Abstract

The present study investigated the effect of culture medium supplementation with mercaptoethanol ( ME)and amino acid (AA) on canine intergeneric embryo development with porcine oocyte cytoplasm. Porcine cumulusoocyte complexes (COCs) were collected from slaughterhouse and matured in TCM-199 supplemented with 26.2mM NaHCO, 3.05 mM glucose, 0.91 mM sodium pyruvate, 0.57 mM L-cysteine, 75 mg/l kanamycin, 10 ng/ml 3epidermal growth factor, equine chorionic gonadotropin (eCG), 10 IU/ml human chorionic gonadotropin (hCG),and 10% (v/v) porcine follicular fluid (pFF) at 39 °C in a humidified atmosphere of 5% CO for 42-44 h and donor cell 2collected from ear skin afghanhound male dog. After somatic cell nuclear transfer (SCNT), embryo developmentwere examined for cleavage rate and 144 hr for final development after cultured in media. The result shows that,amino acid and mercapoethanol addition in culture medium (NCSU-23) have no effect on embryo development.The development rate of embryo until 16 cell stage in NCSU and NCSU supplement are 4.67% and morula stage are3.73% and 4.67%.Key words : intergeneric clone embryo, canine, ( amino acid (AA)

Abstract

Kegiatan desa binaan tahun ke-2 ini sebagai bagian dari desa binaan 3 tahun yang bertujuan menjadikan Kemiri Sebagai Kawasan Petenakan Terpadu. Tahapan tahun ni menitikberatakan pada upaya peningkatan keterampilan budi daya kambing pada masyarakat Kemiri, kecamatan Tanjungsari, Kabupaten Gunungkidul. Kegiatan dilakukan dengan inisiasi budidaya kambing kacang dan konsultatif. Pendampingan lanjutan diperlukan untuk semakin meingkatkan kapasitas peternak dan menyangga kemandirian ekonomi peternak melalui budi daya kambing berupa pemasaran, teknologi pasca produksi, pengolahan pakan dan limbah

Abstract

Lumba-lumba hidung botol (bottlenose dolphin) adalah spesies lumba-lumba yang paling umum danpaling dikenal orang. Gangguan sistem respirasi pada lumba-lumba sering dijumpai, sementara jenisbakteri yang sering menyerang lumba-lumba dari perairan Laut Jawa belum pernah diteliti. Penelitianini bertujuan untuk mempelajari pemeriksaan klinis sistem respirasi lumba-lumba dan melakukan isolasibakteri serta jamur pada sistem respirasi bottlenose dolphin (Tursiops aduncus) dari Perairan Laut Jawa.Penelitian dilakukan dengan studi pustaka, pemeriksaan klinis serta isolasi sampel sistem respirasi. Swabdeep blowhole dilakukan terhadap sepuluh ekor bottlenose dolphin di PT. Wersut Seguni Indonesia.Sampel dikirim ke laboratorium Mikrobiologi FKH UGM untuk isolasi dan identifikasi terhadap bakteridan jamur. Hasil penelitian menunjukan bahwa pemeriksaan klinis lumba-lumba hanya dapat dilakukansecara inspeksi. Hasil isolasi ditemukan bahwa 5 dari 10 lumba-lumba (50%) positif Staphylococcus aureus,sedangkan jamur tidak ditemukan. Berdasarkan hasil penelitian, disimpulkan bahwa pemeriksaan klinislumba-lumba dapat dilakukan dengan cara inspeksi terhadap blowhole, tingkah laku hewan, adanya“chuff” atau “honk”, frekuensi respirasi, sosialisasi hewan dan posisi hewan saat berenang. BakteriStaphylococcus aureus dijumpai pada sistem pernafasan lumba-lumba hidung botol dari perairan LautJawa di PT. Wersut Seguni Indonesia.

Abstract

Berbagai penelitian menunjukkan bahwa ekstrak media penumbuh sel punca mesenkimal (EMPSPM), tanpa sel punca itu sendiri, telah ditemukan terdapat berbagai faktor tropik hasil sekresi sel punca mesenkimal di dalam media kultur yang dapat meregenerasi jaringan yang rusak. Penelitian ini bertujuan untuk mengetahui gambaran kadar malondialdehid (MDA) dalam plasma dan ginjal tikus diabetes melitus tipe 2 (DMT2) dengan terapi EMPSPM. Dua puluh lima ekor tikus wistar digunakan dalam penelitian ini dibagi menjadi 5 kelompok yaitu kelompok DMT2 + 0,05 ml/kg BB EMPSPM (0,05); kelompok DMT2 + 0,1 ml/kg BB EMPSPM (0,1); kelompok DMT2 + 0,2 ml/kg BB EMPSPM (0,2), kelompok kontrol DMT2 (DMT2), dan kelompok sehat (KS). Induksi DMT2 dengan menggunakan streptozotosin nikotinamid (STZ-NA). Terapi mulai dilakukan pada hari ke 7 setelah kondisi DM tercapai, diberikan 4 kali dengan selang waktu 7 hari secara intraperitoneal. Data kadar glukosa darah dan MDA dianalisi secara statistik. Hasil penelitian menunjukkan bahwa induksi DMT2 dengan STZ-NA dapat menaikkan kadar glukosa dan MDA dalam darah (P<0,05). Terapi 0,05; 0,1; dan 0,2 ml/kg BB EMPSPM menunjukkan dapat menurunkan kadar glukosa darah, kadar MDA plasma dan ginjal (P<0,05). Kadar 0,2 ml/kg BB EMPSPM menunjukkan aktifitas lebih baik dalam menurunkan kadar glukosa darah dan MDA. Berdasarkan penelitian ini, EMPSPM dapat menurunkan kadar glukosa darah serta kadar MDA dalam darah dan ginjal tikus DMT2.

