Abstract

Oral and respiratory diseases have been related with the fungal infections like oral candidiasis caused by Candida albicans and chronic rhinitis caused by Aspergillus niger. Both of these micr oorganisms are including in flora normal in the oral and nasal cavity. Aspergillus niger is rarely reported as aeroalergen in allergy rhinitis cases, but in chronic rhinitis its fungals are A. flavus and A. fumingatus. The aim of this study was to analyze the risk factors that influencing rhinitis and oral candidiasis and its relation. This research used rhinitis mucous identified various fungal used pour plate method.And then, cultured with SDA medium (Sabauraud's Dextrose Agar) to identify fung als morphology usedcultured slide.The results showed that Aspergillus niger was found more dominant than A. fumingatus and A. flavus in chronic rhinitis.In conclusion, Aspergillus niger was proved as an aeroallergen had had an important role in chronic rhinitis a nd could trigger the growth of Candida albicans.

Abstract

The present study aimed to isolate gram-negative bacteria infected metacarpal of Panthera tigris sumatrae trapped in Subulussalam, Aceh Province. Swab sample was obtained from metacarpal injury of P. tigris sumatrae. Swab sample was cultured to nutrient broth media using sterile cotton swabs or Pasteur pipettes, and incubated at 37°C temperature for 24 hours. Culture was spared on MacConkey media and incubated again at 37°C temperature for 24 hours. Determination of bacteria colony growth on the surface of MacConkey media based on shape, colour, surface, size, and viscosity (consistency). The bacteria colony stained with Gram staining, and tested biochemically.  The result showed that gram-negative bacteria such as Citrobacter sp., Proteus sp., Providencia sp., Pseudomonas sp.,and Salmonella sp. isolated from metacarpal injury of P. tigris sumatrae.

Abstract

A study was conducted to examine some poultry sellers’ perception of the Avian influenza (birdflu) in nine locations (Ulee Kareng, Batoh, Lamnyong, Kampong Baru, Peunayong, Setui, Kampong Ateuk, and Neusu) in Banda Aceh Municipality, Indonesia. Primary data were obtained with the use of a structured questionnaire administared to ninety nine (99) respondents selected using proportional stratified random sampling, between November to December 2011. Information was obtained using the questionnaire sheets contain questions: knowladge, attitude, availability of facilities, and the effort of Avian influenza prevention strategy. Data obtained were analyzed by descriptive statistics. The result showed that only 52% of the poultry sellers’ had good knowledge of early symptomps of Avian influenza. Amount 59% of the poultry sellers’ had good attitudes. Result also showed that 73% of respondents had good facilities that was available in live poultry market area, but only 70% of respondents were good awareness about the effort of Avian influenza prevention strategy. In addition, the respondents who had more knowledge, good attitudes, and/or good facilities were also those who actually acted more preventively. Based on the result we concluded that the poultry sellers’ perception on prevention strategy for infection of birdflu in Banda Aceh categorized as poor. It is suggested that compliance with preventive measures may be enhanced through behavioural modification and building awareness through communication, training, workshop programs to the live poultry market sellers’ in Banda Aceh

Abstract

Penelitian ini bertujuan mengetahui perbedaan jumlah kontaminasi Salmonella sp pada peralatan pemotongan unggas di pasar Lamnyong Kecamatan Syiah Kuala Kota Banda Aceh, dengan menggunakan prosedur pengenceran berseri  10⁻¹ - 10⁻⁵. Perhitungan Salmonella sp dilakukan pada media SSA dengan metode TPC pour plate secara  duplo. Sampel penelitian diambil dari 5 peralatan pemotongan unggas yaitu bak perebusan, alat pencabutan bulu, bak pencucian, meja dan talenan pada 2 lokasi yang berbeda. Hasil penelitian menunjukkan bahwa terdapat perbedaan jumlah kontaminasi Salmonella sp pada peralatan pemotongan unggas. Pengamatan pada 10 sampel yang diambil, 8 diantaranya positif terkontaminasi salmonella sp dengan jumlah kontaminasi tertinggi terdapat pada talenan yaitu 4,8×10⁴ sedangkan kontaminasi terendah terdapat pada meja karkas yaitu 0,3×10³ dan negatif terdapat pada bak perebusan karkas. Hasil penelitian tersebut dapat disimpulkan bahwa kontaminasi Salmonella sp yang tertinggi terdapat pada talenan.

