Abstract

Biofilm is one of parameters for determining pathogenic pathways of a bacterium. Bacterial virulence activities can be attenuated by antibiotics with inhibiting biofilm. The aim of this study was to examine the characteristics and activities of antibiotics against the biofilm of Stapylococcus aureus isolate which taken from Aceh cattle prepuce. The study was exploratory and experimental, using a Completely Randomized Design (RAL) factorial pattern using three types of antibiotics, three concentrations and replications. Antibiotics were tetracycline, oxytetracycline and phosphomycin with concentrations of 10%, 20% and 30% (mg/mL), respectively. Therefore, the aim of this study was to examine the characteristics and activities of antibiotics to the biofilm of Stapylococcus aureus from Aceh cattle prepuce isolate. U-bottom polystyrene 96 wells microplate and Nutrient Broth (NB) medium was used to biofilm bacterial while biofilm was stained on crystals violet 1%. Optical Density (OD) of biofilm were λ 595 nm. Data were analyzed by using Analysis of Variance (ANOVA). The results showed that the activity of Staphylococcus aureus was moderately positive, i.e 2,667cell/μL. It showed that antibiotics and concetrations have significant effect (P<0.05) to inhibit biofilm staphylococcus aureus. In additioon, tetracycline 30% (mg / mL) has the best ability to inhibit biofilms from 52,18% to 1,262 cell/μL. Based on that fact, it can be concluded that the virulence of Staphylococcus aureus was moderate positive to be false positive.

Abstract

Aspergillosis merupakan penyakit yang disebabkan oleh jamur Aspergillus sp. yang dapat menyerang puyuh muda secara akut dan sebagai penyakit menahun pada puyuh dewasa. Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Aspergillus sp. pada kantung hawa puyuh (Coturnix japonica). Sampel yang digunakan dalam penelitian ini adalah 35 kantung hawa puyuh yang tidak produktif lagi (afkir) diambil secara acak. Isolasi dan identifikasi Aspergillus sp. dilakukan sesuai dengan menggunakan metode Thompson. Sampel dicuci dengan aquadest steril berisi antibiotic gentamicin selanjutnya ditanamkan pada media Sabouraud’s Dextrose Agar (SDA) kemudian diinkubasikan pada suhu kamar selama 3-7 hari. Pertumbuhan koloni Aspergillus sp. diamati morfologinya secara makroskopis dan mikroskopis. Data yang diperoleh dianalisis secara deskriptif. Hasil pemeriksaan menunjukkan bahwa Aspergillus sp. dapat diisolasi dan diidentifikasi pada 24 dari 35 sampel kantung hawa. Berdasarkan hasil penelitian dapat disimpulkan bahwa 68,57% sampel yang diperiksa positif terinfeksi Aspergillus sp. dengan persentase masing-masing spesies Aspergillus fumigatus 40%, Aspergillus flavus 25,7%, dan Aspergillus niger 17,1%.

Abstract

Burung trucukan (Pycnonotus goiavier) merupakan salah satu burung liar yang sering dipelihara karena kicauannyayang merdu. Infeksi ND dapat berupa infeksi yang akut atau kronis yang menyerang burung trucukan dan jenis burunglainnya. Burung liar merupakan salah satu sumber virus ND yang dapat menyebarkan virus ke unggas peliharaan.Penelitian ini bertujuan untuk mengetahui titer antibodi terhadap penyakit tetelo pada burung trucukan liar. Sampel yangdigunakan adalah serum darah dari 50 ekor burung trucukan liar. Parameter yang diamati adalah titer antibodi dalamserum berdasarkan metode Hirst menggunakan uji Haemagglutination Inhibition (HI) untuk melihat titer antibodi. Untukmenghitung titer virus maka uji Haemaglutination (HA) dilakukan sebelumnya. Untuk memastikan titer antigen yangdigunakan adalah 4HAU dilkukan juga back titrasi sehingga didapatkan nilai virus optimal yang akan dipakai pada uji HI.Hasil penelitian menunjukkan 45 dari 50 sampel (90%) mempunyai antibodi positif terhadap ND, dengan titer berkisar 21-29. Berdasarkan hasil yang diperoleh dapat disimpulkan bahwa 90% burung trucukan di Kabupaten Aceh Besar sudahpernah terpapar ND, tetapi hanya 80% yang memiliki titer antibodi yang dapat melingdungi terhadap serangan virus ND.Oleh sebab itu dapat disimpulkan bahwa burung trucukan liar dengan titer antibodi yang protektif berarti sudah pernahterpapar virus ND sebelumnya akan berpotensi sebagai pembawa virus yang dapat ditularkan ke unggas peliharaanmasyarakat di sekitarnya.

