Abstract

Large number of new soybean varieties are mostly derived from crosses of elite genotypes resulted ina narrowing of both the genetic diversity and the phylogenetic relationship between soybean varieties. Thus,discrimination among soybean varieties is becoming more diffi cult, especially when morphological traits wereapplied. In Plant Variety Protection (PVP) system, new varieties of soybeans including granted PVP right, localand breeding varieties registered in PVP offi ce were frequently increased, implicate on increasingly the numberof soybean varieties collections. To assist the management of varieties collections, a standard fi ngerprinting datais further needed. In comparison to the management of plant collection in the fi eld, molecular marker systemswhich are rapid, reliable, informative and relatively simple are continually sought for practical applications ingermplasm conservation, management and enhancement. This study aimed to identify the genetic diversity andphylogenetic relationship of soybean varieties that have earned PVP Right as well as local varieties and breedingvarieties registered in the PVP offi ce using microsatellite or simple sequence repeats (SSR) markers.This study was conducted in Molecular Biology laboratory, Indonesian Center for Agricultural Biotechnologyand Genetic Resources Research and Development (ICABIOGRAD) Bogor, from February to May 2013. The datawere analyzed using the genetic analysis package NTSYSpc 2.02i and PowerMarker V3.25. The result showed arelatively narrow genetic diversity among 45 varieties of soybean analyzed in present study which were indicatedby the small number of genotypes and total number of alleles (NA), and the low value of gene diversity and PICvalues (<0.75). Cluster analysis showed that the grouping varieties are not related to morphological characters butrelated to phylogeny relationship between varieties. Despite the group of varieties were not clustered in accordancewith morphological characteristics, SSR marker can be a powerful tool for discriminating varieties, so that it couldbe useful for initial varieties identity in conjunction with genetic diversity analysis.

Abstract

Kelurahan Kauman is one of the cultural heritage kampong in Surakarta, formerly inhabited by abdi dalem ulama Keraton and batik entrepreneurs. Kauman developed when Pakubuwono III built the Great Mosque in 1757, the kampong then developed into a batik industrial center in the early 1800s, shown by the number of luxury homes built by batik entrepreneurs that year. But in 1939 to 1970 batik companies went bankrupt; Therefore, the ancient houses and buildings were damaged and became a slum neighborhood. Revitalization of the area has been done since 2006, the economy rose again but the environmental conditions have not been optimal. The purpose of this study is to manage the revitalization program that has been done to find a better settlement strategy. The research was conducted by qualitative rationalistic approach. Data were collected through literature studies, interviews and field surveys. The sample selection is purposive, the sample being studied is the impact of revitalization on physical and non-physical environment and community activities of Kauman. Revitalization management is needed to make Kauman more comfortable in the future as a healthy settlement and can be developed into a tourist kampong to improve the welfare of the people who live in it

Abstract

Beras hitam saat ini mulai populer sebagai pangan fungsional seiring dengan kesadaran akan pentingnya kesehatan. Namun, beberapa kendala budidaya mempengaruhi minat petani dalam memproduksi beras hitam, seperti umur tanaman yang panjang, kerentanan terhadap WBC, dan produksi yang rendah. Persilangan dapat dilakukan untuk membentuk tanaman baru dengan sifat yang diinginkan. Metode silang balik digunakan untuk memperbaiki varietas yang sudah mempunyai karakter yang baik, tetapi kurang unggul pada beberapa karakter. Penelitian ini dilakukan untuk memperoleh hasil persilangan backcross 1 dengan tetua (Padi Hitam Jeliteng) sehingga dapat dihasilkan benih generasi backcross 2 untuk dilanjutkan pada backcross generasi berikutnya. Metode persilangan yang digunakan adalah persilangan balik antara 6 nomor (BC1GH8; BC1GH13; BC1GH44; BC1GH46; BC1GH51; BC1GH52) hasil persilangan BC1 antara galur-galur potensial galur padi hitam/Jeliteng//Jeliteng yang disilangkan lagi dengan salah satu tetua, yaitu varietas Jeliteng. Seluruh data hasil pengamatan dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa terdapat perbedaan hasil pada variabel umur berbunga, masa pengisian bulir gabah, dan persentase keberhasilan persilangan. Persilangan backcross 2 menghasilkan keturunan F1 Backcross 2 yang bersifat fertil dan dapat digunakan sebagai tetua untuk persilangan backcross generasi berikutnya. Rata-rata persentase keberhasilan dari keseluruhan hasil set persilangan backcross 2 adalah 44,18%. Penelitian ini diharapkan memperoleh hasil persilangan backcross 1 dengan tetua (Padi Hitam Jeliteng) sehingga dapat dihasilkan benih generasi backcross 2 untuk dilanjutkan pada backcross generasi berikutnya.
