Abstract

ABSTRAK            Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Pseudomonas aeruginosa pada kasus Ear mites kucing domestik di Kecamatan Syiah Kuala, Banda Aceh. Sampel yang digunakan berasal dari 10 ekor kucing domestik yang positif menderita Ear mites. Pengambilan sampel dilakukan dengan metode Cluster Random Sampling terhadap lima desa dari sepuluh desa yang terdapat di Kecamatan Syiah Kuala. Hasil swab liang telinga kucing dimasukan ke media Nutrient Broth (NB) dan diinkubasikan pada suhu 37◦C selama 24 jam, selanjutnya dilakukan penanaman pada media Tryptic Soy Agar (TSA) lalu diinkubasikan kembali pada suhu 37◦C selama 24 jam. Koloni bakteri yang tumbuh terpisah pada TSA dilakukan pengamatan ukuran, warna, elavasi dan pinggiran dari koloni. Pewarnaan Gram dilakukan untuk memastikan bakteri yang terdapat adalah kelompok Gram negatif. Terakhir dilakukan uji IMViC (Indol, Methyl Red, Voges Prokauer, Sulfid Indol Motility, Simmon’s Citrate Agar), uji biokimia Triple Sugar Iron Agar dan uji gula gula (Glukosa, Sukrosa, Maltosa, Laktosa, Manitol). Hasil penelitian ini di analisis secara deskriptif. Hasil pemeriksaan dari 10 sampel ternyata tidak ditemukan adanya infeksi sekunder oleh Pseudomonas aeruginosa pada kasus Ear mites. Oleh karena itu, dapat disimpulkan bahwa Pseudomonas aeruginosa bukan merupakan salah satu penyebab infeksi sekunder pada kasus Ear mites kucing domestik (Felis domesticus) di Kecamatan Syiah Kuala, Banda Aceh. Kata kunci: kucing domestik, Ear mites, Pseudomonas aeruginosa, Banda Aceh ABSTRACT          This study aims to isolate and identify Pseudomonas aeruginosa in the case of Ear mites of domestic cats in the sub district of Syiah Kuala, Banda Aceh. The samples were used 10 domestic cats that are positive suffer from ear mites. The samples were carried out by using the method of cluster random sampling from five out of ten villages in the sub district of Syiah Kuala. The results of cat ear canal swab inserted into media of Nutrient Broth (NB) incubated at 37◦C for 24 hours. Next, using sterile osse the bacteria are modulated in the media of Tryptic Soy Agar (TSA) and incubated at of 37◦C for 24 hours. The morphology colony of bacteria that grew separated in the TSA was observed for size, colour, elevation and margin of those colonies. Gram staining procedure were done to make sure if the bacteria is belongs to Gram negative group. Finally, IMViC test (Indol, Methyl Red, Voges Proskauer, Sulfit Indol Motility, Simmon’s Citrate Agar), biochemical test Triple Sugar Iron Agar and sugars test (Glucose, Maltose, Sucrose, Lactose, Manitol) were conducted. The results of this study were analyzed descriptively. The examination shows none out of 10 samples were found Pseudomonas aeruginosa in the case of Ear mites. Therefore this can be concluded that Pseudomonas aeruginosa were not one of the reason that caused a secondary infection in the case of Ear mites of domestic cats (Felis domesticus) in sub district Syiah Kuala, Banda Aceh.    Keywords: domestic cat, Ear mites, Pseudomonas aeruginosa, Banda Aceh

