Abstract

Tembesu (Fragraea fragans Roxb) is important tree species for furniture due to its decorative structure, but its propagation facing to the problems on seed availability. Determining explant sterilization technique of tembesu (Fagraea fragrans Roxb.) in in vitro condition is needed to produce aseptic explants for shoot induction in micropropagation techniques. Shoot induction of explants depends on plant growth regulator and growth medium compositions. This study was aimed (1)to analyzing the effect of sodium hypochlorite 10% in sterilization process to obtain aseptic explants, (2) to find out plant growth regulator and growth medium composition on shoot induction of tembesu (F. fragrans Roxb.) in in vitro condition. This study was conducted at Tissue Culture Laboratory, Rumpin Seed Source and Nursery Center, The Ministry of Forestry, Bogor on February – June 2014. Explants were sterilized using sodium hypochlorite (NaOCl 5.25%) at 10% of concentrations in various durations (5, 10, 15, and 20 minutes). Shoot induction was done using MS medium, MS modified medium, and MS added with 15% coconut water that combined with 1.5 ppm of BAP. The result showed that explants soaked in sodium hypochlorite 10% within 20 minutes was recommended for explants steriliziton technique of tembesu. Combination treatment between BAP 1.5 ppm and MS modified medium was the most favorable culture medium for shoot induction of tembesu.Keyword: Benzylaminopurine, Fagraea fragrans, micropropagation, Murashige-Skoog modified, sodium hypochlorite

Abstract

Sorghum (Sorghum bicolor L Moenc) is plant for food, feed, and energy (bio-ethanol). There were two types of waste from bio-ethanol production, such as liquid and solid waste (baggase). Liquid waste was produced from ethanol distillation process, rich in nutrients as fertilizer, unfortunately limited information on nutritional content and potential function, especially as medium for forestry seedlings production. This experiment examined the effect of the Granular Organic Fertilizer of Sorghum liquid waste to Leda seedling growth in different soil type of Andosol, Latosol, and Podzolic; and to find the dose of Granular Organic Fertilizer for Leda seedling growth in each type of soil. This study was conducted on Factorial in Completely Randomized design. The result showed, the application of Granular Organic Fertilizer effected to almost all the growth parameters on three soil types. Best seedling quality index of leda was found in the combination of Andosol and 20 granules of Granular Organic Fertilizer with seedling quality index 30 points.Key words: andosol, granular organic fertilizer, latosol, podzolic, sorghum.

Abstract

Mutation breeding is a powerful tool to provide novel trait in plants. However their applications in forestry are still limited. Acacia mangium is a major commercial plantation species in Indonesia and other Southeast Asia countries. Therefore, we were trying to improve its productivity. In 2008, 1200 A. mangium seeds had been irradiated by gamma ray 137Cs. There were 98 trees of M1 generations of 7 years old plantations that survive. Most of them are sterile with high clear bole and natural pruning, small branch with good growth angle,and almost straight cylindrical stem form. Based on growth performance, there were at least 29 M1 superior trees. During the observations, unexpected diversity in the bark colours with high and positive correlation to growth performance were discovered. Using microsatellite markers, the use of irradiations techniques provide surplus of heterozygosity with high value of Shannon’s Information Index. Dendrogram showed that almost all of M1 generation formed separate cluster from M0 generation with 0.71 genetic distance .Key words: mutation breeding, Acacia mangium, microsatellite

Abstract

Pine (Pinus merkusii) is tree species that provides timber and gum rosin. To meet the needs of wood and non wood (gum rosin) products, planting by using superior or high quality seeds are needed. Seed procurements for planting are obtained from seedling seed orchard (SSO). However, Cijambu’s SSO was attacked by pine woolly aphid (Pineus boerneri). Therefore, assessment of Cijambu’s SSO needs to be done to evaluate the severity pest attacks that could affect to the quality and the quantity of seed production. Forest Health Monitoring (FHM) method is one of the methods to assess the health level of a stand. The number of trees found in all cluster plots in Cijambu’s SSO were 270 trees. Based on the value of the VCR (Visual Crown Rating), the trees located in all cluster plot have health level between low to high. Based on the value of the VCR showed 38.52% (104 trees) having VCR’s value was high, 49.26% (133 trees) having VCR’s value was middle; 12.22% (33 trees) having VCR’s value was low; and no tree having very low VCR’s value. The average of VCR’s value in all cluster plots were 3.25 and classified as middle health. Based on the value of TDLI (Tree Damage Level Index) from 270 trees in all cluster plot showed that 189 trees (70.00%) in healthy condition; 69 trees (25.56%) in slight damage condition; 11 trees (4.07%) in middle damage condition; and 1 tree (0.37%) in heavy damage condition. The value of damage in all cluster plots (ALI) was 261.22 and classified as in health condition. The trees located in all cluster plots were mostly suitable to be seed sources as 242 trees (89.63%), while 28 trees (10.37%) were not suitable for seed sources.Keywords: Forest Health Monitoring, Pinus merkusii, seedling seed orchard, Tree Damage Level Index, Visual Crown Rating
