Abstract

This study aimed to investigate the phenotypic and genotypic of antibiotics resistance profile in Escherichia coli. The 30 samples come from cloacal swab of broilers in Cianjur, Indonesia. Isolation and identification of E. coli was performed by culture in McConkey agar, eosin methylene blue agar, Gram staining and five essential biochemical tests (IMViC). In this study, 10 isolates (33.3%) were confirmed E. coli positive. Phenotypic profile was performed by screening all isolates with 8 antibiotics of 6 antibiotic groups. The screening was carried by Kirby-Bauer disk diffusion method based on the standard of CLSI. For genotypic profile, each resistant isolate was detected antibiotic resistance-encoding gene. The result showed all isolates (100%) resistant against tetracyclin, oxytetracycline and erythromycin. Nine isolates (90%) detected nalidixic acid and enrofloxacin-resistant. The ciprofloxacin and gentamicin-resistant isolates were 70% and 40%, respectively. There was no resistant isolate for chloramphenicol. Multi drug-resistant was detected on 90% isolates. Only gyrA (100%) and tetA (80%) genes were detected. This study showed high rate of occurrence of antibiotic resistance in E. coli. Not all resistant isolates were detected in the antibiotic resistance-encoding gene in this study. Future research to detect resistance genes should use more varied target genes.

Abstract

Animal protein needs in Indonesia increase every year. One source of animal protein that is affordable for all levels of society is poultry. Poultry mostly farmed in several parts of Indonesia. The region that has the largest poultry population in Indonesia is the province of West Java. The regions in West Java that contribute the highest poultry populations are Sukabumi and Bogor. The largest number of poultry populations is broiler chicken population, which is 108,304,978 tails.Antibiotics often used in livestock and poultry as a prevention of disease transmission and as a growth promoter. Routine use of antibiotics in the livestock industry, especially chickens, has a negative impact. One of them is the emergence of antibiotic resistance. One of the bacteria that has experienced antibiotic resistance is the Klebsiella species.Klebsiella bacteria are Gram-negative bacteria are normal flora in the oral cavity, skin and intestines, but can become pathogens in animals and humans under certain conditions. This bacterium is one of the causes of bacterial diseases that attack large livestock and poultry.  Many reports say that Klebsiella species have experienced resistance to antibiotics. Klebsiella had experienced resistance to several antibiotics, namely ampicillin as much as 66.7%, Nalidixic Acid 61.8%, tetracycline 59.8% and trimethropin 50%. Klebsiella bacteria that are resistant to antibiotics are very dangerous for animal and human health. Diseases caused by Klebsiella bacteria that are resistant to antibiotics will be difficult to cure. Akova (2016) states that Gram-negative and Gram-positive bacteria that experience resistance will produce infections that are difficult to treat or cannot even be treated using antimicrobials.

Abstract

Most of poultry industries use antibiotics for health management program. They use them as bacterial infection treatment and disease prevention, known as antibiotic growth promotor (AGP) [1]. Now, worldwide concern is about antibiotics resistance. Monitoring programs are done by countries in the world to protect human and animal health [2]. The monitoring programs usually use indicator bacteria such as Escherichia coli [3].Many researchers studied about antibiotics resistance in Indonesia. Escherichia coli isolated from poultry has been resistance to doxycycline (25%) and gentamycin (12.5%) [4]. Seven E. coli isolated from fecal samples shown that resistance to methicillin (85.7%), penicillin G (71.4%) and 42,9% were resistance both doxycycline hydrochloride and streptomycin [5]. Start from Januari 1st 2018 Indonesian Misintry of Agricuture banned antibiotics as AGP, based on Permentan No. 14/2017. They tried to prevent the spread of antibiotics resistance. So, it is necessary to determine antibiotics resistance patterns, especially in E. coli as indicator bacteria.

Abstract

ABSTRAKPermasalahan resistensi Antibiotika pada hewan kesayangan menjadi kendala kesehatan hewan di seluruh dunia. World Health Organisation (WHO) menyebutkan bahwa pada masa mendatang resistensi antibiotika akan menjadi tantangan yang terbesar dalam dunia kesehatan. Penelitian ini bertujuan untuk mengukur tingkat resistensi antibiotika terhadap Escherichia coli yang diisolasi dari kucing pada klinik hewan di Kota Bogor. Hasil penelitian menunjukan bahwa resistensi Escherichia coli tertinggi terjadi pada golongan β-laktam (ampisilin 66% dan amoksisilin 60%) yang diikuti oleh golongan tetrasiklin (oksitetrasiklin 54% dan dosisiklin 24%), serta golongan kuinolon (enrofloksasin 38% dan ciprofloksasin 28%). Hasil dari penelitian ini diharapkan dapat menjadi pertimbangan medis bagi praktisi hewan kesayangan dalam penggunaan antibiotika.Kata Kunci: Escherichia coli, klinik hewan, kucing, resistensi antibiotikaABSTRACTThe problem of antibiotic resistance in pets is obstacles to animal health throughout the world. World Health Organization (WHO) states that in the future, antibiotic resistance will become the biggest challenge in the health concern. This study aims to measure the level of Escherichia coli resistance to antibiotics which is isolated from cats on veterinary clinics in Bogor City. The results showed that the highest resistance of Escherichia coli occurred in the β-lactam group (ampicillin 66% and amoxicillin 60%) followed by tetracycline (oxytetracycline 54% and doxycycline 24%), and quinolone group (enrofloxacin 38% and ciprofloxacin 28%). Abstract

