Abstract

ABSTRAK Pisang mauli (Musa sp. AA) adalah pisang khas Kalimantan Selatan. Kulit buah pisang mauli mengandung flavonoid yang berperan sebagai antihiperlipidemia. Berdasarkan kandungan flavonoid tersebut ekstrak dan agar-agar kulit pisang mauli memiliki potensi sebagai salah satu terapi antihiperlipidemia. Tujuan penelitian ini adalah menentukan kadar flavonoid total, aktivitas antioksidan dan efek antihiperlipidemia dari ekstrak dan agar agar kulit pisang mauli. Penentuan kadar flavonoid total dilakukan dengan metode kolorimetri menggunakan AlCl3. Sedangkan aktivitas antioksidan ditentukan dengan metode DPPH. Penentuan efek antihiperlipidemia dilakukan pada tikus Wistar jantan yang diinduksi dengan Propiltiourasil, tikus dibagi menjadi lima kelompok : kelompok kontrol normal (Na-CMC), kelompok kontrol positif (Simvastatin 40 mg/70Kg BB), kelompok kontrol negatif, kelompok ekstrak 240 mg/Kg BB dan kelompok agar-agar 1 gram/Kg BB. Ekstrak dan agar-agar diberikan secara oral selama delapan hari. Pada hari berikutnya dilakukan pengukuran kadar kolesterol total dan trigliserida. Hasil penelitian menunjukkan bahwa ekstrak dan agar-agar kulit pisang mauli mengandung flavonoid sebanyak 0,8339 dan 0,2021 mg kuersetin/100 mg sampel. Ekstrak dan agar-agar kulit pisang memiliki nilai % inhibisi terhadap DPPH sebesar 58,6794 dan 25,4792 %. Efek antihipelipidemia pada ekstrak 240 mg/kg BB lebih kuat daripada agar-agar 1 gram/Kg BB. Kesimpulan dari penelitian ini adalah kadar flavonoid total, aktivitas antioksidan dan efek antihiperlipidemia ekstrak lebih tinggi dan kuat daripada agar-agar. Kata kunci : Kulit pisang mauli, ekstrak, agar-agar, kadar flavonoid total, aktivitas antioksidan, antihiperlipidemia ABSTRACTMauli Banana (Musa sp. AA) is endemic banana’s from South Borneo. The peel of fruit contains flavonoid which have antihyperlipidemic effect.. Based on the flavonoid content, extract and jelly of mauli banana peel has potential as a therapeutic of antihyperlipidemic. The aim of this research are to determine total flavonoid content, antioxidant activity and antihyperlipidemic effect of extract and jelly of mauli banana peel. The total flavonoid content was determined by colorimetric method using AlCl3. The antioxidant activity was determined by DPPH method. The Antihyperlipidemic effect determined using Propylthiouracil-induced male Wistar rats animals. The rats divided into five group : normal control group (Na-CMC), positive control group (Simvastatin), negative control group, extract 240 mg/Kg b.wt  group and jelly 1 gram/Kg b.wt group. The extract administrated orally within eight days. On the next day the total cholesterol and triglycerides level was determined . The result showed that extract and jelly of mauli banana peel contains 0,8339 and 0,2021 mg quercetin/100mg sample. Extract and jelly of mauli banana peel has 58,6794 and 25,4792 % of  % inhibition value on DPPH. The antihyperlipidemic effect on extract 240 mg/Kg b.wt group stronger than jelly 1 gram/Kg b.wt. The conclusion of this research are total flavonoid content, antioxidant activity and antihyperlipidemic effect of extract higher and stronger than jelly. Keywords :

Abstract

ABSTRAK Rambut jagung mengandung flavonoid. Kandungan total flavonoid dipengaruhi oleh usia tanaman. Flavonoid memiliki potensi sebagai antihiperglikemia. Penelitian bertujuan untuk melihat profil penurunan kadar glukosa darah ekstrak air rambut jagung tua dan muda pada mencit jantan galur Balb-C yang diinduksi aloksan. Penelitian bersifat eksperimental dengan rancangan posttest only control group design. Ekstrak dibuat dengan merebus rambut jagung menggunakan air selama 30 menit. Total flavonoid ditentukan dengan metode AlCl3. Aktivitas penurunan kadar glukosa darah dilakukan dengan metode tes toleransi glukosa oral terhadap mencit jantan galur Balb-C yang diinduksi aloksan secara intraperitonial (140 mg/kgBB). Sebanyak 20 hewan uji dibagi menjadi kelompok kontrol positif (Akarbose 0,1305 mg/20 gBB), kontrol negatif (NaCMC 1%), ekstrak air rambut jagung tua (200 mg/kgBB) dan ekstrak air rambut jagung muda (200 mg/kgBB) yang diberi perlakuan secara oral. Kadar glukosa darah diukur pada menit ke 0, 30, 60, 90, 120, 150, dan 180 pasca pembebanan glukosa oral. Data persen penurunan kadar glukosa darah dianalisis dengan uji Kruskal Wallis dan dilanjutkan dengan uji Mann Whitney. Persen penurunan kadar glukosa darah ekstrak air rambut jagung tua dan muda tidak berbeda bermakna (p>0,10). Ekstrak air rambut jagung muda menurunkan kadar glukosa darah sejak menit ke-90 pasca pemberian glukosa oral dengan persen penuran sebesar 44,55 ± 4,92%. Sedangkan ekstrak air rambut jagung tua menurunkan kadar glukosa darah sejak menit ke-120 pasca pemberian glukosa oral dengan persen penurunan sebesar 30,15 ± 7,25%. Kata kunci: rambut jagung tua, rambut jagung muda, flavonoid, kadar glukosa darah ABSTRACTCorn silk contained flavonoid. Total flavonoid content could be influenced by ages of the plant. Flavonoid has potential effect as anti-hyperglycemia. The purpose of this study was to observe lowering blood glucose levels profile of mature and immature corn silk extract on male mice Balb-C strain. This is an experimental study with posttest only control group design. Aqueous extract was prepared by boiling corn silk with water for 30 minutes. Total flavonoid content was determined by AlCl3 method. Lowering blood glucose levels activity was determined by oral glucose tolerant test method on alloxan induced male Balb-C mice test animals intraperitonialy (140 mg/kgBB). The 20 test animals were divided into positive control group (Acarbose 50 mg/70kgBB), negative control group (NaCMC 1%), mature aqueous corn silk extract group (200 mg/kgBB), and immature aqueous corn silk extract group (200 mg/kgBB) given orally. Blood glucose levels were measured at 0, 30, 60, 90, 120, 150, and 180 minutes after the glucose load. Percentage of lowering blood glucose levels were analyzed by Kruskal Wallis and continued by Mann Whitney test. Percentage of lowering blood glucose levels of mature and immature aqueous corn silk extract was not significantly different. Immature aqueous corn silk extract was lowering blood glucose since 90 minute after the glucose load by 44,55 ± 4,92%.  Whereas mature aqueous corn silk extract was lowering blood glucose since 120 minute after the glucose load by 30,15 ± 7,25%. Key word: mature corn silk, immature corn silk, flavonoid, blood glucose levels