Abstract

Penelitian ini bertujuan untuk mengetahui kontaminasi Salmonella sp pada ayam bakar yang dijual di rumah makan Kecamatan Syiah Kuala Kota Banda Aceh. Sampel diambil dari lima Gampong yaitu Gampong Lampineung, Lamgugob, Jeulingke, Peurada, dan Darussalam. Penelitian ini menggunakan metode Carter yang telah dimodifikasi. Sampel penelitian diamati di rumah makan Kecamatan Syiah Kuala Kota Banda Aceh, dan dikultur pada media Selenite Cystine Broth (SCB). Sampel positif pada media SCB kemudian dikultur pada media selektif Salmonella Shigella Agar (SSA). Selanjutnya dilakukan pewarnaan Gram terhadap koloni yang menciri Salmonella sp. Identifikasi Salmonella sp dilakukan dengan uji biokimia pada media MR-VP, Indol, Simmons’s Citrate Agar (SCA), Triple Sugar Iron Agar (TSIA), Sulfide Indol Motility (SIM), dan media gula-gula (sukrosa, manitol, glukosa, dan laktosa). Hasil penelitian ditemukan semua sampel positif terkontaminasi oleh Salmonella sp. Dapat disimpulkan bahwa, sampel ayam bakar yang dijual di rumah makan Kecamatan Syiah Kuala Kota Banda Aceh telah terkontaminasi oleh Salmonella sp.This study aims to identify the contamination of Salmonella sp in roasted chicken from local restaurans in Syiah Kuala, Banda Aceh. The Samples were taken from five locations in Syiah Kuala district. The locations were Lampineung, Lamgugob, Jeulingke, Peurada, and Darussalam. This study used modified Carter’s method for bacterial identification. Samples collected and inoculated in Selenite Cystine Broth (SCB).Positive sample on SCB media then cultured on Salmonella Shigella Agar (SSA) as selective media for Salmonella sp, Gram stain was done for spesific colonies. Biochemical identificationfor Salmonella spcultured from bacterial isolates from SSA to Methyl Red - Voges Proskauer(MR-VP), Indol, Triple Sugar Iron Agar (TSIA), Sulfide Indole Motility (SIM), Simmon’sCitrat Agar (SCA), and carbohydrate media (sucrose, mannitol, glucose, and lactose. The result showed that all of the samples were positively contaminated with Salmonella sp. Thus, the author can conclude that roasted chicken from five local restaurants in Syiah Kuala Banda Aceh are contaminated with Salmonella sp.

Abstract

ABSTRAK                Penelitian ini bertujuan untuk mengetahui keberadaan dan jenis bakteri enterik pada feses gajah sumatera (Elephas maximus sumatranus) di Pusat Konservasi Gajah (PKG) Saree, Aceh Besar. Sampel feses segar dikoleksi pada pagi hari dari 6 ekor gajah sumatera dengan cara feses langsung diambil dari anus menggunakan swab dan dimasukan dalam Nutrient broth lalu disimpan dalam Cooler box steril. Penelitian ini menggunakan metode Carter (1987) yang sudah dimodifikasi. Untuk isolasi dan identifikasi bakteri enterik pada feses gajah sumatera. Semua sampel feses dikultur pada media MacConkey, Salmonella Shigella Agar (SSA), IMViC (Indol, Methyl red, Voges-Proskauer, sulfid indol motility, Simmons citrate), media Triple Sugar Iron Agar (TSIA), dan uji fermentasi gula-gula (glukosa, laktosa, sukrosa, maltosa). Data hasil penelitian ini dianalisis secara deskriptif berdasarkan keberadaan bakteri enterik yang terdapat dalam feses. Dari hasil penelitian diketahui bahwa di dalam feses lima ekor gajah sumatera yang berumur di atas enam tahun terdapat lebih dari satu bakteri enterik, sedangkan satu ekor gajah yang berumur satu tahun hanya terdapat satu jenis bakteri enterik. Pada gajah bernama Midok ditemukan bakteri E. Coli dan Salmonella sp, pada gajah Amoy bakteri E. Coli dan Salmonella sp, gajah bernama Ella ditemukan bakteri Enterobacter sp dan Salmonella sp, gajah bernama Senna bakteri E. Coli dan Citrobacter sp, gajah bernama Osin ditemukan bakteri E. coli dan Salmonella sp, dan gajah bernama Junaidi ditemukan bakteri E. coli.  Dari penelitian ini dapat disimpulkan bahwa bakteri enterik yang terdapat pada feses gajah sumatera adalah Eschericha coli, Salmonella sp, Enterobacter sp, dan Citrobacter sp.

