Abstract

 This study was conducted to investigate the effect of microencapsulation of lactic acid bacteria (LAB) probiotic isolated from chickens’ gastrointestinal tract on Salmonella enterica serotype enteritidis ATCC 13076 and Escherichia coli EPEC. Probiotic of LAB used were Streptococcus thermophilus strain Kp-2, Lactobacillus murinus strain Ar-3, and Pediococcus acidilactici strain Kd-6. Microencapsulation were conducted by spray drying with inlet/outlet temperatures of 160/80°C using maltodextrin and skim milk powder (20% w/v) as coating materials. Competitive exclusion test was conducted in vitro using well diffusion method. Variable measured in this study was the clear zone observed. The data of clear zone among treatments were analyzed using analysis of variance (ANOVA) one way followed by Duncan multiple range test (DMRT); except the data of clear zone resulted by probiotic before and after microencapsulation that was analyzed using t-test. The result showed that the ability of each strain against pathogen was decreased after being encapsulated. S. thermophilus before and after microencapsulation had the same antagonistic ability against E. coli and S. enteritidis (P>0.05). Microencapsulation process with spray drying method decreased antagonistic ability of probiotic against pathogenic bacteria both in single and multi strain.

Abstract

The aim of this study was to determine the effect of the supplementation of dl-methionine and l-lysine HCl into feed based on cafeteria standards to intestinal morphology of native chickens at 20 weeks-old. A total of 128 fourteen week-old native chickens were used in this study. The native chickens were divided into 4 treatments groups with 4 replications. The dietary treatments were: T0 (feed based on cafeteria standard), T1 (feed based on protein standard of NRC), T2 (cafeteria feed+0.09% dl-methionine+0.22% l-lysine HCl), T3 (cafeteria feed+0.19% dl-methionine+0.42% l-lysine HCl). The data collected were villus height, crypt depth and villus width of duodenum, jejunum and ileum of native chickens aged 20 weeks. The result of this study for T0, T1, T2 and T3 showed that villus height of duodenum were 863.03; 1237.85; 1171.60 and 1261.15 µm, crypt depth 212.70; 192.08; 210.25 and 251.85 µm, villus width 168.74; 168.53; 213.61 and 188.60 µm. Villus height of jejunum were 1381.48; 1088.70; 1072.68 and 1383.15 µm, crypt depth 272.,05; 261.83; 225.00 and 205.65 µm, villus width 188.28; 166.41; 216.26 and 182.95 µm. villus height of ileum were 798.03; 863.35; 882.88 and 828.70 µm, crypt depth 199.58; 199.58; 154.68 and 145.78 µm and villus width 181.13; 154.55; 201.25 dan 175.65 µm, respectively. It can be concluded that supplementation of dl-methionine and l-lysine HCl was not effected upon villus height, crypt depth and villus width of duodenum, jejunum and ileum of native chickens aged 20 weeks.

Abstract

ABSTRACT. Penelitian ini bertujuan untuk melihat aktivitas antibakteri dari bakteri asam laktat (BAL) yang sudah diisolasi dari saluran pencernaan itik lokal asal Aceh. Berdasarkan hasil identifikasi gen 16S rRNA menggunakan metode PCR dan Sekuensing diperoleh lima BAL yaitu Lactobacillus plantarum strain C1; Pediococcus acidilactici strain V2; Lactobacillus fermentum Strain D3; Lactobacillus fermentum Strain I1; Lactobacillus fermentum Strain S4. Penelitian ini mengukur kemampuan aktivitas lima BAL terhadap Salmonella pullorum dan Escherichia coli yang dilakukan dengan difusi (teknik sumuran). Uji ketahanan terhadap kondisi asam (pH 2) dan ketahanan terhadap garam empedu (bile salt) dilakukan dengan metode Total Plate Count (TPC) menggunakan medium agar MRS metode pour plate. Hasil penelitian diperoleh bahwa Pediococcus acidilactici strain V2 memiliki aktivitas antimikroba terbaik terhadap bakteri Salmonella pullorum dan Escherichia coli. Pediococcus acidilactici strain V2 juga memberikan hasil terbaik pada ketahanan terhadap pH 2 dan garam empedu 1%.(Antibacterial activity of LAB isolated from the digestive tract of a native Aceh duck of Salmonella Pullorum and Escherichia coli)ABSTRAK. This study aimed to examine the antibacterial activity of lactic acid bacteria isolated from digestive tract of Local ducks from Aceh. Identification of the 16S rRNA gene using PCR and sequencing methods were obtained five LAB is Lactobacillus plantarum strain C1; Pediococcus acidilactici strain V2; Lactobacillus fermentum Strain D3; Lactobacillus fermentum Strain I1; and Lactobacillus fermentum Strain S4. This study measure the ability five BAL activity test against Salmonella pullorum and Escherichia coli by diffusion (well technique). Test of resistance to acid conditions (pH2) and to bile salt were using the Total Plate Count (TPC) method with MRS Agar medium by pour plate. The results showed that Pediococcus acidilactici strain V2 had the best antimicrobial activity against Salmonella pullorum and Escherichia coli. Pediococcus acidilactici strain V2 also gave the best results on resistance to pH 2 and 1% bile salt.