Antibiotik dapat melemahkan aktivitas virulensi bakteri dengan menghambat pembentukan biofilm. Penelitian ini bertujuan untuk melihat karaksteristik dan aktivitas antibiotik terhadap biofilm Stapylococcus aureus isolat preputium sapi Aceh. Penelitian bersifat eksplorasi dan eksperimental, menggunakan Rancangan Acak Lengkap (RAL) pola faktorial dengan menggunakan tiga jenis antibiotik, tiga konsentrasi dan tiga kali ulangan. Antibiotik yang digunakan adalah tetrasiklin, oksitetrasiklin dan fosfomisin dengan konsentrasi masing-masing 10%, 20% dan 30% (mg/mL). Pengujian biofilm dilakukan pada media Nutrient Broth (NB), menggunakan mikroplat U-bottom polystyrene 96 wells. Pewarnaan biofilm menggunakan kristal violet 1%. Optical Density (OD) biofilm dibaca pada    λ 595 nm. Data hasil penelitian dianalisis dengan menggunakan Analysis Of Variance (ANOVA). Hasil penelitian menunjukkan aktivitas virulensi Staphylococcus aureus isolat preputium sapi Aceh berdasarkan karaksteristik biofilm tergolong positif moderat, yaitu 2,667cell/µL. Uji Biofilm menunjukkan jenis dan konsentrasi antibiotik berpengaruh secara nyata (P<0,05) terhadap daya hambat biofilm Staphylococcus aureus isolat preputium sapi Aceh. Tetrasiklin 30% (mg/mL) mempunyai kemampuan paling baik menghambat pembentukan biofilm Staphylococcus aureus isolat preputium sapi Aceh sebesar 52,18% menjadi 1,262 cell/µL, sehingga aktivitas virulensi Staphylococcus aureus isolat preputium sapi Aceh berdasarkan karaksteristik biofilm dari positif moderat menjadi positif palsu. Kata Kunci: biofilm, antibiotik, Staphylococcus aureus, sapi aceh.

Abstract

This study was aimed to observe correlation between gestational age and fetal growth of Kacang goat for 60 days after mating with transcutaneous ultrasonography visualization. Three female goats of productive age with normal estrus cycle and one male goat were used in this study. The female goats were synchronized with double injection of prostaglandin F2á (PGF2á) intramuscularly with dosage of 1 mL for each female goat with 11 days’ time interval. Observation of oestrus was conducted using male goat after being synchronized. When the sign of estrus were prominent, the female goats were naturally mated. The result was pregnant positive for one female goat. Detection of early pregnancy was observed on the 24th days after mating, with the result of 6.77 mm length embryo by isoechogenic visualization. On 35th days of pregnancy, fetal could be seen clearly in isoechogenic to hyperechogenic visualization with head diameter and fetal length were 17.4 mm and 36.2 mm, consecutively. The observation also found the placentom. On day 45 of pregnancy, head diameter and fetal length were 21.8 mm and 40.6 mm. Later on day 49 of pregnancy, the size of gestational saccus was 44.1 mm, with head diameter of 25 mm, and average placentom diameter size of 12.4 mm. On day 53 of pregnancy, head diameter was 25.2 mm with fetal length of 63.6 mm and placentom diameter of 15 mm. On day 56 of pregnancy, vertebrae of fetal were observed using hyperechogenic visualization and placentom was measured 17.9 mm in diameter. On day 60 of pregnancy, we observed that the fetal length was 79.8 mm and the organs such as eyes, heart, liver, os costae, and fetal extremities, could be observed clearly. The size of developing fetal and organs would grow along with the addition of gestational age.

