Abstract

Penelitian bertujuan untuk mengetahui viabilitas spermatozoa cauda epididimis pada berbagai konsentrasi pengenceran menggunakan air kelapa muda dan kuning telur yang disimpan pada  temperatur 3-5oC.  Pengencer Tris 80% + 20% kuning telur digunakan sebagai kontrol (P1), selanjutnya perlakuan menggunakan pengencer air kelapa muda 90% + 10% kuning telur (P2), air kelapa muda 85% + 15% kuning telur (P3) dan air kelapa muda 80% + 20% kuning telur (P4). Viabilitas  spermatozoa yang diamati setiap hari meliputi persentase motilitas dan persentase hidup sampai mencapai motilitas minimum 30%. Hasil penelitian menunjukkan persentase motilitas tertinggi pada hari ke-4 terdapat pada perlakuan P4 sebesar 31.8 %, berbeda nyata (P<0.05) dengan P3 sebesar 10.5 % dan P2 sebesar 2.5 % serta dengan P1  sebesar 4.2 %.  Pada persentase hidup spermatozoa, perlakuan P4 juga menunjukan nilai 50.3 %, berbeda sangat nyata (P<0.05) dengan perlakuan P1, P2 serta P3 berturut-turut adalah 27.8 %, 26.4 %, dan 32.9 %.  Kesimpulan dari penelitian menunjukkan bahwa pengenceran dengan perlakuan P4 (80% air kelapa dan 20% kuning telur) mampu mempertahankan viabilitas spermatozoa cauda epididimis kerbau rawa selama proses penyimpanan hingga empat hari di dalam refrigerator dengan suhu 3–5oC.

Abstract

The purpose of this research was evaluate the quality of spermatozoa concentration in the caput, corpus and cauda of the swamp buffalo epididymis (Bubalus bubalis carabanensis).  Method of this research was to exploration to 13 epididymides of eight swamp buffaloes were obtained from Banjar and Banjarmasin slaughterhouses,evaluated the quality of spermatozoa in caput, corpus and cauda of epididymis.  Quality of collected-spermatozoa including spermatozoa motility, percentage of live spermatozoa, spermatozoa concentration and percentage of abnormality.  Result of this study showed that mean of each of caput spermatozoa motility, percentage of live spermatozoa, spermatozoa concentration and percentage of abnormality; 0%, 45.43% (31.87–72%), 189,62 x106 (40–480 x106) and 56.16 %(44.34–66.53%), corpus ;2.77% (1–9%), 58.73% (45.14 –76%), 152.31 x106 (45 – 345x106), and 47.61 %(23.92 – 60.15%), cauda;53.46% (20 – 70%), 74.32 % (56.68 – 83%), 1,459.62 x106 (825 – 2,340x106), and 34.60%(15.89 –50.04%). In conclusion, spermatozoaofcaudaepididymis could be used in artificial insemination program.Keywords: Spermatozoa, epididymis, swamp buffalo.

Abstract

The purpose of this research was to examine the effectivity of Tris, sodium citrate, and lactose extenders in maintaining viability of etawa crossbreed (EC) goat spermatozoa preserved at 3–5oC.  Semen was collected using artificial vagina once a week for six times as replicate.  Fresh semen was divided into three tubes then diluted with Tris, sodium citrate, and lactose extenders, respectively.  Diluted-semen were stored in refrigerator at 3–5oC.  Quality of diluted-semen including percentages of motile spermatozoa (MS) and live spermatozoa (LS) were evaluated every day during storage at 3–5oC for four days.  Data were analyzed using completely randomized design with three treatments and six replicates.  Results of this study showed that mean volume, colour, consistency, pH, mass activity, spermatozoa concentration, MS, LS, and spermatozoa abnormal of PE goat fresh semen were 1.05 ml, cream, thick, 6.9, ++ – +++, 3,756.67 million cell/ml, 76.67%, 85.83%, and 4.5%, respectively.  Tris, sodium citrate, and lactose extenders have same ability in maintaining viability of EC goat spermatozoa during preserved at 3–5oC.  At day-4 storage, percentages of MS and LS for Tris (44.17 and 57.83%) was no difference significant with sodium citrate (43.33 and 56.67%), and lactose (43.33 and 54.17%).   In conclusion, Tris, sodium citrate, and lactose extenders have same ability in maintaining viability of EC goat spermatozoa during preserved at 3–5oC.  Tris, sodium citrate, and lactose extenders could be maintaining 40% in percentage of motile spermatozoa for three days after preserved at 3–5oC, and suitable for use in artificial insemination (AI) programKeywords: Tris, sodium citrate, lactose, spermatozoa viability, PE goat.

