Abstract

The objective of this research was to study the influence of biological seed treatment on plant growth, yield of rice, and controlling of bacterial leaf blight. The research was conducted at green house of Indonesian Center for Agricultural Biotechnology and Genetic Resources Development, Bogor and Seed Science and Technology Laboratory, Department of Agronomy IPB from August 2009 to Febru-ary 2010. The result showed that biological seed treatment of Pseudomonas spp. and Bacillus spp. (with and without matriconditioning) could increase plant growth of rice in terms of on plant height, number of seedling, root length, root fresh weight, root dry weight, shoot fresh weight, and shoot dry weight. Seed treatments of matriconditioning + P.aeruginosa, matriconditioning + B.subtillis5/B, and Bacillus subtillis 11/C were the best seed treatments in increasing yield of rice as indicated by the highest number of filled grains/panicle, percentage of filled grain/panicle, percentage of filled grain/plant, and the lowest number of unfilled grain/panicle, percentage of unfilled grain/panicle, and percentage unfilled grain/plant. Seed treatments of Pseudomonas diminuta, matriconditioning + Pseudomonas diminuta and matriconditioning + Bacillus subtilis 11/C were significantly resulted in lower percentage of pathogen diseased leaf area (%DLA) than others seed treatment. Percentages of pathogen diseased leaf area were 15.45%, 15.94% and 19.55%. Key words: Bacillus spp Biological seed treatment, matriconditioning, Pseudomonas spp.

Abstract

Layanan pendidikan merupakan salah satu faktor yang dapat menjadi pendukung keberhasilan di dunia pendidikan. Salah satu usaha yang dilakukan guna meningkatkan layanan pendidikan adalah sebagaimana yang dilakukan oleh MI. Miftahul Ulum 3 (bekerjasama dengan Tim Pengabdian Dosen dari Universitas Qomaruddin), dengan mengimplementasikan sebuah sistem informasi (website sekolah) yang sekaligus difungsikan sebagai sistem penerimaan peserta didik baru (PPDB online). Dengan waktu pelaksanaan yang terhitung singkat (5 bulan), kegiatan pengembangan website tersebut dilakukan dengan menerapkan metodologi pengembangan perangkat lunak extreme programming (XP). Dengan implementasi sistem website sekolah ini, diharapkan dapat meningkatkan layanan pendidikan di MI. Miftahul Ulum 3 dan menjadi inisiasi awal dari bentuk layanan-layanan pendidikan lainnya yang terkomputerisasi. Hasil dari pengabdian ini adalah berupa sistem informasi sekolah (website sekolah) sistem penerimaan siswa baru (PPDB online)

Abstract

The objective of the study was to determine the effect of seed treatment on growth, seed quality, P fertilizer utilization, and rice production in the glasshouse. The experiment was conducted in a split plot design with three replications from January to June 2010.The main plots were P fertilizer rates (0, 50, and 100 kg SP-36 ha-1), and sub plots were (1) negative control (seeds were not inoculated by Xanthomonas oryzaepv. oryzae(Xoo) and without seed treatment); (2) positive control (seeds were inoculated by Xoo); (3) seeds were inoculated by Xoo and immersed in Pseudomonas diminutaA6 suspension; (4) seeds were inoculated by Xoo and immersed in Bacillus subtilis5/B suspension; (5) seeds were inoculated by Xoo and immersed in P. diminutaA6 + B. subtilis5/B suspension; (6) matriconditioning + P. diminutaA6, (7) matriconditioning + B. subtilis5/B, and (8) matriconditioning + P. diminutaA6 + B. subtilis5/B. The results showed that seed treatment with biological agents P. diminutaA6, seed treatment with P. diminutaA6 + B. subtilis5/B were the best seed treatments to improve plant growth and yield of rice. Treatment with matriconditioning + P. diminutaA6 + B. subtilis5/B was the best treatment to enhance seed germination. Soaking the seeds in B. subtilis5/B suspension or P. diminutaA6 + B. subtilis5/B suspension reduced the use of P fertilizer, based on the number of grain, total number of grain, percentage of grain per panicle. Abstract