Abstract

Secretome is a factor found in the stem cell culture medium. This factor may repair the tissues of the organs that were damaged by various degenerative disorders1. A previous study has reported that secretome derived from the fetal human umbilical cord mesenchymal stem cell (HUC-MSC) may be an effective regenerative agent for β-cell pancreatic regeneration in Type 1 diabetes mellitus and for skin regeneration in incisional and burn wound healing.The administration of cisplatin as a chemotherapeutic agent is limited because it causes side effects, such as reproductive toxicity and progresses to testicular dysfunction2. As a degenerative disorder, testicular dysfunction is characterized by failure in the synthesis of reproductive hormones and spermatogenesis. Cisplatin-induced testicular cell damage would promote vimentin and cytokeratin localization in the spermatogenic, Sertoli, and Leydig cells.Studies on the use of secretome in promoting the recovery of various degenerative disorders and its effects on the spermatogenesis of rats with cisplatin-induced testicular dysfunction have not been conducted. The aim of this stud was to investigate the effect of secretome derived from HUC-MSC on cisplatin-induced testicular dysfunction in rats.

Abstract

Mesenchymal stem-cell is a pluripotent cells that can be differentiated into many kinds of cells (1) and have abilities to differentiated into mature cell, secreted cytokines, and releasing many kinds of protein functioned in body body regulation (2). However, the therapeutic effect of stem cell arise from their secreted factor such as growth factors, cytokines, chemokines, and metabolits which acts as biologic regulator in the autocrine and paracrine body function (3). Secreted factors of mesenchymal stem cells known as secretome or mesenchimal stem cell conditioned medium (MSC-CM) have therapeutic effect for antiapoptosis, angiogenic, immunomodulatoric, and chemoattraactive activity (4). Mesenchymal stem cell conditioned medium used as the replacement agent for cell therapy because it contains no cells and save for used in different individual (5).Type 1 diabetes mellitus (DMT1) is a disorder characterized by destruction of the insulin-producing pancreatic β-cell that progressively leads to insulin deficiency and resultant hyperglycemia. Insulin deficiency can lead to progressive metabolic derangement, with worsening hyperglycemia, ketoacidosis, starvation, and death (6). Glucagon is a hormone produced by pancreatic α cells. Glucagon releases by the pancreas to raise the concentration of glucose in the bloodstream when the concentration of glucose in the bloodstream too low (7). Glucagon worked in opposite with insulin, which lowers the extracellular glucose level, while insulin works to increase the extracellular glucose level (8). The aim of this study was to investigate the role of MSC-CM on the structural and functional regeneration of pancreatic α and β cells in Wistar rat (Rattus norvegicus) induced with type 1 diabetes mellitus. The datas in this study will completes the information about the effect of MSC-CM on pancreatic cells regeneration, which has been presented in previous study (9).

Abstract

Hystrix javanica or usually called Sunda porcupine is wildlife animal that only found in Java, Bali, Sumbawa, Flores, Lombok, Madura, and Tonahdjampea (Indonesia) [1]. International Union for Conservation of Nature and Natural Resources (IUCN) has classified this Rodentia into Least Concern criteria, that means the species is relatively widespread and abundant [2]. Nevertheless, their population in nature was threatened, since this mammal was considered as pest by some people.There were anatomical data of Sunda porcupine organ, but it still limited. One of the ways to determine the detail function of the organ can be done by making histological preparations. Lectin histochemistry has been used widely as a probe to detect sugar residues in the organs or tissues because lectin can bind specifically to carbohydrate residues in term of glycoconjugates [3]. Analysis the composition or type of sugar residues in the organs can help to understanding the role of their function.The aims of this study were to detect and find out the distribution of complex oligosaccharide in the lingua of Hystrix javanica using lectin histochemistry method. This data will completes the information that has been obtained before about mucopolysaccharide type of major salivary glands of Hystrix javanica [4].

Abstract

This study was conducted to increase the efficiency of canine embryo production by intergeneric somatic cell nuclear transfer (SCNT) technology. The effect of oocyte recipient for development of intergeneric canine somatic cell cloning embryos were examined. Bovine and porcine cumulus oocyte complexes (COCs) were collected from slaughterhouse ovaries and matured in TCM-199 medium depend on species. Adult dog fibroblasts collected from 3.5 years old Afghanhound dog, and cell between passage 1 and passage 10 used for intergeneric somatic cell cloning using bovine and porcine oocytes as oocyte cytoplasm donor. The result showed that oocytes from bovine and porcine can de-differentiated canine nucleus and no different between bovine and porcine oocyte in fusion and embryo development in vitro. Canine intergeneric cloned embryos developed to morula stages in vitro.

Abstract

The present study investigated the effect of green fluorescent protein on the development of canine intergeneric clone embryo with bovine oocyte recipient. Cumulus oocyte complexes (COCs) were collected from slaughterhouse and matured in TCM-199 supplemented with 10% (v/v) fetal bovine serum (FBS) (Life Technologies), 0.005 U/m1 bovine FSH (Antrin®, Denka Kanagawa, Japan) and 1 pg/m1 estradiol (Sigma-Aldrich) at 39 °C in a humidified atmosphere of 5% CO2 in air and donor cell tranfected with enhanced green fluorescent protein. In this experiment GFP have no negative effect in fusion or embryo development but the expression rate were decreased in 2 cell stage and then the expression gradually decreased with progression of embryo development.