Abstract

ABSTRAK            Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi Pseudomonas aeruginosa pada kasus Ear mites kucing domestik di Kecamatan Syiah Kuala, Banda Aceh. Sampel yang digunakan berasal dari 10 ekor kucing domestik yang positif menderita Ear mites. Pengambilan sampel dilakukan dengan metode Cluster Random Sampling terhadap lima desa dari sepuluh desa yang terdapat di Kecamatan Syiah Kuala. Hasil swab liang telinga kucing dimasukan ke media Nutrient Broth (NB) dan diinkubasikan pada suhu 37◦C selama 24 jam, selanjutnya dilakukan penanaman pada media Tryptic Soy Agar (TSA) lalu diinkubasikan kembali pada suhu 37◦C selama 24 jam. Koloni bakteri yang tumbuh terpisah pada TSA dilakukan pengamatan ukuran, warna, elavasi dan pinggiran dari koloni. Pewarnaan Gram dilakukan untuk memastikan bakteri yang terdapat adalah kelompok Gram negatif. Terakhir dilakukan uji IMViC (Indol, Methyl Red, Voges Prokauer, Sulfid Indol Motility, Simmon’s Citrate Agar), uji biokimia Triple Sugar Iron Agar dan uji gula gula (Glukosa, Sukrosa, Maltosa, Laktosa, Manitol). Hasil penelitian ini di analisis secara deskriptif. Hasil pemeriksaan dari 10 sampel ternyata tidak ditemukan adanya infeksi sekunder oleh Pseudomonas aeruginosa pada kasus Ear mites. Oleh karena itu, dapat disimpulkan bahwa Pseudomonas aeruginosa bukan merupakan salah satu penyebab infeksi sekunder pada kasus Ear mites kucing domestik (Felis domesticus) di Kecamatan Syiah Kuala, Banda Aceh. Kata kunci: kucing domestik, Ear mites, Pseudomonas aeruginosa, Banda Aceh ABSTRACT          This study aims to isolate and identify Pseudomonas aeruginosa in the case of Ear mites of domestic cats in the sub district of Syiah Kuala, Banda Aceh. The samples were used 10 domestic cats that are positive suffer from ear mites. The samples were carried out by using the method of cluster random sampling from five out of ten villages in the sub district of Syiah Kuala. The results of cat ear canal swab inserted into media of Nutrient Broth (NB) incubated at 37◦C for 24 hours. Next, using sterile osse the bacteria are modulated in the media of Tryptic Soy Agar (TSA) and incubated at of 37◦C for 24 hours. The morphology colony of bacteria that grew separated in the TSA was observed for size, colour, elevation and margin of those colonies. Gram staining procedure were done to make sure if the bacteria is belongs to Gram negative group. Finally, IMViC test (Indol, Methyl Red, Voges Proskauer, Sulfit Indol Motility, Simmon’s Citrate Agar), biochemical test Triple Sugar Iron Agar and sugars test (Glucose, Maltose, Sucrose, Lactose, Manitol) were conducted. The results of this study were analyzed descriptively. The examination shows none out of 10 samples were found Pseudomonas aeruginosa in the case of Ear mites. Therefore this can be concluded that Pseudomonas aeruginosa were not one of the reason that caused a secondary infection in the case of Ear mites of domestic cats (Felis domesticus) in sub district Syiah Kuala, Banda Aceh.    Keywords: domestic cat, Ear mites, Pseudomonas aeruginosa, Banda Aceh

