Abstract

Diabetes mellitus (DM) is a disease that can be identified by high levels of blood glucose. Garcinia plants have been widely used for many traditional medicines as antioxidant, anticancer, antidiabetics, and antiinflammation. The antioxidant and antidiabetic activities of (Garcinia picrorrhiza Miq.) or sesoot fruit extract were evaluated in this study and compared with xanthone. The antioxidant and antidiabetic of ethanolic ripe sesoot (G. picrorrhiza Miq.) fruit extract (GpKar) was evaluated by (ABTS) reducing activity, α-glucosidase, β-glucosidase, and α-amylase inhibitor activity. GpKar showed higher ABTS-reducing activity (IC50 = 49.30 µg/mL) than xanthone (IC50 = 404.30 µg/mL). GpKar showed IC50 = 109.32 µg/mL for α-glucosidase inhibitory activity, while xanthones had a better activity (IC50 = 33.97 µg/mL). GpKar also showed lower α-amylase inhibitory activity and  β-glucosidase (IC50 = 126.01 and 9432.09 µg/mL) compared to xanthone (IC50 = 44.32 and 405.03 µg/mL, respectively). The compounds of GpKar are proven to have antioxidant and antidiabetic activities. Therefore, it will be industrially relevant to develop a natural medicine for decreasing DM risk, thus evaluating the antioxidant and antidiabetic effect of G. picrorrhiza by a pre-clinic study is needed.

Abstract

Oxidative stress has been known to contribute to Alzheimer’s disease. Acetylcholinesterase (AChE) enzyme may lead to Alzheimer’s disease as a neurotransmitter. Antioxidants may have protective activities against oxidative damage and Alzheimer’s disease. Acetylcholinesterase inhibitors also can be used in the treatment of various neurological disorders for management of Alzheimer’s disease. This study aimed to determine antioxidant and anticholinesterase effects of Garcinia picrorrhiza Miq. fruit extract (GpKar) and its compounds, xanthone. Antioxidant activity was measured by H2O2 scavenging inhibitory activity, while anticholinesterase activity was measured using modified Ellman method. GpKar has higher H2O2 scavenging inhibitory activity (IC50= 967.28 µg/ml) compared to xanthone (IC50= 1198.95 µg/ml). In the anticholinesterase inhibitory activity, GpKar has lower activity (IC50= 70.25 µg/ml) compared to xanthone (11.80 µg/ml). In summary, GpKar has higher antioxidant activity but lower anticholinesterase activity compared to its compounds, xanthone. However, GpKar has potency as antioxidant agent to prevent Alzheimer’s disease.

