Abstract

Polyurethane is a class of polymers characterized by their substantial tensile strength and high melting points which enables them to be extremely durable. Despite its xenobiotic origin, polyure-thane has been found susceptible to biodegradation by microorgan-isms. The main object of this research was to investigate polyurethane degradation by the bacterium isolated from decayed teak wood (Tec-tona grandis Linn. f.). Polyurethane degrading bacteria were isolated using selective medium in the form of LB (Luria Bertani) with the ad-dition of 0.3% Impranil. Bacterial biodegradation activities shown by the changes of polyurethane structure analyzed by FT-IR spectrosco-py. The bacterial identification was carried out based on observations on the morphological characteristics of the colonies, cell morphology and sequences of 16S rRNA encoding genes. One of the potential iso-late that successfully isolated was K9, which demonstrates the disap-pearance of the 1735/cm peak of the characteristic function urethane in the FT-IR analysis. Analysis of the 16S rRNA encoding gene showed that the potential isolate having 98% similarity index to Bacillus safen-sis strain FO-36b. The activity shown by the isolate suggests that the bacteria could be a promising agent for polyurethane degradation.

Abstract

Rice fields are one of the main sources of high nitrous oxide N2O emission. Soil denitrifiers possessing high N2O reduction activity are important for controlling N2O emission. Nitrous oxide reduction is the last step of denitrification process. The aims of this study were to characterize and to identify denitrifying bacteria isolated from rice soils possessing high activity of N2O reduction. Soil samples were taken from 6 locations of rice fields in Bogor (West Java) and Tangerang (Banten), Indonesia. Physiological characterization was performed using API 20 NE, while molecular identification was conducted based on the 16S rRNA gene sequence. It was found that ten isolates of denitrifying bacteria were able to grow using N2O as an electron acceptor as indicated by decreasing N2O concentration in the headspace of the cultures. The bacterial growth indicated by optical density, increased up to 0.12-0.47 after 5 days incubation. Isolate BL2 had the highest activity of N2O reduction followed by BL1 and BLN1 at up to 5.41, 4.09, and 3.91 μmol mL-1 bacterial cultures, respectively. The BL1, BL2, and BLN1 isolates had some different physiological characteristics. Based on their 16S rRNA sequence, BL1 and BLN1 were closely related to Ochrobactrum anthropi ATCC 49188 with similarity of 99%.

Abstract

Nitrous oxide (N2O) is one of the principal greenhouse gases. Differences in soil microbial community composition affect N2O emission. Ochrobactrum anthropi BL1 and BLN1 isolated from rice field in Tangerang, Banten, Indonesia can grow on and reduce N2O.  This study investigated  the patterns of N2O reduction activity and growth of O. anthropi BL1 and BLN1 on denitrification media and also examined the ability of BLN1 strain to reduce N2O in flooded rice soil. Nitrous oxide reduction activity and growth of strains BL1 and BLN1 occurred simultaneously, indicating that the bacteria used N2O for growth. BL1 and BLN1 showed the same specific growth rate, but the N2O reduction rate of BLN1 was higher than that of BL1. Increase of the N2O concentration in the surface water of flooded soil without BLN1 isolate six hours after the addition of NO3- was significantly greater than the surface water from soil that had been inoculated with the isolate.

Abstract

The aims of this research are to synthesize and determine the formula, characteristics, and complex structure of Cu(II) and Co(II) with 4-aminophenol and to investigate their antibacterial activity. The complexes were synthesized by refluxing a solution of CuSO4·5H2O and CoSO4·7H2O, respectively, with 4-aminophenol in methanol for 1 h. The products were characterized using UV-Vis spectroscopy, atomic absorption spectroscopy, thermal analysis, conductivity, FTIR, and magnetic moment. The formation of the complex was indicated by shifting of maximum wavelength of the metal solution toward shorter, i.e., 817  to 421 nm for Cu(II) and 566  to 450 nm for Co(II). From the characterization, the proposed formulas of the complexes are [Cu(4-aminophenol)4]SO4 and [Co(4-aminophenol)4(H2O)2]SO4·5H2O forming square planar and octahedral geometry, respectively. Both complexes are paramagnetic with negligible antibacterial activity against Staphylococcus aureus, Staphylococcus epidermis, Escherichia coli, and Pseudomonas aeruginosa.