Abstract

The objective of this study was to examine the effect of seminal plasma on viability of Boer goat spermatozoa and effectiveness of sugar palm juice as an alternative extender during preservation at 5ºC. Semen of two Boer goats were collected using an artificial vagina. Fresh semen were evaluated and divided in equal volume into four tubes. Semen in the first and second tubes diluted with 80% sugar palm juice + 20% egg yolk (P1) and Andromed (P2), respectively. Semen in the third and fourth tubes were centrifuged with 3,000 RPM for 20 minutes, and the supernatant removed. diluted with 80% sugar palm juice + 20% egg yolk (P3) and Andromed (P4), respectively. Diluted-semen were preserved in refrigerator at 5oC, and quality of the spermatozoa including motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) were evaluated every day for four days. Results of this study showed that at day-2 preservation, mean percentages of MS, LS, and IPM for P2 (72, 83.4, and 83.4%), P3 (72, 82.6, and 82.2%), and P4 (72, 83, 83.8%) were significantly (P<0.05) higher than P1 (3, 24.8, and 25.2%). At day-3 preservation, mean percentages of MS, LS, and IPM for P2 (57, 65.6, and 69.6%) was significantly (P<0.05) higher than P3 (21, 34.8, and 31.8%), P4 (22, 33.6, and 31.2%), and P1 (0, 0, and 0%). In conclusion, semen of Boer goat to be preserved with extender containing egg yolk should be removed seminal plasma. Sugar palm juice containing egg yolk could be used as an extender for Boer goat semen, but should be applied in the AI program immediately after the semen is diluted.

Abstract

The objective of this research was to examine the effectiveness of coconut water with various egg yolk concentrations in maintaining the viability of epididymal spermatozoa of crossbreed cattle preserved at 5oC. Five testis with epididymides of crossbreed cattle were obtained from slaughterhouse. Epididymal spermatozoa was collected by the combination of slicing, flushing and tissues pressure methods of cauda epididymides with saline solution (0.9% NaCl). Collected-spermatozoa was equally divided in volume into four tubes and diluted with lactose extender containing 20% egg yolk (control), 90% coconut water + 10% egg yolk (CWEY10), 85% coconut water + 15% egg yolk (CWEY15), and 80% coconut water + 20% egg yolk (CWEY20), repectively. Diluted-spermatozoa was stored in refrigerator at 5oC. Quality of dilutedspermatozoa including percentages of motile spermatozoa (MS), live spermatozoa (LS), spermatozoa with intact plasma membrane (IPM) were evaluated every day during four days of storage. Data were analyzed by using completely randomized design with four treatments and five replicates. Means were compared with significant difference test at 0.05 significant level. Results of this study showed that mean of spermatozoa concentration, percentage of MS, percentage of LS, percentage of spermatozoa abnormal, and percentage of IPM of crossbreed cattle fresh epididymal spermatozoa were 1,414 million cell/ml, 72%, 85%, 9%, and 90%, respectively. At day-4 of the storage, percentages of MS, LS, and IPM of control (43, 52.2, 59.2%) and CWEY20 (42, 52, 59%) were significantly (P<0.05) higher than CWEY10 (33, 45.4, 52.8%) and CWEY15 (37, 50, 54.6%). In conclusion, lactose and CWEY20 extenders could be maintaining the quality of epidydimal spermatozoa of crossbreed cattle for three days preservation at 5oC and two days for CWEY10 and CWEY15.

