Abstract

The objective of this research was to examine the effectiveness of coconut water with various egg yolk concentrations in maintaining the viability of epididymal spermatozoa of crossbreed cattle preserved at 5oC. Five testis with epididymides of crossbreed cattle were obtained from slaughterhouse. Epididymal spermatozoa was collected by the combination of slicing, flushing and tissues pressure methods of cauda epididymides with saline solution (0.9% NaCl). Collected-spermatozoa was equally divided in volume into four tubes and diluted with lactose extender containing 20% egg yolk (control), 90% coconut water + 10% egg yolk (CWEY10), 85% coconut water + 15% egg yolk (CWEY15), and 80% coconut water + 20% egg yolk (CWEY20), repectively. Diluted-spermatozoa was stored in refrigerator at 5oC. Quality of dilutedspermatozoa including percentages of motile spermatozoa (MS), live spermatozoa (LS), spermatozoa with intact plasma membrane (IPM) were evaluated every day during four days of storage. Data were analyzed by using completely randomized design with four treatments and five replicates. Means were compared with significant difference test at 0.05 significant level. Results of this study showed that mean of spermatozoa concentration, percentage of MS, percentage of LS, percentage of spermatozoa abnormal, and percentage of IPM of crossbreed cattle fresh epididymal spermatozoa were 1,414 million cell/ml, 72%, 85%, 9%, and 90%, respectively. At day-4 of the storage, percentages of MS, LS, and IPM of control (43, 52.2, 59.2%) and CWEY20 (42, 52, 59%) were significantly (P<0.05) higher than CWEY10 (33, 45.4, 52.8%) and CWEY15 (37, 50, 54.6%). In conclusion, lactose and CWEY20 extenders could be maintaining the quality of epidydimal spermatozoa of crossbreed cattle for three days preservation at 5oC and two days for CWEY10 and CWEY15.

Abstract

The dependence on imports of feed ingredients for ration composer are increasingly expensive and availability of limited and unsustainable local feed, causing the low level of production and reproduction of local Indonesian cattle. This study aims to exploit the potential of plantation waste as an alternative feed of beef cattle, increase the nutrient value and digestibility of waste of palms and leaves and to know the optimal use of inoculums (Trichoderma sp.) through fermentation in improving the digestibility and nutritional value of the feed. The research method used was a complete randomized design (RAL) with five treatments and three replications, with 14 days fermentation. The research treatment includes: PD 0 (without Trichoderma/control), PD 1 (fermentation using Trichoderma viride 3 ml), PD 2 (fermentation using Trichoderma viride 6 ml), PD 3 (fermentation using Trichoderma harzianum 3 ml), PD 4 (fermentation using Trichoderma harzianum 6 ml).O Observation parameters observed included dry matter (DM), crude protein (CP), ash content (AC), coarse fat (CF), organic matter (OM), and total digestible nutrient (TDN). The data obtained were analyzed using variance analysis. The results showed that the best treatment was found in PD 1, had a significant effect on control in increasing total digestible nutrient (TDN) 51.62% and crude fiber decline by 39.14%, although an increase in CP (5.34%) was not equal to the treatment of the PD 4 (5.35%). While the best increase of CP content was found in the treatment of PD 4 of 3.35%. While the best CP content found in the treatment of PD 4 of 5.35%. It was concluded that the use of Trichoderma viride and Trichoderma harzianum can improve the quality of waste nutrient and palm oil leaves by fermentation and the optimal inoculums dose used to produce the best fermentation is the use of Trichoderma sp. 3 ml in 3kg of material.

Abstract

Pengabdian kepada Masyarakat (PKM) ini bertujuan untuk meningkatkan pengetahuan serta keterampilan petani di Kecamatan Sambung Makmur Kabupaten Banjar dalam membuat pupuk organik berbahan kotoran sapi. Khalayak sasaran PKM berupa penyuluhan teknik pembuatan pupuk organik berbahan kotoran sapi dalam rangka menambah ilmu pengetahuan dan keterampilan bagi petani di Kecamatan Sambung Makmur Kabupaten Banjar. Kegiatan PKM dilaksanakan pada 14 Juni 2023, bertempat di BPP Sambung Makmur, Kabupaten Banjar dengan jumlah peserta sebanyak 25 orang yang terdiri dari anggota kelompok tani dan karang taruna di wilayah Kecamatan Sambung Makmur Kabupaten Banjar. Hasil PKM menunjukkan peningkatan pengetahuan dan keterampilan petani di Kecamatan Sambung Makmur Kabupaten Banjar mengenai pembuatan pupuk organik berbahan kotoran sapi. Sebanyak 100% peserta mengetahui apa itu pupuk organik, 100% peserta mengetahui jenis-jenis pupuk organik, 100% peserta mengetahui apa itu pupuk organik kotoran sapi, 100% peserta mengetahui manfaat pupuk organik bagi tanah dan tanaman, 96% peserta mengetahui keunggulan pupuk organik dibanding pupuk kimia, 100% peserta mengetahui bahan-bahan yang digunakan dalam pembuatan pupuk organik kotoran sapi, 100% peserta mengetahui kotoran ternak dapat digunakan sebagai bahan baku pembuatan pupuk organik kotoran sapi, 94% peserta mengetahui proses pembuatan pupuk organik kotoran sapi secara baik dan benar, 88% peserta mengetahui ciri-ciri pupuk organik kotoran sapi yang telah selesai di fermentasi, dan 100% peserta mengetahui cara penggunaan pupuk organik kotoran sapi ke tanaman. Training on Making Organic Fertilizer Made from Cow Manure for Farmers in Sambung Makmur District, Banjar Regency This community service aims to increase the knowledge and skills of farmers in Sambung Makmur District, Banjar Regency, in making organic fertilizer from cow manure. Community service's target audience is in the form of providing information on techniques for making organic fertilizer from cow manure in order to increase knowledge and skills for farmers in Sambung Makmur District, Banjar Regency. The community service activity was carried out on June 14, 2023, at the Sambung Makmur Agricultural Extension Center, Banjar Regency, with a total of 25 participants consisting of members of farmer groups and youth organizations in the Sambung Makmur District, Banjar Regency. The community service results show an increase in the knowledge and skills of farmers in Sambung Makmur District, Banjar Regency, regarding making organic fertilizer from cow manure. As many as 100% of participants know what organic fertilizer is; 100% of participants know the types of organic fertilizer; 100% of participants know what organic cow manure fertilizer is; 100% of participants know the benefits of organic fertilizer for soil and plants; 96% of participants know the advantages of organic fertilizer compared to chemical fertilizers; 100% of participants know the ingredients used in making organic cow manure fertilizer; 100% of participants know that livestock manure can be used as raw material for making organic cow manure fertilizer; and 94% of participants know the process of making organic cow manure fertilizer properly. That's right, 88% of participants know the characteristics of organic cow manure fertilizer that has been fermented, and 100% of participants know how to use organic cow manure fertilizer on plants.

