (p<0.05)compared to control animals. However, digestibility for dry matter, fat, starch and energy wereabout 90%, meaning that the rations were considered adequate in developing obese monkey.The diet formula containing tallow and egg yolk increased body weight based on BMI criteriaabove 25 kg/m2.Key words: obesity, cynomolgus, egg yolk, body mass index, digestibility

Abstract

Ubi jalar ungu (Ipomoea batatas var Ayumurasaki) dilaporkan dapat membantu diet harian bagi penderita diabetes melitus tipe 2 karena memiliki kadar glikemik yang rendah. Salah satu mekanisme menurunkan kadar gula darah pada penderita diabetes adalah dengan menghambat enzim α-glukosidase. Potensi fraksi alkaloid dari daun dan fraksi antosianin dari umbi ubi jalar ungu dievaluasi sebagai inhibitor enzim α-glukosidase. Ubi jalar ungu yang digunakan dari Ciampea, Bogor, Jawa Barat. Fraksi alkaloid daun ubi jalar ungu pada konsentrasi 2,00% aktif menghambat kerja enzim sebesar 61,88%, sedangkan fraksi antosianin umbi ubi jalar ungu tidak aktif (0,25%). Fraksi alkaloid daun ubi jalar ungu dipisahkan lebih lanjut menggunakan kromatografi kolom dengan fase diam silika gel dan kloroform sebagai eluen. Fraksi yang mengandung alkaloid dengan semprotan Dragendorf, yakni F10, kemudian dipisahkan kembali dengan kromatografi lapis tipis preparatif. Fraksi F10.15 positif mengandung alkaloid dan dapat menghambat enzim a-glukosidase sebesar 18,07% pada konsentrasi 1,25%, yang berarti pemisahan menyebabkan penurunan aktivitas. Spektrum inframerah transformasi Fourier fraksi &nbsp;memperlihatkan gugus fungsi –NH−, –OH, dan –CH2– dan diduga mengandung alkaloid.

Abstract

Pomegranate has valuable nutritional content and contains various bioactive compounds, one found in the fruit's peel. The utilization of these bioactive compounds could be used as herbal medicines and supplements, such as antioxidants. This study aimed to determine the antioxidant capacity, phytochemical profile, and pomegranate peel extract grouping using different extracting solvents. The three extracting solvents used were water, 70% ethanol, and ethanol p.a. Antioxidant capacity of the three extracts was measured using the DPPH and CUPRAC methods. We also determined the total phenolic and flavonoid levels and the TLC fingerprint analysis and FTIR spectrum of the pomegranate peel extracts. The 70% ethanol extract owned the largest antioxidant capacity than the other two extracts with a value of 358.67 and 2981.59 µmol trolox/g dried sample using the DPPH and CUPRAC methods, respectively. The three pomegranate peel extracts' total phenolic and flavonoid levels ranged from 287.26–1068.81 mg GAE/g dried sample and 0.24-0.75 mg QE/g dried sample. TLC fingerprint analysis of pomegranate peel extract yielded 2, 6, and 6 bands for water extract, 70% ethanol, and p.a ethanol, respectively. The three extracts can be grouped based on FTIR spectrum data using principal component analysis using three principal components with a total variance of 93%. The results obtained show that using different extracting solvents provides different antioxidant capacities and phytochemical profiles.

Abstract

STUDY ON BEHAVIOUR OF THE LONG TAIL MONKEY (Macaca fascicularis) OBESE IN INDIVIDUAL CAGE. This study was designed to obtain information of development behaviour the obesity of long-tailed monkey (Macaca fascicularis) fed with high energy obes diet. The need of obese monkeys highly to model byomedis obesity. Obesity is now a universal problem that needs to be explored and studied. Therefore, the objective of this research was to develop obese adult male monkeys with high energy food for 12 months and monitoring their behaviour. Fifteen adult male monkeys 6-8 years old with the weight of between 4-5 kg were divided equally into three groups. The first group was body weight average 4.50 kg, the second group was body weight average 5.00 kg; and the third group was body weight average 4.75 kg. The results showed that body weight gave a significant effect (P <0.01) on daily behaviour and also gave a significant effect (P<0.01) on fed behaviour frequently,drink behaviour, agonistic behaviour, grooming behaviour, and locomotion behaviour .Keywords: Behaviour frequently, Long-tailed monkey (Macaca fascicularis), Obesity)

Abstract

High carbohydrate as obese diet is not yet available commercially for monkeys. Therefore, this preliminary study was to carry out nutrient intake and digestibility of cynomolgus monkeys (Macaca fascicularis) fed with high soluble carbohydrate diet compared to monkey chow. Five adult female macaques (average body weight 2.67 kg) were made to consume freshly  diet. Commercial monkey chows (contains 3500 cal/g energy and 35% starch) were fed to three adult females (average body weight 3.62 kg). Nutrient intakes and digestibility parameters were measured using modified metabolic cages. Result showed that  average of protein, fat, starch, and energy intakes in treatment diet were higher than control diet (T-test). Fat intake in the treatment diet was three times higher, while starch and energy intakes were almost two times higher than monkey chow. Digestibility percentage of all nutrients were the same in both diets except for the protein. The study concludes that the freshly prepared high sugar diet was palatable and digestible for the cynomolgus monkeys. Further studies are in progress to develop obese diet high in energy content based on fat and source of starch treatments.         Key words: Macaca fascicularis, obese, metabolic cages, intake, digestibility

