Abstract

Introduction: Aging is a biological process that can induces changes to the structural integrity and physiological function of skin. Structural changes of skin are a result of dermal atrophy, decreased collagen, the loss of subcutaneous fat, the loss of inherent elasticity, and increased melanogen.1,5,6 From the definition itself, a stem cell characterized by its ability to self-renew and also have an ability to differentiate along multiple lineage pathways.2,4 ADSC which is a kind of mesenchymal stem cell from human adipose tissue, have some potential properties as stem cells derived from bone marrow.4,7 Objective: The aim of this study was to verify the comparison effect of ADSCs and their secretome for skin aging treatment. Methods: ADSC were isolated from lipoaspirates obtained from healthy donors after obtaining written consent and ethical approval using liberase enzymatic digestion, washed with steril PBS and centrifuged. After the phase, ADSC were seeded directly after isolation with liberases and cultured, then produced the conditioning media (AAPE) by hypoxia amplifies.3,4,6,7 We analysed the ADSCs (2,5 x 105 cells) and their secretomes (0,1ml in 0,4ml NaCl 0,9%) by subcutaneous injection on the back of a rat (with range age 48 weeks), and followed began every two weeks after injected until six weeks and stained with Van Giesson staining, for measured the density of collagen.3,5,6 Results: Collagen density was increased after ADSC injected to the skin. From statistical analysis showed there was a significant result of collagen density compared to control group and AAPE (p<0.05). We conclude that ADSC had anti-aging potential or regenerative by stimulating collagen synthesis of dermal fibroblast. ADSC will be as new modality treatment for aging skin in future.

Abstract

Abstract Introduction & Objectives : Limbal stem cells (LSCs) play important role in maintaining corneal clarity. Disruption of LSCs or its niche may lead to LSCs deficiency that result in blindness. This study aimed to investigate therapeutic effect of advanced adipose protein extract (AAPE) in LSCs regeneration. Methods : AAPE was derived from adipose derived mesenchymal stem cell secretome purifed by ultrafiltration technique. 2D culture of LSCs were exposed by 0.25 NaOH, then treated with serum-free media as control group, media supplemented with 20% and 50% AAPE as treatment group I and II, respectively. MTT assay was conducted to measure cell proliferation and in vitro scratch assay to assess cell migration at 48 hours after treatment. EGF, FGF, HGF, and KGF from supernatant were measured by ELISA. Data was analyzed using One way ANOVA and post-hoc Tukey. Results : A 50% AAPE successfully promoted the highest proliferation and migration rate of LSCs compared to control group (94.38±2.51%, P 0.001; 68.52±3.51%, P 0.001). EGF (1216.67pg/mL), FGF (988.25pg/mL), HGF (119.33pg/mL), and KGF (793.25pg/mL) from 50% AAPE supernatant group were highest among other groups. Conclusion : A 50% AAPE demonstrated optimal beneficial effect on LSCs regeneration through upregulation of EGF, FGF, HGF, and KGF associated with niche modulation.

Abstract

Glaucoma is the leading cause of irreversible blindness. The aim of this study was to review the profile of secondary glaucoma cases visiting a tertiary hospital in East Java. This is retrospective observational study, completed case records of new patients with secondary glaucoma who presented to glaucoma clinic from January 2014 to April 2016 were included. Out of the 363 case records screened, 66 cases were found to eligible for inclusion. The evaluation included a detailed history and examination performed including vision, anterior segment examination, intraocular pressure (IOP), gonioscopy, and fundus evaluation. Diagnosis of secondary glaucoma was made on the basis of presence of a secondary cause for presence of raised IOP. 66 cases were eligible for inclusion in the study, most of the cases was occurred in the range age 21-50 years. The male female ratio was 1.3:1. Frequent causes of secondary glaucoma were lens factor 30.8%, steroid induced 29.5%, uveitic 20.5%, neovascular15.4%, and surgical complication 3.8%. Most patients with secondary glaucoma have poor vision < 0.1 with high IOP at presentation. Assessment and early detection of underlying cause is the key guide to treatment strategy.

Abstract

Mata adalah organ vital dalam kehidupan manusia terutama bagi Pekerja Migran Indonesia di Malaysia yang sering menghadapi keterbatasan untuk akses ke fasilitas layanan kesehatan. Program Pengabdian Kepada Masyarakat (PKM) yang dilaksanakan oleh Program Studi Doktoral Fakultas Kedokteran Airlangga, Surabaya, Indonesia. Kesehatan mata dipengaruhi oleh berbagai faktor seperti faktor usia, hipertensi, penyakit kardiovaskuler, tiroid dan diabetes mellitus yang memicu gangguan penglihatan bahkan dapat menyebabkan kebutaan.  Konseling mengungkapkan bahwa 90% PMI enggan berobat karena kendala biaya dan administrasi. Program ini menunjukkan bahwa sebesar 25,67% PMI yang hadir mengalami masalah kesehatan mata dan Sebagian besar 55,18% disebabkan oleh kelainan presbyopia, myopia, asthenopia. Sebanyak 27,59% kelainan akomodasi pada kedua bola mata, 24,14% kelainan rabun jauh dan 20,69% kelainan sindrom mata kering. Selain itu program PKM juga berhasil meningkatkan kesadaran dan pengetahuan PMI tentang pentingnya kesehatan mata dibuktikan dengan peningkatan persepsi positip dari 37,5% menjadi 73,5%.  Peserta juga memberikan umpan balik positip dan berharap kegiatan ini berkesinambungan.