Abstract

TOTAL BAKTERI ASAM LAKTAT (BAL) PADA FESES RUSA SAMBAR (Cervus unicolor) DI TAMAN RUSA ACEH BESAR  ABSTRAK          Penelitian ini bertujuanuntuk mengetahui total bakteri asam laktat (BAL) pada feses rusa sambar (Cervus unicolor) di Taman Rusa Aceh Besar. Sampel yang digunakan adalah feses dari 6 ekor rusa sambar.Penghitungan total BAL menggunakanmetode Total Plate Count (TPC). Sampel diencerkan dari 10-1 sampai dengan 10-8,kemudiandiinokulasikan ke dalam media MRS Agar dengan menggunakan metode spread plate.Cawan Petri diinkubasi 37oC selama 48 jam. Koloni bakteri asam laktat yang tumbuh kemudian dilakukan pengamatan berdasarkan morfologi koloni, pewarnaan Gram, uji katalase dan penghitungan total bakteri asam laktat yang tumbuh. Berdasarkan hasil pengamatan morfologi kolonidapat dilihat ada 5 macam BAL yang tumbuhpada media MRSAgar. Bakteri yang tumbuh semuanya termasuk ke dalamkelompok bakteri Gram positif (warna ungu) dan katalase negatif. Hasil penelitian ini dapat disimpulkan bahwa total BAL pada feses rusa sambar adalah 5x109 CFU/ml. The Total of Lactic Acid Bacteria (LAB) on Feces of Sambar Deer (Cervus unicolor)  in Taman Rusa Aceh Besar   ABSTRACT            This study aims to know the total lactic acid bacteria (LAB) on the feces of sambar deer (Cervus unicolor) in Taman Rusa Aceh Besar. The sample used was the feces of  6 sambar deers. Total calculation of BAL used Total Plate Count (TPC) method. Samples were diluted from 10-1 to 10-8, then inoculated into MRS Agar medium by using the spread plate method. The Petri dish was incubated 37oC for 48 hours. The colonies of lactic acid bacteria that grew later were observed based on colony morphology, Gram staining, catalase test and total lactic acid bacteria count. Based on the observation of colony morphology, it can be seen that there were 5 kinds of  LAB growing on MRS Agar medium. The all-growing bacteria belong to the group of Gram-positive (purple) bacteria and negatif catalase. The results of this study can be concluded that the total LAB from feces of  sambar deer was 5 x 109 CFU / ml.

Abstract

ABSTRAK Escherichia coli merupakan salah satu bakteri penyebab foodborne disease yang dapat menimbulkan penyakit berbahaya apabila bahan pangan yang terkontaminasi dikonsumsi oleh manusia. Salah satu bahan pangan asal hewan yang sering dikonsumsi masyarakat adalah daging ayam, baik daging ayam kampung maupun daging ayam broiler. Penelitian ini bertujuan untuk mengetahui angka prevalensi bakteri E. coli pada daging ayam broiler yang dijual di Tiga Pasar Tradisional Kota Banda Aceh. Sampel yang digunakan dalam penelitian ini adalah 25 sampel daging ayam broiler yang diambil dari Tiga Pasar Tradisional. Penelitian ini menggunakan metode total plate count dengan larutan buffered peptone water (BPW) dan media selektif differensial eosin methylene blue Agar (EMBA). Hasil penelitian menunjukkan bahwa sembilan sampel daging ayam broiler positif tercemar E.  coli dengan angka prevalensi yaitu di Pasar Seutui (100%) dan Pasar Peunayong (26,7%). Dapat disimpulkan bahwa cemaran bakteri E. coli melebihi batas normal dengan angka prevalensi secara keseluruhan dari Tiga Pasar Tradisional di Kota Banda Aceh yaitu sebesar 36%.  Kata kunci : Escherichia coli, daging ayam broiler, foodborne disease, prevalensi ABSTRACT Escherichia coli is one of the bacteria that cause foodborne disease which can cause dangerous diseases if contaminated food is consumed by humans. One foodstuff of animal origin that is often consumed by the public is chicken meat, both native chicken meat and broiler chicken meat. This study aims to determine the prevalence of E. coli in broiler chicken meat sold in the Three Traditional Markets in Banda Aceh. The sample used in this study was 25 samples of broiler chicken meat taken from the Three Traditional Markets. This study uses the total plate count method with buffered peptone water (BPW) and differential selective media eosin methylene blue agar (EMBA). The results showed that nine samples of broiler’s chicken meat were positively contaminated E. coli with prevalence rates in Seutui Market (100%) and Peunayong Market (26.7%). It can be concluded that the contamination of E. coli exceeds normal limits with an overall prevalence rate of the Three Traditional Markets in Banda Aceh City is 36%. Keywords : Escherichia coli, broiler chicken meat, foodborne disease, prevalence