Abstract

The objective of this study was to examine the effectiveness of fetal bovine serum (FBS) against thequality of garut ram frozen semen. Semen was collected from one mature garut ram using artificial vagina.Fresh semen were evaluated then divided into four tubes at equal volume and each tube were diluted withTris extender containing 20% egg yolk (TEY-20, as control); TEY-20 + 8% FBS (FBS-8); TEY-20 + 10% FBS(FBS-10); and TEY-20 + 12% FBS (FBS-12), respectively. Semen at the concentration of 100x106 motilespermatozoa was loaded in 0.25 ml mini straw. Semen was equilibrated at 50C for three hours, then freezeand stored in liquid nitrogen container. The quality of the spermatozoa including percentages of motileand live spermatozoa, intact plasma membrane (IPM) were evaluated following diluting, equilibratingand thawing process. A Complete Randomized design using four treatments and five replicates were usedin the study. The results showed that there was no significant difference (p<0.05) in percentage of motilespermatozoa following thawing between the control (44.0%) and FBS-8 (46.0%), FBS-10 (48.0%), andFBS-12 (47.0%), respectively. The percentage of live spermatozoa and IPM were significantly higher (p<0.05)in the FBS-8 (69.0% and 58.2%); FBS-10 (72.4% and 61.2%); FBS-12 (72.2% and 64.4%) compared to thecontrol (64.8% and 52.8%), respectively. In conclusion, the addition of FBS into Tris extender was able tomaintain the viability and integrity of plasma membrane of garut ram frozen semen.

Abstract

Daun kelor mengandung berbagai zat nutrien yang dapat melindungi spermatozoa selama kriopreservasi semen. Tujuan penelitian ini adalah untuk menguji pengaruh ekstrak daun kelor di dalam pengencer tris kuning telur terhadap kualitas semen beku kambing peranakan Boer. Semen segar dibagi ke dalam empat buah tabung reaksi dengan volume yang sama, kemudian disentrifugasi dengan kecepatan 3.000 RPM selama 20 menit. Plasma semen dibuang, dan sedimen (spermatozoa) diencerkan dengan empat pengencer berbeda sebagai perlakuan. Penelitian menggunakan rancangan acak lengkap dengan empat perlakuan, yakni: 73% pengencer dasar tris + 20% kuning telur + 7% gliserol tanpa ekstrak daun kelor (kontrol), 71% pengencer dasar tris + 20% kuning telur + 7% gliserol + 2% ekstrak daun kelor (EDK-2), 69% pengencer dasar tris + 20% kuning telur + 7% gliserol + 4% ekstrak daun kelor (EDK-4), dan 67%pengencer dasar tris + 20% kuning telur + 7% gliserol + 6% ekstrak daun kelor (EDK-6). Semen dikemas di dalam straw mini, diekuilibrasi di dalam lemari es pada suhu 5oC selama empat jam dan dibekukan dengan cara meletakkan straw 10 cm di atas permukaan nitrogen cair selama 15 menit. Peubah yang diamati adalah persentase motilitas, persentase hidup, dan persentase membran plasma utuh (MPU) spermatozoa dievaluasi setelah pengenceran dan thawing. Hasil penelitian menunjukkan bahwa penambahan ekstrak daun kelor ke dalam pengencer tris kuning telur dapat mempertahankan kualitas spermatozoa semen beku kambing peranakan boer. Penambahan sebanyak 4% ekstrak daun kelor merupakan konsentrasi terbaik dalam menghasilkan semen beku kambing peranakan boer. Persentase motilitas spermatozoa setelah thawing pada perlakuan kontrol, EDK-2, EDK-4, dan EDK-6 masing-masing adalah 41,25%, 45%, 50%, dan 47,5%. Dapat disimpulkan bahwa penambahan 4% ekstrak daun kelor ke dalam pengencer tris kuning telur merupakan konsentrasi terbaik dalam menghasilkan semen beku kambing peranakan boer.

Abstract

Sugar palm juice (Arenga pinnata Merr.) can be used as semen extender because its various nutrientscontent which were needed for spermatozoa preservation. The objective of this research was to examineeffectivity of sugar palm juice as swamp buffalo semen extender in artificial insemination (AI) program.Semen of swamp buffalo were collected by using an artificial vagina. Fresh semen were evaluated anddivided in equal volume into four tubes and diluted with lactose extender containing 20% egg yolk (lactoseor control), 85% sugar palm juice + 15% egg yolk (PJEY15), 80% sugar palm juice + 20% egg yolk (PJEY20),and 75% sugar palm juice + 25% egg yolk (PJEY25), respectively. Diluted-semen were preserved inrefrigerator at 5oC, and quality of spermatozoa including percentages of motile spermatozoa, livespermatozoa, and intact plasma membrane (IPM) were evaluated every day for four days. A total of 13female swamp buffaloes were estrous synchronize with PGF2a, and six buffaloes were inseminated withlactose and seven buffaloes were inseminated with PJEY20. Results of this research showed that at dayfourpreservation, mean percentages of motile and live spermatozoa for lactose (36.67 and 51.83%) andPJEY20 (35.83 and 51.5%) were significantly (P<0.05) higher than PJEY15 (31.67 and 44%) and PJEY25 (30 and 45.33%), but no difference significant between treatment for IPM parameter. Pregnancy rate forlactose was 66.67% and 71.43% for PJEY20. Percentage of birth for lactose and PJEY20 were 66.67% and71.43%, respectively. In conclusion, sugar palm juice could be used as an alternative extender in semenpreservation process and AI of swamp buffalo.