Seed quality is an important factor in rice production. Bacterial leaf blight (BLB) caused by Xanthomonas oryzae pv. oryzae (Xoo) is a seedborne disease of rice that cause serious yield losses in Indonesia. Experiments were conducted at the Laboratory of Rice Research Babakan, IPB, Bogor; Laboratory of Seed Science and Technology at IPB, Bogor; and the Laboratory of Seed PT. EWSI, Purwakarta, from April to August 2011. The objective of this research was to determine the effectiveness of coating Xoo contaminated rice seeds with biological agents on plant growth and crop yield. The experiments were arranged in a randomized complete block design with one factor (seed coating treatment),i.e. T0 = negative control, healthy seed; T1 = positive control, the seeds contaminated with Xoo; T2 = P. diminuta A6 and B. subtilis 5/B; T3 = alginate 3% + 1% peat + P. diminuta A6 and B. subtilis % 5/B; T4 = arabic gum 3% + 1% gypsum + P. diminuta A6 and B. subtilis 5/B; T5 = CMC 1.5% + 1% talc + P. diminuta A6 and B. subtilis 5/B, and T6 = bactericide streptomycin sulfat 4%. The results showed that seed coating treatment with with 3% alginate + 1% peat + P. diminuta A6 + B. subtilis 5/B was the best treatment based on the percentage of full and empty grain weight per panicle, and the percentage of number of full and empty grains per panicle.

Abstract

ABSTRACTThe research objectives were to evaluate biological agent treatments in controlling bacterial leaf blight (BLB) and increasing plant growth and seed production of rice. The experiment was conducted in the greenhouse and field using the same experimental design (randomized block design with two factors) and three replications. The first factor was seed treatments, i.e. negative control, positive control, streptomycin sulphate 0.2%, Pseudomonas diminuta + Bacillus subtilis, matriconditioning + streptomycin sulphate 0.2%, and matriconditioning + P. diminuta + B. subtilis. Spraying plants (second factor): untreated control, streptomycin sulphate 0.2%, biological agent F112, biological agent F198, and biological agent F57. In the greenhouse, matriconditioning + P. diminuta + B. subtilis  improved seed germination, plant height, and plant dry weight. Spraying plants with biological agent F112 increased plant dry weight. Meanwhile, matriconditioning + P. diminuta + B. subtilis followed by spraying plants with biological agent F112 reduced the BLB severity. In the field, matriconditioning + P. diminuta + B. subtilis improved seedling dry weight. Matriconditioning + P. diminuta + B. subtilis and spraying plants with biological agents F112 controlled BLB and increased plant growth. Abstract

ABSTRACTXanthomonas oryzae pv. oryzae (Xoo), is a seedborne pathogen causing bacterial leaf blight (BLB) disease, and reduces the quality of seed and rice production. One of the efforts to control the BLB disease and to improve the quality Xoo infected seeds is the seed coating technique enriched with biological agents. The experiment was aimed to study the effect of coating on seed quality and storage life, as well as the Xoo and biological agents resistence (P. diminuta A6 and B. subtilis 5/B) on the seeds. The experiment was carried out from August 2011 to March 2012, using a split plot design with four replications. The main plot was storage period, i.e. 0, 1, 2, 3, 4, 5, 6, and 7 months. The sub plot was seed coating treatment consisted of negative control (healthy seed); positive control (seeds contaminated with Xoo); seed infested with biological agens;  alginate 3% + 1% peat + biological agents; arabic gum 3% + 1% gypsum + biological agents; CMC 1.5% + 1% talc + biological  agents;  and  bactericide  streptomycin  sulfat 20%. The  coated  seeds were  stored an air-conditioned room (18-20 °C,  RH  48-50%). The results showed that the treatments were able to maintain seeds quality during storage, i.e. germination percentage, uniformity percentage, and vigor index, better than those of the positive control. The P. diminuta A6 was still presence (0.08 x 106 cfu mL-1) in seeds coated after 7 month storage, and the B. subtilis 5/B was still presence (0.07 x 106 cfu mL-1) up to 6 month storage with 3% arabic gum + 1% gypsum + biological agents.  Keywords: Bacillus subtilis 5/B, Pseudomonas diminuta A6, seed quality, storage space, Xanthomonas oryzae pv. oryzae