Abstract

AKBISprob merupakan ampas kedelai dan bungkil inti sawit yang difermentasi dengan Aspergillur niger. Penelitian bertujuan mengetahui total mikroba pada telur ayam yang diberi pakan tambahan AKBISprob 4% selama penyimpanan pada suhu refrigerator dan suhu  ruangan selama 14 hari. Penelitian menggunakan 12 ekor ayam petelur strain ISA Brown berumur 16 bulan. Sampel yang digunakan adalah telur ayam. Sampel diambil pada hari ke 29 dari setelah pemberian AKBISprob. Rancangan penelitian menggunakan Rancangan Acak Lengkap Faktorial (RAL Faktorial) terdiri atas dua faktor yaitu faktor pengaruh suhu penyimpanan terhadap kualitas  telur dan faktor lamanya penyimpanan telur penelitian ini terdiri dari 4 perlakuan dan 3 ulangan. Perlakuan P0 (pakan komensial 324-2R 110 g); P1 (AKBISprob 4% yang diberikan setiap hari); P2 (AKBISprob 4% yang diberikan tiga hari sekali); dan P3 (AKBISprob 4% yang diberikan lima hari sekali). Total mikroba telur selama penyimpanan pada suhu refrigerator dan suhu  ruangan dianalisis dengan menggunakan analisis varian. Rata-rata (±SD) total mikroba telur dari ayam yang diberi AKBISprob pada penyimpanan suhu ruangan dan suhu refrigerator P0, P1, P2, dan P3 berurutan adalah total mikroba pada penyimpanan suhu ruangan 4,57±0,96, 5,03±0,09, 4,82±0,16 dan 4,86±0,29. Total Mikroba pada penyimpanan suhu refrigerator 4,95±0,06, 4,78±0,16, 4,43±0,03 dan 4,53±0,99. Hasil menunjukkan penyimpanan telur pada suhu refrigerator berpengaruh nyata (P0,05) terhadap penyimpanan pada suhu ruangan. Pemberian AKBISprob 4% tidak berpengaruh secara nyata (P0,05) terhadap total  mikroba pada telur. Tidak ada interaksi antara suhu dan perlakuan AKBISprob 4% terhadap jumlah total mikroba.(This study aimed to determine total microbes ofeggs which were suplemented with AKBISprob 4% while stored in refrigerator and room temperature for 14 days. This study used 12 ISA Brown Strain Layer Poultry aged 16 months. Sample were taken in 29th days since first time AKBISprob was fed. This study used Factorial Randomized Design based on two factors and divided to 4 treatments and 3 repetitions. P0 (commercial feed 324-2R 110 g); P1 (AKBISprob 4%, fed everyday);P2 (AKBISprob 4%, fed once in three days); and P3 (AKBISprob 4%, fed once in five days).Total of microbes while stored in refrigerator were analyzed by using ANOVA (Analysis of Varians).Total of microbes (±SD) from AKBISprob treatmentand eggs were produced then stored in room and refrigerator temperature ofP0, P1, P2, and P3respectively, total microbes in room temperature were4,57±0,96, 5,03±0,09, 4,82±0,16and4,86±0,29 andafter refrigerator storage were4, 95±0,06, 4,78±0,16, 4,43±0,03 and4,53±0,99.This result showed that refrigerator storage has significantly affect (P0,05) to room temperature storage. but, thathas not significantly (P0,05) to total microbes on eggs.So there are no relation between temperature andAKBISprob 4%treatment to total microbes on eggs.)

Abstract

Penelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Escherichia coli dan Salmonella sp pada telur ayam kampung yang gagal menetas di Laboratorium Lapangan Peternakan Universitas Syiah Kuala. Sebanyak 20 butir telur ayam kampung yang gagal menetas diperiksa di Laboratorium Mikrobiologi Fakultas Kedokteran Hewan, Universitas Syiah Kuala Banda Aceh. Sampel diisolasi berdasarkan metode Carter yang dimodifikasikan. Pada media Brilliance E.coli/ Coliform Selective Agar koloni bakteri tumbuh berbentuk bulat berwarna ungu, pada pewarnan Gram bakteri bewarna pink berbentuk batang pendek. Pada media SSA koloni bakteri berbentuk bulat bewarna hitam dan pada pewarnaan Gram bakteri bewarna pink berbentuk batang panjang. Hasil uji IMViC dan gula-gula sesuai dengan karakteristik Escherichia coli dan Salmonella sp. Hasil penelitian dari 20 sampel telur ayam kampung yang diteliti 3 sampel positif terinfeksi Escherichia coli dan 12 sampel positif Salmonella sp. Maka dapat disimpulkan Escherichia coli dan Salmonella sp merupakan salah satu penyebabab kegagalan menetas pada telur ayam kampung yang gagal menetas di Laboratorium Lapangan Peternakan Universitas Syiah Kuala.Kata kunci : telur ayam kampung, gagal menetas, Escherichia coli, Salmonella spThis study aims to isolat and identify bacteria Escherichia coli and Salmonella sp from failed to hatch domestic chicken in Field Laboratory of Animal Husbandry Syiah Kuala University. Twenty unhatched domestic chicken eggs were examined in microbiology laboratorium of Veterinary Faculty of Syiah Kuala University, Banda Aceh. Modified Carter methode was used to isolate bacterial from the samples. On media Brilliance E.coli/ Coliform Selective Agar colonies bacterial were purple and circular, on Gram staining it formed pink color cocobacil shape. On media SSA the colonies were black and circular, on Gram staining it formed pink color bacil shape. The result of Biochemistry test this showed that 3 of 20 domestic chicken eggs were positive infected by Escherichia coli and 12 of 20 sample were infected by Salmonella sp. It can conclude that Escherichia coli and Salmonella were one of the causes of failed to hatch of domestic chickens  egg’s.Keywords : domestic chicken eggs, fail to hatch, Escherichia coli, Salmonella sp

