Abstract

Permasalahan sektor perikanan saling terkait antar sub sektor perikanan tangkap, budidaya, serta olahan dan pemasaran hasil perikanan. Penelitian bertujuan menganalisis potensi keberlanjutan usaha multi sub sektor perikanan skala kecil - menengah oleh masyarakat di Kabupaten Sidoarjo. Penelitian deskriptif dilaksanakan April 2015 s/d April 2018. Survey terestris dengan teknik rekam data tertutup dan terarah menggunakan alat bantu kuesioner. Data diberi bobot dan dianalisa menggunakan analisis shift share dan statistik untuk mengetahui keterkaitan antar sub sektor perikanan terhadap potensi keberlanjutannya. Hasil analisis keberlanjutan usaha tangkap (kode 01.T) bernilai terendah 2,3529 gap 6,0 dari nilai tertinggi 8,3529. Nilai regresi usaha penangkapan ikan Y = 0,005 + 0,961 X menunjukkan usaha penangkapan ikan belum mampu memberdayakan sektor lain. Tingkat signifikansi uji T tidak nyata 22,2%. Nilai R Square 0,005 dan Adjusted R Square -0,061 merepresentasikan tingkat kepercayaan usaha penangkapan ikan sangat rendah. Keberlanjutan usaha perikanan budidaya di tambak (simbol 02.Y) bernilai terendah 2,9783. Nilai regresi linier sebesar Y = 0,980 + 3,375 X menunjukkan usaha budidaya memberikan keberdayaan bagi sub sektor lain secara signifikan 97,8%*. Nilai R Square 0,225 dan Adjusted R Square 0,188 merepresentasikan keberlanjutan usaha budidaya kurang menjanjikan. Keberlanjutan olahan dan pemasaran hasil perikanan (kode 03.U) bernilai terendah 7,2600 dengan shift share gap positif 0,2600. Nilai regresi linier Y = 6,031 + 3,235 X signifikansi 100% menunjukkan usaha olahan dan pemasaran berpengaruh terhadap usaha lainnya, dengan nilai R Square 0,651 dan nilai Adjusted R Square 0,636. Hasil penelitian menyimpulkan bahwa sub sektor olahan dan pemasaran hasil perikanan berpeluang besar untuk ditumbuh-kembangkan.Title: Analysis of Potential Sustainability of Multi Fisheries Sub Sector Business in the Sidoarjo RegencyThe problems of fisheries sector are interrelated between the capture fisheries, cultivation, processing and marketing of fishery products. The research aims to analyze the potential sustainability of small and medium scale multi sub-sector fisheries businesses by people in Sidoarjo Regency. Descriptive research was conducted from April 2015 to April 2018. Terrestrial survey with closed data recording techniques and questionnaires were used in the study. Data were measured and analyzed using shift share matrix and statistics to find out the relation between fisheries sub-sectors to their potential sustainability. Results of the capture business sustainability analysis (code 01.T) have the lowest value of 2.3529 gap 6.0 from the highest value of 8.3529. The regression value of fishing business Y = 0.005 + 0.961 X indicates that fishing businesses have not been able to empower other sectors. The significance level of the T test is not real 22.2%. The R Square value of 0.005 and Adjusted R Square -0.061 represents the relatively low level of trust in fishing businesses. The sustainability of aquaculture business in the pond (symbol 02.Y) has the lowest value of 2.9783. The linear regression value of Y = 0.980 + 3.375 X indicates that cultivation provides empowerment for other sub-sectors significantly of 97.8%*. The value of R Square 0.225 and Adjusted R Square 0.188 representing the sustainability of aquaculture is less promising. Sustainability of processed and marketing of fishery products (code 03.U) has the lowest value of 7.2600 with a positive shift share gap of 0.2600. The linear regression value Y = 6.031 + 3.235 X 100% significance indicates that the processed business and marketing affect other businesses, with the value of R Square 0.651 and the value of Adjusted R Square 0.636. It is concluded that the processed and marketing of fishery products subsector have a great opportunity to be developed.

