Abstract

Research on the separation and characterization of chemical compounds from taro tuber (Colocasia esculenta Schott var. antiquorum) has been done. The maceration results of 850.00 g simplicial with 70 % ethanol solvent obtained 102.16 g brown viscous extract. The result of chemical components identification showed that the ethanol extract of taro tuber was positive for alkaloids, flavonoids, and terpenoids. Separation and purification of the ethanol extract were done by a liquid-liquid partition, thin layer chromatography and two-dimensional chromatography with a combination of n-hexane and ethyl acetate (2:8) as mobile phase. We found 4 bands (I-IV) with maximum wavelength were 407.0, 268.50, 253.00, and 209.50 nm. The presence of absorption at a wavelength of 268.50 nm and 253.00 nm indicating the presence of a class of flavonoids. The infrared spectrum showed that the isolates had typical groups such as O-H, C=O, C-C, C-O, and C=C.

Abstract

Klika faloak (Sterculia populifolia) telah digunakan oleh masyarakat secara tradisional untuk mengobati beberapa penyakit seperti hepatitis, kanker, gangguan saluran pencernaan, diabetes, reumatik dan peningkat sel darah merah dll. Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi senyawa alkaloid ekstrak metanol klika faloak. Ekstraksi dilakukan secara maserasi menggunakan pelarut metanol. Ekstrak metanol dipartisi dengan n-Heksan. Fraksinasi menggunakan metode kromatografi cair vakum, sebagai fase diam digunakan silika gel dan fase gerak adalah campuran n-heksan : etil asetat : metanol dengan sistem kepolaran ditingkatkan. Fraksi positif alkaloid yang diperoleh dipisahkan menggunakan kromatografi lapis tipis preparative dan dilakukan uji kemurnian menggunakan kromatografi lapis tipis 2 dimensi. Isolat selanjutnya dikarakterisasi menggunakan instrumen spektrofotometer UV-Vis dan inframerah (FTIR).  Hasil analisis spektroskopi UV-Vis dan FTIR isolat diduga adalah senyawa alkaloid yang mempunyai karakteristik gugus fungsi N-H, C-N, C-H dan C=O serta serapan UV pada panjang gelombang 269 dan 302 nm yang merupakan serapan n→ π* dari gugus N-H.

Abstract

The Ziziphus spina-christi L. plant, commonly known as Arabian bidara, are rich in potentially health-beneficial compounds, particularly those with antibacterial properties. This research aimed to investigate the effectiveness of an ethanolic leaf extract in inhibiting the growth of pathogenic bacteria. The leaves were subjected to maceration using 70% ethanol, followed by an assessment of their antibacterial properties using the agar diffusion method. Subsequently, the active extract was analyzed through bioautography using thin-layer chromatography (TLC), and its phytochemical spotted was screened. The results of the antibacterial activity using the ethanolic leaf extract revealed that it exhibited antibacterial activity when applied at a concentration of 2 mg per paper disk. Subsequently, it showed inhibition zones with diameters of 19.44 mm for Bacillus subtillis; Escherichia coli (17.06 mm); Staphylococcus aureus (16.32 mm); Pseudomonas aeruginosa (12.96 mm), and Salmonella typhi (14.50 mm). The TLC-bioautography analysis of the ethanolic leaf extract demonstrated inhibitory activity against Bacillus subtillis of Rf value with 0.96, Escherichia coli (0.31) and Staphylococcus aureus (0.89). However, no inhibitory activity was observed against Salmonella typhi. The phytochemical screening results suggested the presence of flavonoids, terpenoids, and tannins in the ethanolic leaf extract.
