Abstract

The rumen of cattle is well known one of a bacterium cellulolitic production that have ability to degrade cellulose for produce cellulase enzymes. The aims of the present study is to determine the type of bacteria produced cellulase enzymes in the rumen of cattle aceh. The diversity of cellulolitic bacteria is determined by  analysis of 16S rRNA genes. Isolation of cellulolytic bacteria, total DNA isolation, agarose gel electrophoresis, amplification of the 16S rRNA gene, determining the DNA sequence, DNA sequence homology and alignment of DNA sequences. Isolation of bacterium cellulolitic, isolating total DNA, gel electroforesis agarosa, amplified of gen 16S rRNA, determination the DNA barcode,  homology DNA barcode, and alignment of DNA sequences. The sample of rumen liquid was taked from RPH , then maked isolation of total DNA to get pure DNA from bacterium whos result and amplification DNA of bacterium cellulolitic separated by electroforesis gel agarosa. Gene of bacterium cellulolitic was isolated based Polymerase Chain Reaction methode. Alignment is done by using BlastN program, then use CLustalW program.  It can be concluded that the species of bacteria (FKH_USK_6) in rumen has a closeness with Coronabacter sakazakii bacteria strain PNP8 of 97% according to the sizeof the DNA sequence and the same position on an existing gene in GenBank.

Abstract

This study aimed to isolate and identify fungus that causes aspergillosis in the quail (Coturnix japonica). The samples used in this study were 30 pairs quail lungs taken randomly from the Hanin Group Quail Farm, Garot village, Darul Imarah subdistrict, Aceh Besar regency. The growth of the fungus was observed for Aspergillus sp. macroscopically and microscopically. The data were analyzed descriptively. The results of this study revealed that from 30 pairs quail lungs, Aspergillus sp. can be isolated from 26 out of 30 lungs. It can be concluded that 86.67% of the lung samples were infected with Aspergillus sp.

Abstract

This study aimed to determine the number of lobes and thymus regression time of ALPU chickens. The samples were 76 ALPU chickens aged 1 – 19 weeks, consisted of 38 male and 38 female chickens. Two male and 2 female chickens were sacrificed every week and the thymus were collected and weighed. The results showed that the increase in thymus weight occurred in the age range of 1 – 15 weeks and reached the maximum weight at the age of 15 weeks. Thymus was decreased in weight at the age of 16 – 17 weeks. Thymus regressesion occurred at the age of 18-19 weeks. The thymus had 5-9 lobes. The result of T test on the average weight of ALPU chicken thymus showed that the weight of male ALPU chicken thymus of starter phase compared with grower phase was significantly different (p0.05), while the grower phase with finisher phase was not significant or there was no difference (p0,05). The result of T-test for weight of female ALPU thymus showed that the weight of thymus of starter phase compared to grower phase chickens was significantly different (p0.05), whereas grower phase with finisher result was not significantly difference (p0.05).Therefore, it was concluded that the number of thymus lobes of ALPU chickens are 5 - 9 lobes and the time of thymus regression of ALPU chicken occurs at the age of 18 - 19 weeks.

Abstract

This study aimed to determine the regression time of bursa Fabricius in ALPU chicken. In this research, 72 chickens of ALPU consisted of 36 males and 36 females, aged 1 week, 2 weeks,, until 18 weeks old were used. Each age group consisted of 2 male chickens and 2 female chickens. Bursa Fabricius of every age group was collected and weighed. The results showed that the weight of bursa Fabricius increased at 1 to 15 weeks old, and reached the maximum weight at 15 weeks of age. After reaching maximum weight, bursa Fabricius underwent regression that occurred at 16 to 18 weeks old. Bursa Fabricius’s weight increased in the starter phase at 1 to 6 weeks, grower at 7 to 12 weeks and will regress in the finisher phase at 13 to 18 weeks. The results of the average weight analysis of bursa Fabricius using t-test was observed by comparing the weight of bursa Fabricius ALPU chicken starter phase with grower phase and grower phase with finisher phase. The weight of bursa Fabricius of male ALPU on starter phase with grower phases was significanly difference (p0.05) whereas grower phase with finisher phase was not significantly difference (p 0,05). The weight of bursa Fabricius of female ALPU on starter phase with grower phases was significanly difference (p0.05), whereas grower phase with finisher phase was not significant (p 0,05). Therefore, it can be concluded that regression of bursa Fabricius occurred at the age of 16 to 18 weeks. The weight of bursa Fabricius in starter phase and grower phases are significantly difference, however there was no significantly difference was observed in finisher phase.