Abstract

Background.In our previous research, when cell culture were exposed to high glucose, this will cause the increase of H2O2. At the exposure to 22 mM glucose on 3rd day, the increase of H2O2 that induced the activation of Phospholipase C (PLC) have caused 1P3 (Inositol tri-phosphate) mobilizing the release of Ca²+ from the depo Endoplasmic reticulum (ER). Thus, causing the increase of cytosolic Ca²+. Giving thapsigargin (TG) will cause significant increase in Cytosolic Ca²+ so that the most contribution to the increasing of Cytosolic Ca²+ derives from the ER . On the 7th day exposure, H2O2 played the same role as TG, causing direct incease in Cytosolic Ca²+ and an addition of Ca²+ free/buffer ethyleneglyco bis (ßaminoethyl ether).&NNN’N’– tetraacetic acid (EGTA) caused significant decrease of cytosolic Ca²+ basal and the greatest contribution to the increase of cytosolic Ca²+ on the 7th day, comes from extracellular. Administrating Cyclosporin A (CSA) 10 µM on the 9th day, caused significant decreasing on cytosolic Ca²+ basal, the ability of CSA in decreasing Ca²+ basal concentration was less than the 3rd and 7th days. At a high glucose condition with different length of exposure, a change of new cytosolic Ca²+ homeostatic regulation occurred and this enable a change in the dynamic interaction among ER, extracellular and mitochondria.Method.HUVECs culture exposed to 22 mM glucose for 3, 7 and 9 days. The cells were incubated with FURA2-AM. The evaluation of fluorescence cytosolic Ca²+ was done by epifluorescence Nikon digital camera-computerized analyser. To measure the cytosolic Ca²+ concentration we use Histogram Image Corel Draw Photo Paint 12.Result. Exposure to glucose 22mM on the 3rd day (65.4 ± 12.2) it showed the increase of cytosolic Ca²+ by giving Ca²+ free/EGTA 1 mM and CSA 10 mM caused the decrease of cytosolic Ca²+ (33.2 ± 4.47) TG1µM and CSA caused the decrease of cytosolic Ca²+ basal (53.07 ± 2.75) and Ca²+ -free/EGTA, TG and CSA (68.59 ± 5.71). On the 7th day exposure (92.74 ± 7.66) the decrease of cyto -solic Ca²+ basal occurred at the giving of Ca²+ -free/EGTA, TG (50.52 ± 9.23). EGTA and CSA (45.59 ±6.2). TG and CSA (73.55 ± 7.30), Ca²+ -free/EGTA and TG much more decrease the concentrate of cytosolic Ca²+ basal (17.58 ±4.5). On the 9th day of exposure to glucose (72.32 ±7.46), the giving of Ca²+ -free/EGTA, TG and CSA(35.76 ± 5.25) have caused the decrease of cytosolic Ca²+ basal. Conclusion.HUVECs culture exposed to 22mM glucose will cause the increase in H2O2and cytosolic Ca²+ basal. ER, mitochondria and extracellular regulate the Cytosolic Ca²+ and a dynamic interaction occurred among them to obtain a new homeostatic.

Abstract

Penelitian yang bersubyek pada petani tradisional Arfak dari Subsuku Hatam telah dilaksanakan di Kampung Syoubri, Kwau dan Mokwam Distrik Warmare Kabupaten Manokwari yang bertujuan mengkaji dan mendeskripsikan tipologi kebun campuran petani Hatam eksisting dan permasalahannya. Penelitian ini merupakan penelitian survei deskriptif yang didesain menggunakan pendekatan kualitatif naturalistik yang dimaksudkan untuk mendeskripsikan secara sistematis, faktual dan akurat terkait fakta-fakta, karakteristik serta hubungan antar fenomena yang diteliti. Metode pengumpulan data yang digunakan adalah primary methods yang menggunakan beberapa teknik sekaligus seperti teknik partisipasi, observasi langsung, wawancara mendalam dan studi literasi/pustaka (Morrisan, 2016). Jumlah sampel ditetapkan sebanyak 5 orang per kampung dan menggunakan teknik sampling bola salju mengingat subyek penelitian bersifat homogen (Yunus, 2010).Variabel yang diamati adalah karakteristik kebun campuran meliputi lama bera, luas lahan, jenis tanaman, cara bercocok tanam, produksi, dan pemasaran. Hasil penelitian menunjukkan bahwa kebun campuran petani tradisional Hatam saat ini termasuk dalam tipologi agroforesti sederhana semi subsisten, yang dicirikan dengan masa bera 2 -5 tahun, terbatasnya jenis tanaman yang ditanam, terdiri dari tanaman hortikultura dan tanaman pangan semusim, tidak menggunakan input kimia, tanpa olah tanah dan pola tanam tidak teratur, produksi terbatas dengan kualitas yang rendah dimana hasil kebun separuh untuk kebutuhan sendiri dan lainnya untuk dijual. Permasalahan utama yang dihadapi antara lain pendapatan rendah dari hasil kebun, meningkatnya kebutuhan ekonomi, rendahnya pengetahuan dan ketrampilan petani karena belum adanya pembinaan yang intens dari Dinas terkait, terbatasnya lahan pertanian potensial yang dapat dikelola, faktor resiko kegagalan panen cukup tinggi akibat bencana alam dan ketiadaan lembaga tataniaga yang membantu pemasaran produk petani.
