Abstract

ABSTRACT Runtunuwu, S.D. and E.F. Lengkong. 2005. Identification of Isozyme Markers (PER-7, PER-8, and RAPD OBI7375) of Dwarf Salak Coconut (GSK) and its Tall Hibrids. Eugenia 11 (1): 8-17. The objective of this research was to identify PER-7 and PER-8 isozymes, and OB17 375 RAPD as molecular markers for resistance to NF Phytophthora disease in Genjah Salak (GSK) dwarf coconut and its several hybrids with Tall coconut at The Research Institute for Coconut and Palmae (RICP) Manado. Coconut resistance to NF Phytophthora disease was determined based on disease lesion size of inoculated coconut fruits at 7 days after inoculation. PER-7 and PER-8 isozymes marker were identified using horizontal starch gel electrophoresis, and OB17375 RAPD marker was identified based on polymerase chain reaction (PCR) using kit B number 17 primer (Operon Technologies, California). PER-7 isozyme marker could be used to select GSK x DTA (Dalam Tenga) and GSK x RLT (Rennell Tall) hybrid coconuts that resistant to NF Phytophthora disease. PER-7 isozyme without OB17375 markers (PER-7/-) could be used to select resistant GSK x WAT (West African Tall) hybrid coconut to the disease. Abstract

ABSTRACT Response of rice to paclobutrazol (PBZ) depends on plant variety. This experiment were done to know the influence of PBZ to growth and yield of local rice Wesel using randomized block design (RBD). The treatment consist of four dosages of PBZ with four replications. The results showed PBZ treatmant enhance of grain yield per plant, number of productive tillers (NPT),  weight of 1.000 grains, crop growth rate (CGR), and leaf chlorophyll content.Keywords: Paclobutrazol (PBZ), crop growth rate (CGR), chlorophyll, yield, local rice

Abstract

ABSTRACT   Runtunuwu, S.D. et al. 2008. Assembling Hybrid Coconut of GSK x DMT Based on RAPD (RANDOMLY AMPLIFIED POLYMORPHIC DNA) Marker. Eugenia 14 (1) : 134-152.   The aimed of this research was : 1. assembling hybrid coconut GSK x DMT (Genjah Salak x Dalam Mapanget) that seeds growth was relatifly homogeneous based on RAPD (Randomly Amplified Polymorphic DNA) marker and 2. to found the assembling method of hybrid coconut that will produce massive seeds relatifely short time will homogeneous plant. It was 65 individu trees observe for the average of famale flower per bunch. The result was 25 individu of coconut GSK has the average flower production > 40 per bunch was analyze the homogeneous genetic with the RAPD marker. Based on the analyze RAPD that were 25 individu of GSK coconut trees have the same genetic average 88 % and 14 individu among that was 100 % have same genetic. Further more that 14 individu of GSK was crossing with the 3 individu of DMT that have high yield per year its was DMT 1188, 1172 and 781. Based on the evaluation for the color of buds, high of buds, the steam circle, the petiole color and the germination time of hybrid coconut seeds from the crossing of GSK x DMT 1188 produce more than    70 % seeds that have same genetic, also for crossing of GSK x DMT 1172 have 9 combination and have more than 70 % that same genetic, 10 combination from crossing GSK x DMT 781 have more than 80 % same seeds growth. Therefore, using the RAPD marker were successfully produced 28 crossing of the hybrid coconut GSK x DMT that have relatifly homogeneous seeds growth.   Keywords : assembling, hybrid coconut GSK x DMT, RAPD.

Abstract

ABSTRACT   This research aims to produce a shorter cloves and generate more interest and qualified by the presence of paclobutrazol treatment. The results showed that the high accretion paclobutrazol can produce the content of chlorophyll a (0,032 mg g-1) and chlorophyll b (0,004 mg g-1), dry weight of 1000 grains (207 g) and dry weight / plant (6.038 kg) was 8 years old clove higher than the control. Recommended further research to determine the maximum effect of paclobutrazol application on plants. Paclobutrazol at a dose of 2.5 g / tree height increment can deliver maximum results of cloves. Abstract

ABSTRACT   The study aimed to examine the influence of time of application and concentration of paclobutrazol on growth and yield of potato crop Supejohn held in District Modoinding South Minahasa regency, from March to June 2012, using a randomized block design with two factors. The first factor is the concentration of Paclobutrazol with four standards are: 0 ppm, 75 ppm, 100 ppm, 125 ppm and the second factor is the provision of a three-level applications MST 4, 5 and 6 MST MST (Week After Planting). Based on research, the results of analysis of variance showed an interaction between application timing and concentration of paclobutrazol. Combination treatment with 125 ppm paclobutrazol 6 WAP application time can give significant effect on plant height, number of chlorophyll content, percentage weight class tuber yield components in the form of increased crop production amounted to 45.04 kg / plot (24 plants) / range 52. ton / ha by pressing the vegetative growth of plants. Abstract