Abstract

The endophytic fungi that live in endemic plants are a promising bio‐prospect as the producers of antibacterial compounds. This research is aimed to evaluate the endophytic fungi antibacterial compound from Mangifera casturi. The bioactive compounds of 13 endophytic fungi were extracted using ethyl acetate and evaluated for antibacterial activity using disk diffusion assay. The minimum inhibitory concentration (MIC) was measured using the serial broth dilution method. Scanning Electron Microscopy (SEM) was used to examine cell damage because of the extract. The antibacterial compounds were then detected using GC‐MS analysis. The endophytic fungi were identified morphologically and molecularly based on ITS rDNA sequencing Among the 13 isolates, the endophytic fungi identified as Botryosphaeria rhodina AK32 produced the antibacterial compounds that exhibited the highest activity and a broad spectrum. Moreover, they were capable against resistant bacteria, Methicillin Resistant Staphylococcus aureus (MRSA) with an MIC value of 1.56% for all the test bacteria. The mechanism of action of AK32 ethyl acetate extract seemed to affect the condition of bacterial cell walls, causing morphological alteration such as shrinkage of the cell, warted cells, and hollow cells. Based on GC‐MS, the antibacterial compounds of AK32 ethyl acetate extract were di‐n‐octyl phthalate, benzyl alcohol, high‐oleic (CAS) safflower oil, benzene acetonitrile, and benzotriazole.

Abstract

Universias Sebelas Maret (UNS) Surakarta has a piped drinking water distribution system. Drinking water from a piped system is called tap water. Tap water is a public facility provided by the government in European cities but not in Indonesia. However, consumption of tap water by the academic community at UNS Surakarta was relatively low. This was caused by the trust and worthiness of UNS’s tap water. This research aimed to determine the quality of tap water at UNS Surakarta, both in aspects of physics, chemistry and microbiology. Selection of sampling points based on the location of the distribution pipes and sampling using the grab sampling method. The tap water analyzed by Indonesian National Standard (SNI) test for drinking water. The results were compared with the Indonesian Government Regulation on Drinking Water, namely PERMENKES 492/2010. The quality of physical, chemical and microbiological aspects of tap water at UNS Surakarta were meet Indonesian Government Regulations, namely Permenkes 492/2010. Thus, tap water from UNS Surakarta is equivalent to bottled drinking water and worth to drink.

Abstract

Health education or promotion is part of hypertension management to control hypertension. Visual media is often used in health promotion activities, because the material displayed is easy to understand and interesting. So far, society has controlled or reduced hypertension only pharmacologically, and is not familiar with complementary therapies for managing hypertension. Good public knowledge will influence positive attitudes and behavior. The aim of this research is to determine the effect of using visual media regarding hypertension management on the attitudes and behavior of hypertension sufferers. Research with one group pretest posttest design with a sample size of 30 respondents from Gesono Village, Wonosegoro District, Boyolali Regency. Data analysis used the independent t test. The results of data analysis show that there is an influence of visual media regarding hypertension management on attitudes (p<0.001) and behavior (p=0.001) in hypertension sufferers. There is a significant influence of visual media on the management of hypertension with the behavioral attitudes of hypertension sufferers in Gesono Village, Wonosegoro District, Boyolali Regency.

Abstract

This study aims to analyze internal control over the procurement cycle of government goods/services in one of the government agencies, namely the National Land Agency (BPN) Cilacap. From the implementation of the controls carried out, further analyzed its conformity with Government Regulation (PP) No. 60 of 2008 concerning the Government Internal Control System. The method used in this study is a qualitative method by conducting interviews with informants to obtain primary data and documentation to obtain secondary data. The informants in this study were a Commitment Making Officer (PPK) and a Government Employee with a Contract Agreement (PPPK) who served as the operator of SAKTI application assets. The result of this study is that the application of internal control over the procurement cycle of goods/services at BPN Cilacap has been implemented and in accordance with PP No. 60 of 2008. However, not fully internal control has been implemented because there are still sub-elements of internal control that have not been implemented.