Abstract

ABSTRAKEscherichia coli dikenal sebagai bakteri indikator sanitasi, keberadaanya dalam pangan menunjukkan bahwa pangan tercemar oleh lingkungan. Penelitian ini bertujuan mengetahui angka prevalensi bakteri E. coli pada meja dan peralatan pedagang daging ayam broiler. Sampel yang digunakan yaitu 20 swab meja, pisau, dan talenan pedagang daging ayam broiler di dua Pasar Tradisional Kota Banda Aceh. Penelitian ini menggunakan metode Total Plate Count (TPC) dengan larutan Buffer Peptone Water (BPW) dan mengidentifikasi E. coli menggunakan media selektif diferensial, yaitu dengan Eosin Methylene Blue Agar (EMBA). Hasil penelitian menunjukkan bahwa 2 meja, 2 pisau, dan 3 talenan di Pasar Tradisional Seutui positif tercemar E. coli dengan angka prevalensi cemaran E. coli pada meja (40%), pisau (40%), dan talenan (60%), sedangkan di Pasar Tradisional Peunayong terdapat 2 meja dan 2 pisau positif tercemar E. coli dengan angka prevalensi cemaran  E. coli pada meja (13%) dan pisau (13%). Hasil kesimpulan yang didapat yaitu rata-rata angka prevalensi cemaran E. coli secara keseluruhan pada peralatan pedagang dari dua Pasar Tradisional di Kota Banda Aceh yaitu 18,33%.Kata kunci : Escherichia coli, prevalensi, peralatan pedagang, pasar tradisional  ABSTRACTEscherichia coli is known as an indicator of sanitation bacteria, its presence in food indicates that food is polluted by the environment. This study aims to determine the prevalence of  E. coli bacteria on the table and equipment of broiler chicken meat traders. The samples used were 20 table swabs, knives, and cutting boards of broiler traders in two Traditional Markets in Banda Aceh City. This study uses the Total Plate Count (TPC) method with Buffered Peptone Water (BPW) solution and identifies E. coli using differential selective media, namely Eosin Methylene Blue Agar (EMBA). The results showed that 2 tables, 2 knives, and 3 cutting boards in the Seutui Traditional Market were positively contaminated with E. coli with the prevalence of E. coli contamination on the table (40%), knives (40%), and cutting boards (60%), while in the Peunayong Tradisional Market there are 2 tables and 2 positive knives contaminated with E. coli with the prevalence rate of E. coli contamination on the table (13%) and knives (13%). The conclusion obtained is the average prevalence rate of E. coli contamination as a whole on the equipment of traders from two Traditional Markets in the City of Banda Aceh is 18.33%. Key words : Escherichia coli, prevalence, equipment traders, traditional markets

