Abstract

Penelitian ini bertujuan untuk mengetahui valid atau tidaknya suatu metode analisis. Pengembangan metode yang dilakukan yaitu menggunaan fase gerak asetonitril : 0,1 M dapar fosfat pH 4 (70 : 30). Selanjutnya dilakukan validasi ciprofloksasin. Parameter validasi dalam penelitian meliputi selektivitas, akurasi, presisi, linieritas, LOD dan LOQ. Kondisi HPLC meliputi perangkat HPLC merk Waters e2695 Separations, kolom SunfireTM C18 (150mmx 46 mm, 5µm), dan detektor PDA dan kecepatan alir 1,0 mL/menit. Penelitian ini diperoleh hasil selektif, hal ini dinyatakan dengan tidak munculnya puncak pada matriks. Metode analisis HPLC pada ciprofloksasin memenuhi kriteria keberterimaan nilai akurasi % recovery larutan konsentrasi ≥100 µg/mL adalah 100,40-103,88%. Memenuhi syarat uji presisi %RSD untuk larutan konsentrasi ≥100 µg/mL adalah <5,3%. Nilai r pada linieritas ≥0,9974 maka memenuhi syarat. Nilai LOD adalah 5,2471 µg/mL dan nilai LOQ adalah 17,4905 µg/mL. Kata kunci : ciprofloksasin, HPLC, validasi

Abstract

ABSTRACT Extracts of mengkudu fruit and mahkota dewa leaf each contain compounds that can be efficacious antibacterial Staphylococcus Aureus (acne-causing bacteria), but at high concentrations. The combination of both is expected to improve the efficiency and effectiveness of the use if the dosage form is formulated in a transparent soap. Selection of dosage forms based on a transparent soap acceptabilitas reasons. This study aims to determine the effect of the use of each and interactions extract of mengkudu fruit and mahkota dewa leaf in diameter clear zone antibacterial activity test by the method of pitting and dilution method with a parameter number of colony can then be determined comparison of the two extracts were having antibacterial activity optimum. The test results stated good use of each and combinations can enhance the clear zone according to the equation Y = 0.811 + 0.011 + 0.007 B + A 1.56. 10-5 AB, the parameters of the number of colonies also give the same result Y = 42.781 to 0.316 A - 0.228 0.006 A B + B. Optimal combination obtained extract concentration of 30% mengkudu fruit extract and 40% mahkota dewa leaf extract. Test the antibacterial activity of a combination of transparent soap mengkudu fruit and mahkota dewa leaf extracts with an optimal combination produces a clear zone diameter of 2.509 cm. Abstract

ABSTRACTThe purpose of this study is identification and analysis of the macronutrient glucomannan from porang tuber (Amorphophallus onchopyllus). The stages done in this research are extraction, identification, and macronutrient analysis of glucomannan of the porang tuber. Prior to extraction, the oxalic acid was removed from porang tuber using mixture of 2% lime juice and 5% lime, then the glucomannans was extracted using ultrasonic method. The glucomannan then was identified using Fourier transform infra red spectrometer (FTIR) instrument and macronutrient analyzer such as Soxhlet method for fats and fibers, Kjedahl method for protein, and Antrone method for carbohydrates. The results of identification using FTIR showed the presence of β-pyranose at wave numbers of 900-810 cm -1 . The carbonyl on on acetyl group was found at 1726 cm-1 . The area between 870-800 cm-1 showed the β-glycosidic and β-manosidic compounds. The C-O ether group was shown in the area of 1260-1200 cm -1 , while for C-O alcohol was shown in the area of 1050 cm-1 showed the C-O bond in alcohol. At 2925 cm-1 it showed C-H, while at 3000-3700 cm-1 O-H group. The levels of porang glucomannan macronutrients included fat at 0.50%, protein at 1.05%, fiber at 22.34%, and carbohydrates at 31.33%.Key word: Porang tuber, glucomannan, FTIR, macronutrient analysis.

Abstract

ABSTRACTIndonesia is a country with abundant natural resources, one of which tubers. Plants types of tubers that have a lot of glucomannan content of the tuber porang. Glucomannan is used as a diet in patients with hypercholesterolemia. Glucomannan from tubers porang (Amorphophallus oncophyllus Prain ex Hook.f.) serves as a water-soluble fiber and have a low calorie.The purpose of this study was to determine the effect of glucomannan tuber porang (Amorphophallus oncophyllus Prain ex Hook.f.) to decrease total blood cholesterol mice fed a high-fat diet and to determine what dose of glucomannan tubers porang most effective for lowering blood cholesterol.The treatment in this study is glucomannan flour dose of 25 mg / kg; 50 mg / kg; and 100 mg / kg. The population in this study was a white rat (Rattus norvegicus) males aged 2-3 months with BB ± 200 g.Giving glucomannan porang tubers dose 25 mg / kg; 50 mg / kg; and 100 mg / kg give effect to the reduction in total blood cholesterol mice fed a high-fat diet. Effective dose for lowering total cholesterol levels of mice that is 50 mg / kg.Keywords: glucomannan tuber porang (Amorphophallus oncophyllus Prain ex Hook.f.), total cholesterol.

Abstract

Obat tradisional banyak digunakan di Indonesia. Parameter mutu sangat perlu untuk menjamin keamanan dan khasiat dari senyawa bioaktif  yang terkandung dalam obat tradisional tersebut, salah satunya daun kenikir. Metode KLT Densitometri adalah metode sederhana, spesifik, dan tinggat kepekaannya tinggi digunakan untuk analisis kadar kuersetin fraksi etil asetat daun kenikir (Cosmos caudatus H.B.K). Daun kenikir dimaserasi menggunakan etanol 80%. Fase gerak yang digunakan adalah toluen : etil asetat : asam formiat (5:4:0,2), fase diam silika gel F254, dan panjang gelombang maksimal 372 nm. Larutan baku kuersetin pada rentang 0,7-2,0 µg didapatkan persamaan regresi linear y = 4,9982x10-3x + 2,0925x10-3 dengan koefisien kolerasi 0,9933 dan nilai koefisien variasi fungsi (Vxo) 2,54%. Persen recovery didapatkan antara 98,01-101,24% dan nilai persen deviasa standar relative sebesar 1,42%. Kadar kuersetin tertinggi pada fraksi etanol 10%: etil asetat (1:1) sebesar 576,733 ± 6,1101 µg/mL.Kata kunci: kenikir, kuersetin, KLT-Densitometri, verifikasi.
