Abstract

ABSTRAK                Penelitian ini bertujuan mengisolasi bakteri Escherichia coli pada telur puyuh yang gagal menetas di Desa Garot Kecamatan Darul Imarah, Kabupaten Aceh Besar. Sebanyak 30 butir telur puyuh yang gagal menetas diperiksa di Laboratorium Mikrobiologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala Banda Aceh. Sampel diisolasi berdasarkan metode Carter yang dimodifikasikan. Telur dibuka, diambil embrionya dan diswab dengan swab steril. Selanjutnya dimasukkan dalam media Nutrien Broth (NB) dan inkubasi pada suhu 37ºC selama 24 jam. Kemudian dengan menggunakan ose steril, biakan dipindahkan ke media Eosin Methilen Blue Agar (EMBA). Bakteri yang tumbuh diamati morfologi koloninya dan dilakukan pewarnaan Gram. Hasil penelitian menunjukkan bahwa 3 dari 30 sampel (10%) telur burung puyuh yang diteliti positif terinfeksi Escherichia coli . Oleh sebab itu dapat disimpulkan bahwa Escherichia coli merupakan salah satu penyebab kegagalan menetas pada telur puyuh dipeternakan Desa Garot Kecamatan Darul Imarah, Kabupaten Aceh BesarABSTRACTThis study aimed to isolate Escherichia coli from quail eggs that were not hatched in a farm at Garot village, Darul Imarah subdistrict, Aceh Besar District. Thirty unhatched quail eggs were examined in microbiology laboratorium of Veterinary Faculty of Syiah Kuala University, Banda Aceh. The eggs opened and the embrio were swabbed, and put into Nutrien Broth (NB) and incubated at 37o C for 24 hours. The culture were then transfered onto Eosine Methilen Blue Agar (EMBA). Grown bacteria were observed for it’s colony  morphology and stained with Gram stain. The result showed that 3 out of 30 unhatched samples (10%) were positively infected by Escherichia coli. Based on this study, it can be concluded that Escherichia coli was one of many factors that caused quail eggs failed to hatch in a farm of  Garot village, Darul Imarah subdistrict, Aceh Besar District.

Abstract

ABSTRAKIkan yang dijual di pasar tidak menutup kemungkinan terkontaminasi oleh bakteri. Penelitian ini bertujuan untuk mendeteksi adanya cemaran bakteri Shigella sp. pada ikan kuniran (Upeneus sulphureus) di pasar Al-Mahira Banda Aceh. Penelitian ini menggunakan metode survei cross-sectional dan pengambilan sampel menggunakan metode sampling jenuh (sensus). Pemeriksaan sampel menggunakan metode Carter. Sampel yang digunakan pada penelitian ini sebanyak 12 ekor ikan, yang diambil dari 4 pedagang ikan di pasar Al-Mahira Banda Aceh. Ikan dibedah secara aseptis dan ususnya diambil kemudian dimasukkan ke dalam media Nutrient Broth (NB) dan diinkubasi pada suhu 37℃ selama 24 jam. Kemudian dilakukan penanaman pada media Salmonella Shigella Agar (SSA) dan diinkubasikan kembali pada suhu 37℃ selama 24 jam. Hasil penanaman pada media SSA menunjukkan bahwa semua sampel positif terkontaminasi Shigella sp. yang ditandai dengan adanya bulat berwarna bening pada koloni yang ditanam. Selanjutnya koloni bakteri yang tumbuh dilakukan pewarnaan Gram dan diperiksa secara mikroskopis. Hasil pewarnaan Gram menunjukkan bakteri yang terlihat berwarna pink/merah dan berbatang pendek, yang menunjukkan bakteri tersebut adalah genus Shigella sp. Dari hasil penelitian dapat disimpulkan bahwa semua sampel yang diperiksa positif tercemar bakteri Shigella sp.ABSTRACTFish sold in the market does not rule out being contaminated by bacteria. This study aims to detect the presence of bacterial contamination Shigella sp. on kuniran fish (Upeneus sulphureus) at Al-Mahira market Banda Aceh. This study used a cross-sectional survey method and sampling using the sampling jenuh method. Samples were examinated using the Carter method. The samples used in study were 12 fish taken from 4 fish traders in the Al-Mahira market Banda Aceh. Fish were dissected aseptically and their intestines were removed and the put in Nutrient Broth (NB) media and incubated at 37 ℃ for 24 hours. Then it was planted on Salmonella Shigella Agar (SSA) media and re-incubated at 37 ℃ for 24 hours. The results of planting on SSA media showed that all positive samples were contaminated with Shigella sp. which was indicated by presence of clear colored circles on the planted column. Futhermore, the column of bacteria that grew was stained with Gram stain and examined microscopically. The result of Gram staining showed that the bacteria were pink/red and had short trunks, which indicated that the bacteria were of genus Shigella sp. The result of the study can be concluded that all samples used were positively contaminated with Shigella sp.