Leptospirosis is a zoonotic disease caused by bacteria Leptospira sp. which causes infection in animals and humans. Dogs infected with leptospirosis showed symptoms such as anorexia, fever, vomiting, weakness, diarrhea and often experience yellowing of the eye area and mucosa around the mouth (icteric) with fatal systemic complications and multi-organ dysfunction, especially in the kidneys and liver. Leptospirosis is an endemic disease in Jakarta. This study aims to identify risk factors that can contribute to canine mortality based on early clinical symptoms that are found when the dog is in an animal health service facility such as a veterinary clinic, veterinary hospital or independent practice veterinarian. Method were used in this study is clinical manifestations and laboratory examinations and medical records of dogs with suspected leptospirosis. Criteria inclusion were based on aspects of the clinical symptoms of dogs in and around Jakarta. Analysis data used the chi-square with confidence of interval (CI) 95%. Dogs used during the study had ages for puppies (less than 1 year) totaling 13 or 32.50%, for adult dogs over 1 year amounted to 27 or 67.50%, 80% male dogs and 20% female. with 80% maintenance system not housed by the owner. Risk factors for clinical symptoms such as myalgia, symptomatic vomiting of the pulmonary area or shortness of breath and abdominal pain, conjunctival suffusion, anorexia and diarrhea contributed to the high mortality rate leptospirosis during study in dogs 2020.

Abstract

Chicken is one of the important protein source in Indonesia. Moreover, the largest population of chicken layer and poultry in Indonesia is known situated at West Java province with Bogor manicipality as the main producer. The aims of this study were to determine the antibiotic resistance pattern of Staphylococcus aureus isolated from poultry and layer farm in Bogor. The study also identified gene encoded the resistance. Cloacal swab samples were collected from chicken broiler and layer farm in Bogor manicipality. The samples were then cultured in Mannitol Salt Agar (MSA) medium to obtain S. aureus. Suspected colony was then confirmed by biochemical test. Positive strains were tested against several antibiotics and the diameter of clear zone arround of colony was interpreted based on Clinical and Laboratory Standard Institute. Furthermore, the DNA from resistant strains were then extracted, followed by detection of the resistance gene by using polymerase chain reaction (PCR) method. A total of 14 isolates of S. aureus were positive from poultry farm, and 15 isolates from layer farm. Most of all were resistant to tetracycline, ampicillin, oxytetracycline, erythromycin and nalidixic acid. On the other hands, several strains were sensitive to gentamycin and chloramphenicol. The study showed 28 isolates out of them were multi-drug resistant. Resistant gene such as blaTEM, gyrA and tetA were also identified in some isolates except for ErmB gene which was found in isolates originated from poultry farm. In conclussion, S. aureus in both farm showed mostly multi-drug resistant to several antibiotics which were supported by identification of resistant gene among isolates.