Abstract

Air tebu dapat dimanfaatkan sebagai pengencer semen karena mengandung berbagai nutrien yang dapat menunjang kehidupan spermatozoa selama preservasi. Penelitian ini bertujuan untuk menguji kemampuan air tebu dalam mempertahankan daya hidup spermatozoa kambing PE selama preservasi pada suhu 3-5oC. Semen ditampung menggunakan vagina buatan dan segera dievaluasi secara makroskopis dan mikroskopis. Semen segar dibagi ke dalam lima buah tabung reaksi kemudain masing-masing diencerkan dengan 80% pengencer dasar laktosa + 20% kuning telur sebagai kontrol (P0), 20% air tebu + 60% akuabidetislata + 20% kuning telur (P1), 30% air tebu + 50% akuabidestilata + 20% kuning telur (P2), 40% air tebu + 40% akuabidestilata + 20% kuning telur (P3), dan 50% air tebu + 30% akuabidestilata + 20% kuning telur (P4). Semen yang telah diencerkan disimpan di dalam refrigerator lemari es pada suhu 3-5oC. Motilitas dan daya hidup spermatozoa masing-masing perlakuan dievaluasi setiap hari hingga motilitas mencapai 40%. Hasil penelitian diperoleh karakteristik semen segar kambing PE yaitu volume 1,15 ml, konsentrasi 3.870 juta/ml, motilitas 80%, spermatozoa hidup 87,75%, dan spermatozoa abnormal 7,5%. Persentase motilitas dan spermatozoa hidup pada hari keempat preservasi perlakuan P2 (41,25% dan 50%) nyata (P<0,05) lebih tinggi dibandingkan dengan P0 (37,5% dan 47%), P1 (32,5% dan 44%), P3 (30% dan 36,25%), dan P4 (18% dan 29,25%). Dapat disimpulkan bahwa Kombinasi pengencer terbaik untuk semen kambing PE adalah terdiri atas 30% air tebu + 50% akuabidestilata + 20% kuning telur. ABSTRACT Sugarcane juice can be used as an alternative semen extender because it contains various nutrients that can support the life of spermatozoa during preservation. This study aimed to examine the ability of sugarcane juice to maintain the ability of Etawah crossbreed goat spermatozoa during preservation at 3-5oC. Semen was collected using an artificial vagina and immediately evaluated macroscopically and microscopically. Fresh semen was divided into five tubes and diluted with 80% lactose-based extender + 20% egg yolk as a control (T0), 20% sugarcane juice + 60% aquabidestilata + 20% egg yolk (T1), 30% sugarcane juice + 50% aquabidestilata + 20% egg yolk (T2), 40% sugarcane juice + 40% aquabidestilata + 20% egg yolk (T3), and 50% sugarcane juice + 30% aquabidestilata + 20% egg yolk (T4), respectively. The diluted-semen was stored in the refrigerator at 3-5oC. The motility and viability of spermatozoa were evaluated every day until the motility reached 40%. The results showed that the characteristics of fresh semen of Etawah crossbreed goat were volume 1.15 ml volume, spermatozoa concentration 3870 million cells/ml, spermatozoa motility 80%, live spermatozoa 87.75%, and abnormal spermatozoa 7.5%. The percentage of motility and live spermatozoa on the fourth day of preservation for T2 (41.25% and 50%) was significantly (P<0.05) higher than T0 (37.5% and 47%), T1 (32.5% and 44%), T3 (30% and 36.25%), and T4 (18% and 29.25%). It can be concluded that the best extender combination for Etawah crossbreed goat semen is composed of 30% sugarcane juice + 50% aquabidestilata + 20% egg yolk.
