Abstract

Indonesia is a country with the majority of its population adhering to Islam, and Muslims are obligated to consume halal food. Mixing products with non-halal ingredients, especially pork, is still often done to obtain economic benefits. One of the food products that are at risk of mixing is processed meat products. A method that can be used to detect the presence of pork in food products with accurate results and can save cost and time is a DNA-based analysis method developed from Polymerase Chain Reaction (PCR), namely Multiplex Polymerase Chain Reaction. This study used samples in the form of beef meatballs without halal logos and without BPOM numbers as many as 3 pieces, which were then extracted and amplified using Multiplex Polymerase Chain Reaction, then the results were read using electrophoresis with 2% agarose gel. The electrophoresis results showed a specific band with a length of 279 bp in the positive control of beef and beef meatball samples, and 951 bp in the positive control of pork. Based on these results, it can be concluded that all three samples were from beef and did not contain pork. These results indicate that samples of beef meatballs circulating in traditional markets that do not have clear information on the packaging purely contain beef and do not contain pork. These results can also show that the Multiplex Polymerase Chain Reaction (PCR) method can be used to detect the presence of porcine DNA from food products of animal origin with accurate results.