Abstract

Penelitian ini bertujuan menghitung jumlah koloni bakteri selulolitik pada sekum ayam kampung. Sampel yang digunakan yaitu isi sekum ayam kampung yang diambil di Tempat Pemotongan Unggas (TPU) Lambaro Aceh Besar sebanyak 5 ekor ayam kampung. Sampel dilakukan pengenceran 10-2 sampai 10-5, kemudian diinokulasi ke dalam media BHM-CMC agar dengan metode tuang (pour plate). Selanjutnya, diinkubasi dengan suhu 37˚C selama 96 jam. Koloni bakteri selulolitik yang tumbuh kemudian dilakukan pengamatan morfologi koloni (warna, posisi, pinggiran, dan diameter koloni), diameter zona bening, penghitungan jumlah total bakteri, dan pewarnaan Gram. Hasil penelitian menunjukkan bahwa koloni bakteri berwarna putih, berada di dalam dan di permukaan agar, memiliki pinggiran rata, diameter koloni 2-3 mm dan diameter zona bening 18 mm dan 29 mm. Bakteri berbentuk basil bereaksi negatif terhadap pewarnaan Gram, sedangkan bakteri berbentuk kokus bereaksi positif dan negatif terhadap pewarnaan Gram. Jumlah total bakteri pada isi sekum ayam kampung yaitu 5.3 x 105 cfu/g.Kata kunci: Ayam kampung, bakteri selulolitik, sekum, Carboxy Methyl Cellulase.                The aim of this study was to count the total count of cellulolytic bacteria colony in caecum of native chicken. The sample used was caecum contents of native chicken which taken from the poultry slaughterplace at Lambaro Aceh Besar. Sample were diluted from 10-2 to 10-5, then inoculated to agar BHM-CMC media with pour plate method. Then, incubated with temperature 37oC about 96 hours. A colony of cellulolytic bacteria which were grown then were observed by using colony bacteria criteria (colour, position, edge, and diameter of the colony), diameter of the bacterial zone, counting a total of bacteria which was grown, and Gram stain. The results showed that there is white bacterial colony, located inside and surface agar, has flat periphery, diameters of colony 2-3 mm and diameters of bacterial zone 18 and 29 mm. Bacill-shaped bacteria react negative to Gram staining, whereas coccus-shaped bacteria react positive and negative to Gram staining. Number of total bacteria in the native chicken’s caecum was 5.3 x 105 cfu/g Keywords: Native chicken, cellulolytic bacteria, caecum, Carboxy Methyl Cellulose.

Abstract

Hasil penelitian menunjukkan bahwa terdapat morfologi koloni berwarna putih bening, posisi di dalam dan di permukaan agar, pinggiran bergerigi, diameter koloni 2-4 mm dan zona bening 19 mm sampai 26 mm. Bakteri selulolitik berbentuk basil Gram negatif dan kokus Gram positif. Rata-rata jumlah total bakteri pada isi ilium ayam kampung yaitu 3,3 x 105 cfu/g. The aim of this study was to count the total sum of cellulolytic bacteria colony in the illium of native chicken. A colony of cellulolytic bacteria which were grown then were observed by using colony bacteria criteria (colour, position, edge, and diameter of the colony), diameter of the bacterial zone, counting a total of bacteria which was grown, and Gram stain. The results showed that there is white translucent colony morphology, position inside and surface agar, cogged periphery, diameters of colony 2-4 mm and bacterial zone 19 to 26 mm. Cellulolytic bacteria Gram-negatif bacilli, Gram-negative cocci and Gram-positive cocci. The average number bacteria of total bacteria in the native chicken’s illium was 3.3 x 105 cfu/g.

Abstract

Lactic Acid Bacteria (LAB) has many benefits for human and animal health and has been widely used as a probiotic. One of the LAB is the genus Lactobacillus which consists of many species used for fermentation and food preservation. This study was conducted to isolate and identify the LAB of the genus Lactobacillus from the faeces of wild Sumatran Orangutan (Pongo abelii) at the Suaq Belimbing Research Station in South Aceh. Bacterial isolation was carried out using Man Rogosa Sharpe Agar (MRSA). Colonies that grew on MRSA media were observed for morphology and were Gram stained. Biochemical tests were conducted using KIT API 50 CHL. Data analysis used the Apiweb computer program Version V-5.2. The results showed that the OUL isolate was a species of Lactobacillus delbrueckii ssp delbrueckii, with an identity of 93.8%. Based on this, it can be concluded that there is a Lactic Acid Bacteria of the Lactobacillus in the faeces of wild Sumatran Orangutan (Pongo abelii) at the Suaq Belimbing Research Station in South Aceh.

Abstract

A major current problem in public health is the issue of antimicrobial resistance of Escherichia coli in humans and poultry. In Indonesia, multidrug-resistant E. coli are of specific concern since such E. coli may cause public health problems in humans. The prevalence of multidrug-resistant chicken E. coli strains and the E. coli resistance genes, which are tet(A) and tet(B) genes, were investigated in the present study. A total of 57 swabs were collected from layer and broiler breeder farms in West Java, Indonesia, and used in the experiment. Eighteen isolates were identified as E. coli by the disk diffusion method. The isolates classified as drug-resistant and intermediate were then identified using PCR for the antimicrobial resistance genes. The results showed that 18 isolates of E. coli from layer-breeder and broiler-breeder farms in West Java were resistant to ampicillin (100%), nalidixic acid (94%), tetracycline (88%), oxytetracycline (83%), gentamicin (27%), and chloramphenicol (22%). PCR identification of E. coli antimicrobial-resistant genes in 18 isolates showed tet(A) and tet(B) genes. This study reports antimicrobial resistance genes among E. coli on layer and broiler breeder farms in West Java. This present study showed that E. coli isolated from layers-breeder and broiler-breeder farms in West Java of Indonesia carried tet(A) and tet(B) genes, the multidrug-resistance genes.