Abstract

The mammary gland contains adult stem cells that are capable of self-renewal and are likely target for neoplastic transformation leading to breast cancer. In this study, we developed a cell culture derived from the mammary glands of cynomolgus monkeys (Macaca fascicularis)(MfMC) and furthermore identified the expression of markers for stemness and estrogenreceptor-associated activities. We found that the primary culture can be successfully subcultured to at least 3 passages, primarily epithelial-like in morphology, the cultured cells remained heterogenous in phenotype as they expressed epithelial cell markers CD24, CK18, and marker for fibroblast S1004A. Importantly, the cell population also consistently expressed the markers of mammary stem cells (ITGB1 or CD29 and ITGA6 or CD49f), mesenchymal stem cells (CD73 and CD105) and pluripotency (NANOG, OCT4, SOX2). In addition to this, the cells were also positive for Estrogen Receptor (ER), and ER-activated marker Trefoil Factor 1, suggesting an estrogen responsiveness of the culture model. These results indicate that our cell culture model is a reliable model for acquiring a population of cells with mammary stem cell properties and that these cultures may also serve as a reservoir from which more purified populations of stem cell populations can be isolated in the future.

Abstract

Bone marrow mesenchymal stem cells have been widely used, because plasticity, specific surface markers, self-renewal to transform into various lineages including cardiomyocytes. Information about the connexin (Cx) cardiac conduction systems of the pigtail macaque (Macaca nemestrina) is limited. This study aimed to evaluate cardiomyocyte differentiation from bone marrow mesenchymal stem cells of pigtail macaques and to clarify the Cx cardiac conduction system. Bone marrow aspirates were obtained from the proximal humerus of four adult male pigtail macaques, collected into heparinized tubes, then centrifuged to obtain mononuclear cells that were isolated and cultured in an incubator. After these cells reached 70–80% monolayer confluency as homogeneous fibroblast-like cells, they were subcultured. On the second subculture passage, the cells were pelleted to extract the mRNA, which was analysed by reverse transcription–polymerase chain reaction, and then cultured for a third passage. Cells were positive for CD73 and CD105 and the reference gene glyceraldehyde-3-phosphate dehydrogenase, and negative for CD34 and CD45. Osteogenic, chondrogenic, adipogenic, and cardiomyocyte differentiation was confirmed based on specific staining. The pigtail macaque bone marrow mesenchymal stem cells can be isolated and subcultured. The transcription of genes and translation of proteins of the connexin cardiac conduction systems was successfully identified.

Abstract

Microencapsulation techniques have been used to improve the stability of carotenoids and in the development of food fortification. The aim of this study was to determine the effect of the ratio of the coating material formulation (maltodextrin (MD) and soy protein isolate (SPI)) to the core material crude palm oil (CPO) on the physicochemical characteristics of the CPO microcapsules. Microencapsulation were carried out with ratios of coating material (MD:SPI = 2:1(w/w)) and CPO of 1:1.0, 1:1.2, 1:1.4, and 1:1.6 (w/w). The mixtures were homogenized at 12.000 rpm for 3 minutes and then spray dryed at inlet temperature of 180°C and outlet temperature at 80°C. The CPO microcapsules were analysed for yield, moisture content, solubility, and encapsulation efficiency. The results showed that the formulation ratio of 1:1.6 (w/w) was the best because it had the highest yield (20.12±0.02%), total carotenoids (295.2±0.32 µg/g) and encapsulation efficiency (59.95±1.50%). The best CPO microcapsules had spherical shape and smooth surface with an average size of microcapsules of 1-12 μm. The results of X-ray diffraction (XRD) showed that microcapsules were in crystalline state with crystallinity and amorphous value of 69.3 and 30.7%, respectively.

Abstract

Nikotin dengan dosis tertentu telah dilaporkan memiliki pengaruh terhadap penurunan nafsu makan pada tikus, tetapi belum pernah dilaporkan pada hewan model monyet ekor panjang. Oleh karena itu penelitian ini bertujuan untuk mengetahui pengaruh nikotin terhadap bobot badan, indeks massa tubuh (IMT) dan kadar glukosa darah monyet ekor panjang (Macaca fascicularis) yang diberi diet berenergi tinggi selama tiga bulan. Sepuluh monyet jantan dewasa secara acak dibagi menjadi dua kelompok. Kelompok pertama diberi pakan dengan bahan utama dari lemak sapi dan kelompok kedua dengan pakan monyet komersial (monkey chow). Cairan nikotin ditambahkan kedua pakan dengan dosis 0,75 mg/kg berat badan. Rancangan yang digunakan dalam penelitian ini adalah rancangan acak lengkap (tersarang) dalam waktu. Data yang dikumpulkan dianalisis untuk mengetahui hubungan antara waktu dan perlakuan. Pengukuran dilakukan setiap empat minggu terhadap berat badan, indeks massa tubuh dan kadar glukosa darah.