Abstract

ABSTRAK                Penelitian ini bertujuan mengisolasi bakteri Escherichia coli pada telur puyuh yang gagal menetas di Desa Garot Kecamatan Darul Imarah, Kabupaten Aceh Besar. Sebanyak 30 butir telur puyuh yang gagal menetas diperiksa di Laboratorium Mikrobiologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala Banda Aceh. Sampel diisolasi berdasarkan metode Carter yang dimodifikasikan. Telur dibuka, diambil embrionya dan diswab dengan swab steril. Selanjutnya dimasukkan dalam media Nutrien Broth (NB) dan inkubasi pada suhu 37ºC selama 24 jam. Kemudian dengan menggunakan ose steril, biakan dipindahkan ke media Eosin Methilen Blue Agar (EMBA). Bakteri yang tumbuh diamati morfologi koloninya dan dilakukan pewarnaan Gram. Hasil penelitian menunjukkan bahwa 3 dari 30 sampel (10%) telur burung puyuh yang diteliti positif terinfeksi Escherichia coli . Oleh sebab itu dapat disimpulkan bahwa Escherichia coli merupakan salah satu penyebab kegagalan menetas pada telur puyuh dipeternakan Desa Garot Kecamatan Darul Imarah, Kabupaten Aceh BesarABSTRACTThis study aimed to isolate Escherichia coli from quail eggs that were not hatched in a farm at Garot village, Darul Imarah subdistrict, Aceh Besar District. Thirty unhatched quail eggs were examined in microbiology laboratorium of Veterinary Faculty of Syiah Kuala University, Banda Aceh. The eggs opened and the embrio were swabbed, and put into Nutrien Broth (NB) and incubated at 37o C for 24 hours. The culture were then transfered onto Eosine Methilen Blue Agar (EMBA). Grown bacteria were observed for it’s colony  morphology and stained with Gram stain. The result showed that 3 out of 30 unhatched samples (10%) were positively infected by Escherichia coli. Based on this study, it can be concluded that Escherichia coli was one of many factors that caused quail eggs failed to hatch in a farm of  Garot village, Darul Imarah subdistrict, Aceh Besar District.

Abstract

ABSTRAKTelur ayam ras merupakan bahan pangan asal hewan yang mengandung protein tinggi dan banyak dikonsumsi oleh masyarakat Indonesia. Kualitas isi telur ayam sangat dipengaruhi oleh suhu dan waktu penyimpanan. Oleh karena itu, penelitian ini dilakukan untuk mengetahui pengaruh suhu dan waktu penyimpanan, terhadap cemaran bakteri Salmonella typhimurium pada telur ayam ras yang telah dipaparkan dengan bakteri Salmonella typhimurium, pada kerabang telur. Penelitian ini menggunakan 15 butir sampel telur ayam yang berkualitas baik. Telur dibagi menjadi 2 kelompok, masing-masing kelompok dipaparkan dengan 0,5 mg (1,5 x108) Salmonella typhimurium dalam suspensi putih telur. Telur pada kelompok pertama disimpan pada suhu ruangan (25OC), dan kelompok kedua disimpan pada suhu refrigerator (4OC). Pengamatan dilakukan pada umur telur 0 hari, 3 hari, 6 hari, 9 hari, 12 hari, 15 hari, 18 hari dan 21 hari. Hasil penelitian yang dilakukan sampai hari ke- 21, tidak terdapat penetrasi bakteri Salmonella typhimurium pada isi telur ayam yang diperiksa. Berdasarkan hasil penelitian ini dapat disimpulkan bahwa, tidak ditemukan adanya penetrasi Salmonella typhimurium ke dalam isi telur ayam baik pada penyimpanan suhu refrigerator dan suhu ruangan selama 21 hari.Kata Kunci : Salmonella typhimurium. suhu, waktu, telur ayam.ABSTRACT                Chicken eggs are food ingredients of animal origin that contain high protein and are widely consumed by the people of Indonesia. The quality of the contents of chicken eggs is strongly influenced by temperature and storage time. Therefore, this study was conducted to determine the effect of temperature and storage time on Salmonella typhimurium on broiler eggs that have been exposed to Salmonella typhimurium bacteria, on egg shells. This study used 15 samples of good quality chicken eggs. Eggs were divided into 2 groups, each group was exposed to 0.5 mg (1.5 x108) Salmonella typhimurium in egg white suspension. Eggs in the first group were stored at room temperature (25OC), and the second group was stored at refrigerator temperature (4OC). Observations were made at the age of 0 days, 3 days, 6 days, 9 days, 12 days, 15 days, 18 days and 21 days. The results of the research carried out until the 21st day, there was no penetration of Salmonella typhimurium on the contents of the chicken eggs examined. Based on the results of this study, it can be concluded that there was no penetration of Salmonella typhimurium into the contents of chicken eggs both at refrigerator temperature and room temperature for 21 days.Keyword : Salmonella typhimurium. temperature, time, chiken egg
