Abstract

The aim of this research was to find the effect of interaction time on adsorption of Pb (II) by chitosan.The research was done in three stages: (i), the isolation of chitin from Bakau Crab’s Shell (Scylla sp); (ii), chitin deacetilation becomes chitosan; (iii), interaction of chitosan to Pb (II). Qualitative analysis was done by ninhidrin test, while quantitatively was carried out by infra red spectra. Langmuir Hinshelwood kinetica equation was applied to determine adsorption velocity of chitosan to Pb (II), and one way analysis of variance to determine the effect of interaction time to adsorption of Pb (II) (α = 5%). Interaction time was 10, 30, 50, 70, 90, 110, and 130 minutes. The results showed that: (a) deacetilation degree of chitin and chitosan were 35.94 and 65.28 % respectively, (b) ninhidrin test was resulting purple colour for chitosan, (c) period time of equilibrium to adsorp Pb (II) was 70 minutes. From Langmuir- Hinshelwood kinetica equation resulting that k1 = 0.0014 minutes-1 and K = 1189.6 M-1. Range of chitosan adsorption velocity to Pb (II) was 2977. 10-7 to 3.275. 10-7 mol/L minutes.

Abstract

The aim of the research were isolation, purification and characterization of Bacillus subtilis phytase from Holiwood Gresik. The research was done in two stages; the first include enzyme isolation, precipitation with amonium sulphate, dialysis, gel filtration chromatography, SDS-PAGE analysis, while second determining optimum pH, optimum temperature, the effect of pH and temperature to enzim stability, the values of KM and Vmax Bacillus subtilis phytase from Holiwood Gresik. The first stage research design were One Shot Case Study and Post Test Only Control Group Design, while the second stage were Post Test Only Control Group Design and Factorial Design. The data being analyzed by one-way and two-way Anova. The results of research showed that Bacillus subtilis phytase has the molecular mass of 36.5 kDa, optimum pH at 6.5–7.0, optimum temperature at 41° C and it was found to be stable for 30 minute incubation at pH 7 or 30° C with 2% or 3% lost of its activity respectively. KM value was 0.62 mM and VMax 0.393 mmol/ml/minute.

Abstract

The aim of the study were to find: 1)The effect of high intakes of dietary fiber on iron absorption and haemoglobin regeneration efficiency percentages, through combination of pH 4 and 7 with length of boiling time of 0 (raw), 5, 15, and 25 minutes. 2)The role of iron binding to dietary fiber on haemoglobin regeneration efficiency. The experiment involved yard long beans and 110 of male Rattus norvegicus (Wistar strains) as dietary fiber sources and animal model respectively. The results revealed that: 1) High intakes of yard long bean fiber will decreased iron absorption and haemoglobin regeneration efficiency. pH, boiling time and its interaction affected on iron absorption and haemoglobin regeneration efficiency (p < 0,05). On dietary fiber treatment, the highest iron absorption and haemoglobin regeneration efficiency at raw- pH 4 were higher than standard feeding treatment; while the lowest at pH 7- boiling time 5 minutes treatment. The treatment’s results at raw-pH 7 and pH4- boiling time 25 minutes were identical with standard feeding treatment. 2) An increase and decrease of boiling time and pH respectively increased iron absorption and haemoglobin regeneration efficiency, due to a decrease of iron binding to dietary fiber. The same results also occurred with non boiling treatment (raw condition) due to the role of exhancer substances on iron non heme absorption.

Abstract

Target utama yang diharapkan melalui kegiatan Program KKN-PPM meliputi dua hal yaitu 1) memberdayakan masyarakat dan mengembangkan potensi alam, 2) memberi keterampilan mahasiswa keterampilan bersosialisasi dan memecahkan masalah di masyarakat. Luaran utama dari kegiatan ini adalah kesejahteraan masyarakat yang meningkat melalui budidaya dan pemanfaatan tanaman yacon di desa Argosari, Kecamatan Senduro, Kabupaten Lumajang. Kegiatan KKN-PPM dilakukan selama 3 bulan dimulai pada bulan April sampai dengan Juli 2016. Namun untuk pelaksanaan evaluasi hasil budidaya tetap dilakukan sampai panen umbi yacon (sekitar 8 bulan). Hasil kegiatan: Yacon merupakan tanaman yang sesuai untuk ditanam di Desa Argosari Kecamatan Senduro Kabupaten Lumajang Jawa Timur. Tanaman tersebut telah berhasil ditanam dan diolah menjadi teh yacon oleh petani di desa tersebut. Mahasiswa KKN telah mampu bersosialisasi dengan masyarakat Desa Argosari melalui program penanaman yacon. Mahasiswa juga menunjukkan sikap tanggung jawab yang dapat dilihat dari perawatan tanaman yacon selama kegiatan KKN. Mahasiswa telah mampu mengembangkan keterampilan mereka, khususnya dalam hal bercocok tanam yacon. Kegiatan yang dilakukan mahasiswa selama KKN meliputi pelatihan penanaman yacon, penanaman bibit, dan perawatan tanaman yacon.