ABSTRACT Runtunuwu, S.D., et.al. 2007. The Become Fruit Rate in Crossing Coconut Genjah Salak x Dalam Mapanget (GSK x DMT). Eugenia 13(1):97-108. The aim of this research was 1) individu tree crossing of coconut Genjah Salak x Dalam Mapanget (GSK x DMT), 2) to known the become fruit rate in crossing individu tree of coconut GSK x DMT. The method of crossing used crossing a pair between fourteen trees individu of coconut GSK. With three trees coconut DMT, so it become fourty two combination crossing. Coconut DMT 1188 crossing with stem number one of coconut GSK, DMT 1172 with stem number two coconut GSK and DMT 781 with stem number three. The become fruit rate of the crossing result between fourteen individu trees of coconut GSK and tree individu trees of DMT with the tree number 1188, 1172 and 781 in three mounth in sequence was have variation from 0,00 – 27,27 %, 3,76 – 32 % and 2,86 – 27,16 %. Therefore the lowest success rate of these crossing was in combination of GSK coconut tree number fourteen and DMT tree number 1188 was 0,00 %, while the highest success rate was in the combination crossing between coconut GSK tree number 12 and DMT tree number 1172 was 32 %. Eugenia 13 (1):109-118.   The goal of this research to study the variety of DNA tape form and the population variety of coconut Genjah Salak (GSK) based on RAPD code (Randomly Amplified Polymorphic DNA). 25 trees individu of coconut GSK was amplification the genomic of DNA using 35 primer Operon Kit A and Kit B. The coconut GSK individu tree that were analyse was selected based on the female flower production, with average above 40 female flower. The electrophoresis of DNA result of amplication PCR get tape form monomorfic and polimorfic between 25 individu coconut GSK trees that was analyzed. OPA 09 primer and OPB 06 get the result of monomorfic tape form, other wise the OPA 13 primer, OPB 05 and OPB 10 get polimorfic tape form. The genetic similarity in coconut Genjah Salak (GSK) population based on RAPD code rate was 88 % or have the variety genetic of 12 %. Therefore based of this similarity the coconut GSK population was clustering in five groups. Abstract

ABSTRACT The purpose of this research was to study the effect of the concentration of growth inhibitor Paclobutrazol (Pbz) against the high growth of clove seedlings (Syzygium aromaticum (L.) Merryl & Perry) and get Pbz concentrations that can produce seed cloves in a relatively short (dwarf). The results showed that spraying Paclobutrazol (Pbz) with 25-100 ppm affect height growth of clove seedlings. Pbz the higher concentration applications, the shorter the length of apical segments produced shoots. In other words, the higher the concentration Pbz, the higher the emphasis on high growth of clove seedlings. Pbz emphasis on high growth of clove seedlings varied from the lowest, ie. 3.79% (25 ppm) up to the highest, ie 69.85% (100 ppm). Based on the comparison of height increment of plants sprayed with Paclobutrazol, the dose applications can be used to get the seed cloves that dwarf is a concentration of 50 to 100 ppm. Abstract

Clove (Syzigium aromaticum L.) is one of the important commodity crop farming, especially as a basic material in the kretek cigarette industry, also belong to the volatile oil-producing spice crops used as basic materials for pharmaceutical and food industries. The study aims were: 1) manipulating the canopy shape of clove plants to be shorter than normal, (2) obtaining the right dose to increase the productivity of young clove plants. Benefit study is to provide information technology for the farmers to increase crop productivity clove.  Research was done in the  Popareng Village,  District of Tatapaan, South Minahasa Regency in November 2011 to May 2012. Research compiled in Randomized Block Design (RBD) with one factor of treatment, consisting of four dosage of paclobutrazol (P), namely: P0 = 0 g tree-1; P1 = 1.0 g tree-1; P2 = 1.5 g tree-1 : P3 = 2.0 g  tree-1. The results showed that paclobutrazol inhibited the high growth of the apical buds is 72% at a dosage of 1.0 g tree-1, 73% at a dosage of 1.5 g tree-1 and 81% at a dosage of 2.0 g tree-1 that produces clove crop 3-year-old to be shorter than normal growth. Paclobutrazol applications increase weight 1000 grain dried flowers tree-1 and production tree-1. Highest production achieved in the paclobutrazol dosage 2.0 g tree-1, which is  0.024 kg of 1000 grain weight of dried flowers and production  tree-1 is 3.350 kg, while the lowest production in a dosage of 0.0 g paclobutrazol (control), is 0.192 kg of 1000 grain weight dried flowers and production tree-1  is 1.012 kg.
