Abstract

The efficacy and safety of guajava leaf (Psidium guajava L.) extract as antidiarrheal has been tested through pra-clinical and clinical studies. Now dosage form of guajava leaves extract as antidiarrheal has limitation for childrens because it is difficult oto be swallowed, it has high disintegration time and unpleasent smell and flavor so it needs an acceptable dosage formulation for childrens. The aim of this research is to optimize formula dispersible tablets with combination disintegrants of croscarmellose sodium (CCS) and sodium starch glycolate (SSG). Dispersible tablets were produced by dry granulation with five formulas, i.e. formula I (100% CCS), II(75%:25%), III(50%:50%), IV (25%:75%) and V (100% SSG). Standarization of the extract used organoleptic parameters, loss on drying value and thin layer chromatography (TLC). The dispersible tablets were tested for weight uniformity, friability, hardness, disintegration time, dispersion time, wetting time and TLC. Data were analyzed using sofware Design Expert® versi 7.1.4.0 to determine the optimum formula. The result showed that the different compositions of CCS-SSG influence the physical charasteristics of dispersible tablets. The higher of SSG content will increase the hardness, the disintegration time, the dispersion time and the wetting time. The optimum formula was the formula with combination of 71% CCS and 29% SSG.

Abstract

Fungi endofit IP-2 adalah fungi endofit yang diisolasi dari tanaman Artemisia annua L. Fungi ini diketahui mempunyai aktivitas sebagai inhibitor polimerisasi hem. Dalam penelitian ini akan diteliti mengenai media yang sesuai untuk pertumbuhan maupun pembentukan metabolit aktifnya serta menentukan waktu yang sesuai untuk pemanenan metabolit aktifnya. Fungi endofit difermentasi dalam 4 jenis media selama 14 hari. Setiap hari dilakukan pengambilan sampel yang akan digunakan untuk mengetahui bobot sel keringnya yang selanjutnya digunakan untuk analisis pertumbuhannya serta digunakan untuk uji aktivitas inhibitor polimerisasi. Uji dilakukan dengan menggunakan metoda yang dikembangkan oleh Bassilico dkk (1998). Data bobot sel kering dan prosentase penghambatan inbihitor polimerisasi dianalisis sehingga diperoleh kurva pertumbuhan dan kurva produksi metabolitnya yang  akan digunakan untuk menentukan  waktu yang tepat untuk memanen metabolit aktifnya. Hasil penelitian menunjukkan bahwa dalam media PDB fungi endofit IP-2 menunjukkan pertumbuhan yang paling baik dan mampu menghasilkan metabolit aktif paling tinggi dibandingkan 3 media lainnya. Dalam media PDB, pada hari ke-7 terlihat jumlah metabolit aktif yang dihasilkan paling banyak sehingga waktu panen metabolit yang tepat adalah pada hari ke-7.

Abstract

Senggugu root bark has mucolytic activity and has been used empirically, so it needs to be formulated as lozengeswhich can be used practically and comfortable for the patients. Senggugu root bark powder was extracted by maceration using aethanol 70%. Lozenges was optimized using lactose-sorbitol filler mix through three formulas, formula A ( 100% lactose), formula B (100% sorbitol), formula C (50% lactose-50% sorbitol). Lozenges was made by wet granulation method. The optimum formula was obtained from theresults of physical granul test and lozenges using SLD, and analyzed by its granul flow, hardness, dissolution time, and taste responsiveness, and also qualitative and quantitative analysis. The results show that lactose-sorbitol filler mix can increase hardness and taste responsiveness, decrease granul flow and dissolution time. The oprimum formula from this research is 100% sorbitol:0% lactose.

Abstract

Senggugu root bark (Clerodendrum serratum L. Moon) is known as a mucolitics. Senggugu root bark is made in the dosage form of lozenges in combination with sucrose, mannitol and lactose in order to obtain good flavor and comfortable when consumed. The purpose of this study was to determine the effect of the combination of fillers on the physical properties of granules and tablets as well as the composition of the combination of excipients to produce lozenges of extract of senggugu root bark with optimum physical properties. Seven formulas were made with a combination of mannitol, lactose and sucrose as follows: FI (100%: 0%: 0%), FII (0%: 100%: 0%), FIII (0%: 0%: 100%), FIV (50 %: 50%: 0%), FV (50%: 0%: 50%), FVI (0%: 50%: 50%), and FVII (33.3%: 33.3%: 33.3% ). The results showed that the combination of mannitol, lactose and sucrose affected the physical properties of the granules and lozenges of extract of senggugu root bark i.e. flow rate, hardness, dissolved time, compactibility and perceptive sense. The dominant amount of sucrose and lactose can improve the physical properties of granules and tablets. The optimal composition of the combination of mannitol, lactose and sucrose obtained from Design Expert 7.1.5. program was 5.491%: 37.387%: 57.122%, respectively.

Abstract

Rutin and quercetin are   flavonoid compounds containing in the extract of mulberry (Morus alba L.) leaves  have an influence on the cardiovascular system. This study aims to assessment of the blood pressure effect and cardiac puls after giving ethanolic extract of  mulberry leaves.  The extract was made using 95% ethanol by remaseration method. The content of rutin and quercetin in the extract were determined by using HPLC(High Performance Liquid Chromatography).  The formula of the encapsulation containing extract 0.25% w/v, chitosan 0.5mg/mL and TPP (sodium  tripolyphosphate) 1mg/mL. Characterization of extract encapsulated was done on the particle size, encapsulation efficiency and FTIR. Extract encapsulated was given to rats orally,  subsequently arterial blood pressure and heart rate were measured using Coda Non-Invasive Blood Pressure System. The results showed that mulberry extract contains rutin (0.879±0.138) mg/g  and  quercetin (4.427±0.065) mg/g. Ratio of chitosan-TPP with 0.5:1  forms particle size were   25.11-45.2 nm, encapsulation efficiency  (76.14±2.29)%. Mulberry leaves extract and encapsulated extract could decrease arterial blood pressure.