Abstract

Ethylene diamine tetraacetic acid (EDTA) is a molecular structure that binds to heavy metals andother toxins which needed to protect the spermatozoa during the cryopreservation process. The objectiveof this research was to examine EDTA in maintain the quality of garut ram frozen semen. Semen werecollected from mature Garut ram by artificial vagina. Fresh semen were evaluated and divided in equalvolume into three tubes and diluted with Tris extender containing 20% egg yolk (TEY-20) as control, TEY-20 + 0.01% EDTA (EDTA0.01), and TEY-20 + 0.02% EDTA (EDTA0.02), respectively. Semen was loaded in 0.25 mL mini straw with the concentration of 100 million motile spermatozoa. Semen was equilibrated at5oC for three hours, then frozen and stored in liquid nitrogen container. Quality of spermatozoa includingpercentages of motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) wereevaluated after diluting, equilibrating, and thawing. Data were analyzed using completely randomizeddesign with three treatments and five replicates. Means were compared significant difference test at 0.05significant level. Results of this research showed that mean volume, color, consistency, pH, mass movement,spermatozoa concentration, percentage of MS, percentage of LS, percentage of abnormal spermatozoa,and percentage of IPM of garut ram fresh semen were 0.87 mL, cream, thick, 6.98, 3, 4,296 million cell/mL,75%, 86.33%, 4.4%, and 85%, respectively. Percentages of MS, LS, and IPM after thawing in control (42.5,52.5, and 52.17%) was not significantly difference with EDTA0.01 (44.17, 52.5, and 52.5%) and EDTA0.02(39.17%, 51%, and 48.33%). In conclusion, the addition of EDTA in Tris extender can maintain quality ofGarut ram frozen semen, and its suitable using in artificial insemination program. Addition of EDTA cannot enhancing quality of garut ram frozen semen.

Abstract

Biogas is a renewable energy source needed as an alternative potential fuel.  This research aims to 1) analyse perceptions and participations of community to the development of biogas in Kecamatan Tamban Catur, Kabupaten Kapuas, 2) identify factors influencing the success of biogas development in Kecamatan Tamban Catur, Kabupaten Kapuas, and 3) analyse the benefits of using biogas for community in Kecamatan Tamban Catur, Kabupaten Kapuas.  The research was carried out in Sidomulyo, Sidorejo, and Warnasari Villages in Kecamatan Tamban Catur, Kapuas Regency. The research was conducted using survey data, and the data were further analyzed through descriptive statistics involving 33 respondents who have received the program of biogas development in Kecamatan Tamban Catur Kapuas Regency.  The dependent variables were perceptions and participations of community while the independent variables were ages, the number of cattle, education, supporting program, and duration of biogas utilization. The results indicated that community perceptions of the biogas development were positively increasing (78.8%). The F-Test showed that independent variables influencing community perceptions significantly (P<0.05) with 35.1% of coefficient determination (R2).  Although community participations to biogas development were relatively high (72.7%), there was no apparent impact (P>0.05) of independent variables to the community participations. Factors influencing the success of biogas development in Kecamatan Tamban Catur were passion, the number of cattle, floor types, applicable technology, and supporting programs from the government agencies.  There are several advantages of using biogas for the community. First, the use of biogas improves cost efficiency per month by reducing the usage of fuel/kerosene for about 15 liters (Rp.150.000,-) and the usage of 60 bundles of fire-wood (Rp.120.000,-).  Moreover, utilizing solid and liquid organic fertilizer increased daily income for Rp. 10.000,- and Rp. 48.000,- respectively.  In addition, biogas application is more environmentally-friendly.