Abstract

ABSTRAKSalmonella sp. merupakan penyebab penyakit salmonellosis akibat foodborne disease yang bersifat zoonosis. Penelitian ini bertujuan untuk mengetahui perbandingan infeksi Salmonella sp. pada ayam kampung dan broiler yang di potong di Pasar Lambaro Aceh Besar. Sampel penelitian yang digunakan berupa usus ayam yang terdiri dari 15 usus ayam kampung dan 15 dari usus ayam broiler. Isolasi Salmonella sp. dilakukan dengan memasukkan swab isi usus ayam ke dalam media SCB, yang diinkubasikan dengan suhu 37ᵒC selama 24 jam. Selanjutnya apabila media SCB menjadi orange maka dilanjutkan dengan penanaman pada media SSA dan diinkubasikan pada suhu 37ᵒC selama 24 jam. Kemudian dilakukan pengamatan morfologi koloni yang tumbuh lalu dilakukan pewarnaan Gram. Data penelitian dianalisis secara deskriptif. Hasil penelitian menunjukkan bahwa ayam kampung dan ayam broiler 100% terinfeksi Salmonella sp. Berdasarkan hasil penelitian dapat disimpulkan bahwa tidak ada perbedaan jumlah ayam yang terinfeksi bakteri Salmonella sp. pada ayam kampung dan ayam broiler yang di potong di Pasar Lambaro Aceh Besar. Oleh sebab itu maka dapat disimpulkan bahwa tidak ada perbedaan infeksi oleh Salmonella sp. pada ayam kampung dan ayam broilerKata kunci: infeksi, foodborne disease, Salmonella sp., salmonellosis. ABSTRACTSalmonella sp. is a cause disease salmonellosis due to  foodborne disease, which zoonotic. The aim of this study to know the comparison of Salmonella sp. in native chickens and broilers that are slaughtered at the Lambaro Aceh Besar Market. The research sample used in the form of chicken intestines consisting of 15 native chicken intestines and 15 of bowel ayam broiler. Isolation of Salmonella sp. done by inserting a swab of chicken intestine contents into the media SCB, which was incubated at 37ᵒC for 24 hours. Furthermore, if the SCB media becomes orange then continued with planting on SSA media and incubated at 37ᵒC for 24 hours. Then, the morphology of the growing colony was observed and then Gram stained was carried out. The research data were analyzed descriptively. The results showed that 100% native chickens and broilers were infected with  Salmonella sp. Based on the results of the study it can be concluded that there is no difference in the number of chickens infected with Salmonella sp. in free-range chickens and broiler chickens that were slaughtered at Lambaro Market, Aceh Besar. By Therefore, it can be concluded that there is no difference in infection by Salmonella sp. in free-range chickens and broilers.Keywords: infection, foodborne disease, Salmonella sp., Salmonellosis.

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Gram positif kokus yang diduga menjadi penyebab pada kasus ear mites kucing domestik di Kecamatan Syiah Kuala Kota Banda Aceh. Sampel yang digunakan berasal dari 10 ekor kucing domestik yang positif menderita ear mites. Hasil swab liang telinga kucing dimasukan ke media Nutrient Broth (NB) dan diinkubasikan pada suhu 37°C selama 24 jam, selanjutnya menggunakan osse steril dilakukan penanaman pada media Manitol Salt agar (MSA) dan media Plate Agar Darah (PAD) dengan teknik goresan T lalu diinkubasikan kembali selama 24-48 jam. Morfologi koloni yang tumbuh terpisah diamati bentuk, ukuran, warna dan elevasi sebelum dilakukan pewarnaan Gram. Uji biokimia (Manitol dan Glukosa) dan Uji katalase dilakukan pada bakteri yang berbentuk kokus. Hasil penelitian ini dari 10 sampel dinyatakan 8 sampel (80%) penyebab infeksi sekunder pada kasus ear mites adalah bakteri Gram positif dari kelompok Staphylococcus sp dan 2 sampel (20%) dari kelompok Streptococcus sp. Dari 8 sampel yang terinfeksi bakteri kelompok Staphylococcus, 3 sampel (37,5%) merupakan infeksi tunggal Staphylococcus aureus dan 2 sampel (20%) merupakan infeksi tunggal Staphylococcus epidermidis, selain itu terdapat infeksi ganda antara Staphylococcus aureus dan juga Staphylococcus epidermidis sebanyak 2 sampel (20%) dan infeksi ganda Staphylococcus aureus dengan Micrococcus sp sebanyak 1 sampel (12,5%). Kata Kunci: Kucing domestik (Felis domesticus), Ear mites, bakteri Gram positif kokus ABSTRACT             The objectives of this study was to isolate and identify the Gram positive coccus bacteria which were suspected as one of the cause of secondary infection in ear mites case of domestic cats in Subdistric of Syiah Kuala, Banda Aceh. In this study, 10 of domestic cats suffer from ear mites were used. The swab ear canal from each cat  was inoculated in Nutrient Broth (NB) and incubated at 37°C for 24 hours. Then, using steril osse bacteria were cultured in Manitol Salt Agar (MSA) and Blood Agar Plate (BAP) with T streak method and incubated for 24-48  hours. The morphology coloni of bacteria were observed for shape, size, colour and elevation before Gram staining procedure was applied. Biochemical (manitol and glucose) and catalase test then conducted for the colonies which were identified as coccus in shape. The result of this study showed, 8 samples (80%) out of 10  samples were positive infected by the group of Staphylococcus sp and 2 samples (20%)  are positive infected by the group of Streptococcus sp. Frome those 8 samples  infected by the group of Staphylococcus sp, 3 samples (37,5%)  only infected by Staphylococcus aureus and 2 samples (25%) only infected by Staphylococcus epidermidis while the others 2 samples (25%) were infected by both Staphyloccocus aureus and Staphylococcus epidermidis and only 1 samples (12,5%) was infected by Staphylococcus aureus and Micrococcus sp. It can be concluded that  the group of Gram positive bacteria was the one of the cause of  secondary infection in the cats by ear mites.    Keywords: Domestic cat (Felis domesticus), Ear mites, Gram positif coccus bacteria