Abstract

A myokine is one of the proteins that are produced and released by myocytes in response to muscular contractions when doing physical exercise. One protein that is thought to function as myokine is FGF23.The purpose of this study was to determine the effect of acute physical exercise with moderate intensity on the expression of FGF23 gene in Wistar rat heart. This was an animal experimental study using 24 male Wistar rats that were divided into 4 groups:treatment groups that performed 30 minute acutephysical exercise with moderate intensity (20 m/min) for 3 days, 6 days, and 15 days and a control group without physical exercise. The study was conducted in the Animal Laboratory and Central Laboratory of Universitas Padjadjaran during the period of February to July 2019. Data observed were the FGF23 gene expressions in Wistar rats heart. Data were analyzed using Kruskal-Wallis and Mann-Whitney tests. The results from the Kruskal-Wallis test showed that acute physical exercise with moderate intensity did not increase the FGF23 gene expression in Wistar rat heart (p>0.05), and the average of relative ratios of FGF23/GAPDH gene expression were as follows: control (0.970±0,03), 3 days (0.992±0.03), 6 days (1.014±0.05), and 15 days (1.056±0.02). GAPDH was used in this study as a housekeeping gene since its expression is very constant. This study proves that FGF23 is more likely to take a role in the cardiac remodeling process, especially those associated with cardiac hypertrophy after chronic exercise with no effect observed after acute physical exercise with moderate intensity in Wistar rat heart. Pengaruh Latihan Fisik Akut Intensitas Sedang terhadap Ekspresi Gen FGF23 pada Jantung Tikus Galur WistarMiokin diproduksi dan dilepaskan oleh miosit sebagai respons terhadap latihan fisik. Salah satu protein yang diduga berfungsi sebagai miokin adalah FGF23. Penelitian ini bertujuan mengetahui pengaruh latihan fisik akut intensitas sedang terhadap ekspresi gen FGF23 pada jantung tikus galur Wistar. Penelitian ini menggunakan desain eksperimental dengan tikus galur Wistar jantan yang berjumlah 24 tikus. Tikus dibagi menjadi 4 kelompok untuk diberi perlakuan latihan fisik intensitas sedang (20 meter/menit) dengan durasi 30 menit, selama 3 hari, 6 hari, dan 15 hari serta kelompok tanpa latihan fisik. Penelitian ini dilakukan di Lab Hewan dan Lab Sentral Universitas Padjadjaran pada bulan Februari hingga Juli 2019. Ekspresi gen FGF23 pada jantung tikus galur Wistar dilihat dengan PCR. Analisis data menggunakan uji Kruskal-Wallis dan Mann-Whitney. Uji statistik tidak mendapatkan peningkatan ekspresi gen FGF23 setelah dilakukan latihan fisik akut intensitas sedang selama 3 hari, 6 hari dan 15 hari (p>0,05) dan didapatkan rasio relatif ekspresi gen FGF23/GAPDH otot jantung pada : kontrol (0,970±0,03), hari 3 (0,992±0,03), hari 6 (1,014±0,05), hari 15 (1,056±0,02). GAPDH digunakan dalam studi ini sebagai gen ‘housekeping’ karena ekspresinya yang sangat konstan. Studi ini membuktikan bahwa FGF23 mungkin lebih berperan dalam proses remodeling jantung, terutama yang berhubungan dengan hipertrofi jantung setelah latihan fisik kronik. Simpulan, tidak terdapat pengaruh latihan fisik akut intensitas sedang terhadap ekspresi gen FGF23 pada jantung tikus galur Wistar.

Abstract

Free radicals in the body will increase with excessive of ultraviolet (UV) light exposure, induce oxidative stress with the formation of Reactive Oxygen Species (ROS). A visible effect on skin tissue known as photoaging, including the process of melanogenesis catalyzed by the tyrosinase enzyme, risked for pigmentation or melanoma disorder.The use of natural ingredients has been widely used by the community to prevent the aging process. Natural compounds from a plant can be a source of antioxidants and have anti-aging abilities through inhibition of the tyrosinase enzyme. Black soybean (Glycine max (L.) Merr.) are high in isoflavone compound, one of which is daidzein.This study evaluates the antioxidant and anti-aging potential of black soybean extract with daidzein. Antioxidants using the DPPH method and anti-aging tests carried out, namely the inhibition of the tyrosinase enzyme, are very important in the aging process.Daidzein has an IC50 value of DPPH scavenging activity around 109.34±2.80 µg/mL lower than black soybean extract with 116.52±2.50 µg/mL. Results on tyrosinase enzyme inhibition activity, black soybean extract had an IC50 value of 70.71±1.83 µg/mL lower compared to daidzein with 72.65±2.81 µg/mL. In contrast to the better antioxidant activity of daidzein, the black soybean extract are more potential to inhibit the enzyme tyrosinase

Abstract

The potential utilization of flavonoid compounds, especially scutellarin, and apigenin, contained in basil (Ocimum basilicum L.) leaf extract to manage the aging effects on the skin, that occurs because of over-activated hyaluronidase enzyme and oxidative stress due to hydrogen peroxide (H2O2) radicals, is not well known. This study was conducted to assess the H2O2 scavenging activity and the inhibition of hyaluronidase from scutellarin and apigenin at various concentrations. The study was conducted by measuring H2O2 scavenging inhibition and hyaluronidase inhibition of scutellarin and apigenin in various concentrations using the spectrophotometry method. The various activity was tested using the One-Way ANOVA test followed by Tukey post hoc test. IC50 values were calculated based on linear regression equations of H2O2 scavenging inhibition and hyaluronidase inhibition. The analysis showed the highest H2O2 scavenging activity was found in scutellarin with IC50 158.76 μg/mL. Scutellarin has greater scavenging activity than apigenin. Hyaluronidase inhibition of scutellarin with IC50 35.25 µg/mL, while apigenin was 162.86 µg/mL. Scutellarin has higher hyaluronidase inhibition activity than apigenin. Antioxidant and antiaging effects of basil leaf extract caused by phytochemical compounds contained, especially scutellarin.