Abstract

ABSTRAKPenelitian ini bertujuan untuk mengisolasi dan mengidentifikasi bakteri Gram positif kokus yang diduga menjadi penyebab pada kasus ear mites kucing domestik di Kecamatan Syiah Kuala Kota Banda Aceh. Sampel yang digunakan berasal dari 10 ekor kucing domestik yang positif menderita ear mites. Hasil swab liang telinga kucing dimasukan ke media Nutrient Broth (NB) dan diinkubasikan pada suhu 37°C selama 24 jam, selanjutnya menggunakan osse steril dilakukan penanaman pada media Manitol Salt agar (MSA) dan media Plate Agar Darah (PAD) dengan teknik goresan T lalu diinkubasikan kembali selama 24-48 jam. Morfologi koloni yang tumbuh terpisah diamati bentuk, ukuran, warna dan elevasi sebelum dilakukan pewarnaan Gram. Uji biokimia (Manitol dan Glukosa) dan Uji katalase dilakukan pada bakteri yang berbentuk kokus. Hasil penelitian ini dari 10 sampel dinyatakan 8 sampel (80%) penyebab infeksi sekunder pada kasus ear mites adalah bakteri Gram positif dari kelompok Staphylococcus sp dan 2 sampel (20%) dari kelompok Streptococcus sp. Dari 8 sampel yang terinfeksi bakteri kelompok Staphylococcus, 3 sampel (37,5%) merupakan infeksi tunggal Staphylococcus aureus dan 2 sampel (20%) merupakan infeksi tunggal Staphylococcus epidermidis, selain itu terdapat infeksi ganda antara Staphylococcus aureus dan juga Staphylococcus epidermidis sebanyak 2 sampel (20%) dan infeksi ganda Staphylococcus aureus dengan Micrococcus sp sebanyak 1 sampel (12,5%). Kata Kunci: Kucing domestik (Felis domesticus), Ear mites, bakteri Gram positif kokus ABSTRACT             The objectives of this study was to isolate and identify the Gram positive coccus bacteria which were suspected as one of the cause of secondary infection in ear mites case of domestic cats in Subdistric of Syiah Kuala, Banda Aceh. In this study, 10 of domestic cats suffer from ear mites were used. The swab ear canal from each cat  was inoculated in Nutrient Broth (NB) and incubated at 37°C for 24 hours. Then, using steril osse bacteria were cultured in Manitol Salt Agar (MSA) and Blood Agar Plate (BAP) with T streak method and incubated for 24-48  hours. The morphology coloni of bacteria were observed for shape, size, colour and elevation before Gram staining procedure was applied. Biochemical (manitol and glucose) and catalase test then conducted for the colonies which were identified as coccus in shape. The result of this study showed, 8 samples (80%) out of 10  samples were positive infected by the group of Staphylococcus sp and 2 samples (20%)  are positive infected by the group of Streptococcus sp. Frome those 8 samples  infected by the group of Staphylococcus sp, 3 samples (37,5%)  only infected by Staphylococcus aureus and 2 samples (25%) only infected by Staphylococcus epidermidis while the others 2 samples (25%) were infected by both Staphyloccocus aureus and Staphylococcus epidermidis and only 1 samples (12,5%) was infected by Staphylococcus aureus and Micrococcus sp. It can be concluded that  the group of Gram positive bacteria was the one of the cause of  secondary infection in the cats by ear mites.    Keywords: Domestic cat (Felis domesticus), Ear mites, Gram positif coccus bacteria