Abstract

Bacterial leaf blight, the disease caused by Xanthomonas oryzae pv. oryzae (Xoo), a seedborne pathogen, has been known to reduce rice yield by 50%. In this study, two consecutive experiments were conducted in order to investigate the effect of various biological seed treatments applied on rice seeds artifi cially infected by Xoo on seed quality and seedling growth. Laboratory experiment was arranged in a completely randomized design while greenhouse experiment was arranged in a completely randomized block design. In both experiments, 12 seed treatments were applied: un-infected seeds (negative control) without seed treatment, Xoo infected seeds (positive control) without seed treatment, infected seeds soaked in bactericide Agrept 0.2%, infected seeds soaked in biological agent suspension isolate A6, infected seeds soaked in A54 isolate, infected seeds soaked in 5/B isolate, infected seeds soaked in 11/C isolate, infected seeds matriconditioned + A6 isolate, infected seeds matriconditioned + A54 isolate, infected seeds matriconditioned + 5/B isolate, and infected seeds matriconditioned + 11/C isolate. Soaking seeds either in Agrept 0.2% or in biological agent suspension was conducted for 30 h. Matriconditioning was conducted using ratio of seeds to carrier (burned rice hull 32 mesh) to biological agent suspension or bactericide solution of 1.0 (g) :0.8 (g): 1.2 (mL) for 30 h in air-conditioned room ca. 25 oC. Results of experiments showed that all biological seed treatments could suppress Xoo in rice seeds. Matriconditioning plus biological agent (isolate A6), biopriming with isolate A6 or isolate A54 were the best seed treatments to improve seed viability and vigor. In greenhouse experiment, matriconditioning plus isolate A54 was the best seed treatment to increase seedling growth.Keywords: biopriming, matriconditioning, rhizobacteria, seed health, viability

Abstract

The objectives of this research were to find the most compatible coating formula for rice seeds (Experiment 1), and to evaluate the effect of coating formula on viability, vigor and storability of the seeds (Experiment 2). This research was conducted in Laboratory of Seed Science and Technology at IPB, Bogor and Laboratory of Seed Technology at PT East West Seed Indonesia, Purwakarta from December 2010 to February 2011. In Experiment 1, the ten coating materials (formula) were arranged in single factor of Completely Randomized Design with four replications, i.e. uncoated seed, 3% alginate + 1% talc, 3% alginate + 1% gypsum, 3% alginate + 1% peat, 3% arabic gum + 1% talc, 3% arabic gum + 1% gypsum, 3% arabic gum + 1% peat, 1.5% CMC + 1% talc, 1.5% CMC + 1% gypsum, and 1.5% CMC + 1% peat. Experiment 2 used Split Plot Design with four replications. The main plot was storage periods i.e. 0, 1 and 2 months. The seeds were stored in two storage conditions: ambient and air conditioned room. The subplot was coating formula (same as in Experiment 1). The results of Experiment 1 showed that the most compatible coating material for rice seeds were 1.5% CMC + 1% talc, and 1.5% CMC + 1% gypsum, these formula were able to maintain vigor of the coated seeds. In Experiment 2, coating formula of 1.5% CMC + 1% peat generated higher seed vigor than uncoated seeds after two months storage, 3% gum arabic + 1% gypsum was able to keep vigor for one month storage in ambient condition. The coating formula of 3% alginate + 1% peat was able to keep viability and vigor of the coated seeds for two months in air conditioned room.