Abstract

Ternak kambing merupakan salah satu jenis ternak ruminansia kecil tetapi memberikan manfaat yang besar, selain menghasilkan daging dan susu, feses kambing juga dapat dijadikan bahan pupuk berkualitas tinggi. Ternak kambing juga dapat dibudidaya secara kerakyatan karena tidak memerlukan lahan yang luas, biaya produksi relatif rendah, pemeliharaan yang sederhana, serta produksi dan panen tidak membutuhkan waktu lama. Sebagian umat Buddha di Vihara Dhammapanna Dusun Krajan bermata pencaharian sebagai petani. Namun, penghasilan sebagai petani tidak menentu karena bersifat musiman dan bergantung pada kondisi cuaca. Kemampuan ekonomi mempengaruhi kesejahteraan umat, sehingga perlu adanya kegiatan untuk meningkatkan perekonomian.  Sosialisasi perekonomian umat Buddha dan pemberdayaan masyarakat melalui peternakan kambing mampu memberikan hasil yang positif. Diperolehnya pengetahuan mengenai budidaya ternak kambing dan kesehatannya. Salah satu keberhasilan dari program ini adalah Salah satu penerima kambing berhasil mengembangbiakkan kambingnya dan menghasilkan 1 ekor anak kambing. Selain itu, kambing juga tampak tumbuh dan berkembang dengan baik. Akan tetapi terdapat beberapa hal yang dievaluasi terkait tentang kendala pemeliharaan ternak kambing dan perkembangan ternak kambing. Adapun kendala dalam pemeliharaan ternak kambing yaitu keterbatasan lahan yang menyebabkan beberapa kambing ditempatkan dalam satu kandang. Letak kandang kambing ada yang di dalam dan di luar rumah. Semua penerima menggunakan model kandang lemprak dengan berasalaskan jerami. Permasalahan yang muncul dalam pengelolaan peternakan kambing, yaitu adanya kambing yang mengalami penyakit mata (pink eye). Oleh karena itu, diperlukan pemberian obat dan pemeriksaan secara rutin.

Abstract

Tujuan penelitian yaitu; (1) mendeskripsikan peningkatan keterampilan membaca pemahaman cerita jataka melalui penerapan metode CIRC (Cooperative, Integrated, Reading, Composition); (2) penerapan metode CIRC (Cooperative, Integrated, Reading, Composition) dalam pembelajaran pendidikan agama Buddha pada meteri jataka. Jenis penelitian yaitu penelitian tindakan kelas. Tempat pelaksanaan penelitian yaitu di Homeschooling Kusalamitra, Gunung Kidul. Penelitian ini berfokus pada peningkatan keterampilan membaca pemahaman cerita jataka siswa kelas X pada pembelajaran agama Buddha. Pengumpulan data dilakukan melalui pengamatan, wawancara, dokumen, serta tugas atau tes. Data yang sudah terkumpul lalu divalidasi dengan teknik triangulasi data dan review instrumen kunci untuk selanjutnya dianalisis, direduksi, dan disimpulkan. Hasil dari penelitian ini yaitu penerapan metode CIRC mampu meningkatkan keterampilan membaca pemahaman siswa kelas X Homeschooling Kusalamitra pada pembelajaran agama Buddha dalam materi Jataka. Rata-rata nilai keterampilan membaca pemahaman siswa pada saat pra tindakan yaitu 57,63. Pelaksanaan siklus 1 nilai-nilai rata meningkat menjadi 71. Hasil pelaksanaan siklus 2 meningkat menjadi 78,25. Penerapan metode CIRC mengkondisikan siswa aktif membaca, menemukan ide pokok dari cerita jataka, dan terlibat diskusi kelas.
